    Abstract Severe arteriosclerotic stenosis of the renal artery with at least 60-70% narrowing of the lumen can lead to various diseases: in the case of unilateral stenosis it can lead to renovascular hypertension, in the case of bilateral narrowing (or in a stenotic solitary kidney) also to an often progressive renal insufficiency (ischemic kidney disease) and/or to acute pulmonary edema (pulmonary flash edema). Renal artery stenosis may be treated by revascularization using either percutaneous (balloon angioplasty with or without stenting) or less commonly open surgical procedures, both with excellent primary patency rates of over 90%; however, randomized trials of catheter-based interventions have failed to demonstrate a longer term benefit with respect to blood pressure control and renal function as well as improved overall survival over optimal medicinal management alone. Due to improved clinical outcomes interventional revascularization is justified in cases with critical stenoses and clinical sequelae, such as pulmonary flash edema and progressive renal failure. Careful patient selection is essential to maximize a potential clinical benefit.

In most cases arteriosclerotic disease is the underlying cause; less prevalent are the various manifestations of fibromuscular disease. Renal artery stenosis may be treated by revasularization, using either percutaneous (balloon angioplasty, stenting) or rarely open surgical procedures, both with excellent primary patency rates. However, randomized trials of renal artery angioplasty or stenting in patients with arteriosclerotic lesions have failed to demonstrate a longer-term benefit with regard to hypertension control and renal dysfunction over medical management alone. Careful patient selection is essential to maximize the potential benefit (e.g., in patients with refractory hypertension, progressive renal failure or recurrent pulmonary flash edema).

    Abstract |Today's story in Matthew's gospel is likely a familiar one to most people. Jesus goes out in a boat with his disciples and the sea's turn for the worse. The disciples are scared and believe they may die and all the while Jesus is asleep. They call on him to wake up. I can picture myself on that boat as the sky turns dark, the winds blow hard as the waves come up over the sides of the boat. I would be terrified, no doubt. After I read the story, however, the familiarity of it changed as I became fixed on the idea of Jesus being asleep while all this was going on.|Many of my thoughts these days seem to focus on racism and the struggles that people of color experience in today's world. I think about being a white man and how I have greatly benefited from those two unearned characteristics. I have done nothing to earn the many privileges that just seem to be part of my normal life. For most of my life I just kind of assumed everyone lived like me and had the same opportunities and benefits that I did. I am beginning to understand, however, that this is not so – and unfortunately, far from the true life experiences of my brothers and sisters of color. I have been asleep while the storm blew all around me.|Racism, sexism, consumerism, war, disease, politics, and other noxious doings of life today foster a sense of dualism – or a separation of humanity where we do not see our similarities, but only our differences. If there were ever a time when we should recognize the sameness of all humanity, it should be now. The COVID-19 virus does not discriminate based on color, race, gender, or political alliance. Nor too, do the feelings of hurt and anger after watching people of color murdered by those with power and privilege.|The storm of racism and injustice isn't scary to those who are asleep. The inability to see the happenings of today, either by choice or by ignorance, is a position of unearned privilege. It is time to wake from our sleep and take action to calm the storm – just as Jesus did. It's time for everyone who has been asleep to get up and rebuke the wind and the sea, and perhaps then we can have great calm.

    Abstract |My reflection on the readings of the day usually draw me towards the gospel. Today's gospel reading from Mark certainly has a great deal of good stuff to reflect upon as Jesus ascends into heaven and his disciples go forth and preach everywhere.|But, my attention from today's readings are drawing me to Peter's first letter. It seems to say so much about what the world is experiencing right here and now. Peter says, "Cast all your worries upon him because he care for you." How perfect for the worries of the newness we live in today. But, Peter goes on to teach us about something that is perhaps as equally important - our sameness with one another. A few lines later he says, "…knowing that your brothers and sisters throughout the world undergo the same sufferings." If there is one positivity that we can take away from this pandemic is the realization of our sameness in our humanity.|Sometimes our fast-paced lives cause us to get caught up inside the issues of our personal lives only. It's easy to feel like our problems are worse than others. This "otherness" leads to an individualistic view and does not allow us to see the interconnections among all of us. It doesn't allow us to see that we are just part of the whole, rather than being "the whole". Peter reminds us that people everywhere experience the same difficulties and challenges regardless of where you live. Likewise, the COVID-19 pandemic is teaching us the same. It doesn't matter your gender, race, nationality, socio-economic status, or even political party, no one is immune to the challenges of the day. We are all experiencing physical isolation from one another, anxiety from not knowing what the immediate and long-term future holds, and perhaps a longing for "the good 'ol days" of pre-pandemic life. But, knowing that we are not suffering inside of our individual-ness is somehow comforting because it allows us to connect with everyone across the globe at a deeper level. And, perhaps if we reflect upon that notion for long enough, we can realize that in order for us to feel whole as an individual, we need to paradoxically come to terms with the idea that we are only part of the whole and share the greater wholeness with the rest of humanity.

    Abstract Chemical biology and the application of small molecules has proven to be a potent perturbation strategy, especially for the functional elucidation of proteins, their networks, and regulators. In recent years, the cellular thermal shift assay (CETSA) and its proteome-wide extension, thermal proteome profiling (TPP), have proven to be effective tools for identifying interactions of small molecules with their target proteins, as well as off-targets in living cells. Here, we asked the question whether isothermal dose-response (ITDR) CETSA can be exploited to characterize secondary effects downstream of the primary binding event, such as changes in post-translational modifications or protein-protein interactions (PPI). By applying ITDR-CETSA to MAPK14 kinase inhibitor treatment of living HL-60 cells, we found similar dose-responses for the direct inhibitor target and its known interaction partners MAPKAPK2 and MAPKAPK3. Extension of the dose-response similarity comparison to the proteome wide level using TPP with compound concentration range (TPP-CCR) revealed not only the known MAPK14 interaction partners MAPKAPK2 and MAPKAPK3, but also the potentially new intracellular interaction partner MYLK. We are confident that dose-dependent small molecule treatment in combination with ITDR-CETSA or TPP-CCR similarity assessment will not only allow discrimination between primary and secondary effects, but will also provide a novel method to study PPI in living cells without perturbation by protein modification, which we named "small molecule arranged thermal proximity coaggregation" (smarTPCA).
