    Abstract We propose a mutant detection approach based on endonuclease IV and DNA ligase in combination with qPCR. The enzymes functioned cooperatively to facilitate PCR for low abundance DNA detection. We demonstrate that our approach can distinguish mutations as low as 0.01%, indicating the potential application of this strategy in early cancer diagnosis.

    Abstract Full employment is important to promote the high-quality development of the urban economy. Using urban-level data on China from 2004 to 2018, we analyse the effects and mechanism of expanding imports on urban manufacturing employment. We use the Guiding Opinions on Strengthening Import to Promote Balanced Development of Foreign Trade issued by the China State Council in 2012 as a natural experiment to solve the endogeneity problem. We find that expanding imports significantly increases urban manufacturing employment. This conclusion is still robust after a series of robustness tests. Further mechanism tests reveal that productivity improvements and upgrades to product quality from expanding imports can explain increased urban manufacturing employment. The results of the heterogeneity analysis show that expanding imports promote manufacturing employment in large and medium-sized cities but not small cities. Expanding imports increases employment in manufacturing in cities in different regions, with the largest effects on eastern cities, the second largest effects on western cities, and the smallest effects on central cities. These results suggest that expanding imports is an effective channel for increasing employment.

    Abstract The work presented in this paper is in the area of brain tumor detection. We propose a fast detection system with 3D MRI scans of Flair modality. It performs 2 functions, predicting the gray level distribution and location distribution of the pixels in the tumor regions and generating tumor masks with pixel-wise precision. To facilitate 3D data analysis and processing, we introduce a 2D histogram presentation encompassing the gray-level distribution and pixel-location distribution of a 3D object. In the proposed system, specific 2D histograms highlighting tumor-related features are established by exploiting the left-right asymmetry of a brain structure. A modulation function, generated from the input data of each patient case, is applied to the 2D histograms to transform them into coarsely or finely predicted distributions of tumor pixels. The prediction result helps to identify/remove tumor-free slices. The prediction and removal operations are performed to the axial, coronal and sagittal slice series of a brain image, transforming it into a 3D minimum bounding box of its tumor region. The bounding box is utilized to finalize the prediction and generate a 3D tumor mask. The proposed system has been tested extensively with the data of more than 1200 patient cases in BraTS2018∼2021 datasets. The test results demonstrate that the predicted 2D histograms resemble closely the true ones. The system delivers also very good tumor detection results, comparable to those of state-of-the-art CNN systems with mono-modality inputs. They are reproducible and obtained at an extremely low computation cost and without need for training.

    Abstract Background: Arteriovenous fistula (AVF) is the preferred vascular access for patients undergoing haemodialysis (HD). AVF malfunction remains a major clinical problem and is a significant independent risk factor for death. Although far-infrared (FIR) therapy has been shown to reduce complications and improve the patency rate of AVFs in various studies, it has been cautiously recommended by the Kidney Disease Outcome Quality Initiative (KDOQI) guidelines for AVF care due to insufficient evidence. Therefore, it is necessary to identify more effective methods for preventing AVF dysfunction. Many in vitro studies and few clinical studies have examined the effects of near-infrared (NIR) therapy on the vasculature. This study will examine the effects of NIR therapy on AVF.
    Methods: A randomised, controlled, open-label, multicentre trial will compare the effect of NIR on AVF patency after 1 year of therapy with that of a control group of patients with existing AVF. One group of patients received NIR treatment above their AVFs, whereas the control group received regular care. The primary outcome is the primary fistula patency rate within 12 months. In addition, acute changes in inflammatory, vasodilatory and haemodynamic parameters after a single treatment in the first 40 participants will be examined. This study was registered in the Clinical Trials Registry (ChiCTR2300071305) at https://register.clinicaltrials.gov/.
    Discussions: This study will explore the long-term and acute effects of NIR on AVFs. The study findings will provide information that can be used to develop new technical support for the prevention of AVF dysfunction in patients undergoing haemodialysis.
