    Abstract In this work, the topological effect on binding interaction between a G-quadruplex and thioflavin T (ThT) ligand was systematically investigated on a platform of an intramolecular split G-quadruplex (Intra-SG). Distinct fluorescence changes from ThT were presented in the presence of distinct split modes of Intra-SG structures and an intriguing phenomenon of target-induced fluorescence light-up occurred for split modes 2 : 10, 5 : 7 and 8 : 4. It was validated that hybridization between the Intra-SG spacer and target did not unfold the G-quadruplex, but facilitated the ThT binding. Moreover, the 3' guanine-rich fragment of Intra-SG was very susceptible to topology variation produced by the bound target strand. Additionally, a bioanalytical method was developed for ultrasensitive gene detection, confirming the utility of the ThT/Intra-SG complex as a universal signal transducer. It is believed that the results and disclosed rules will inspire researchers to develop many new DNA-based signal transducers in the future.

    Abstract DNA as an intriguing organic ligand has been widely employed for synthesizing metal nanoclusters and engineering their properties. This review aims to present recent progress on DNA-encoded metal (Ag, Cu, Au, Ag/Pt, Cu/Ag, etc.) nanoclusters (DNA-MNCs), focusing on their applications in the fields of analysis, logic operation, and therapy based on properties including fluorescence, electrochemiluminescence (ECL), and antibacterial and catalytic activity, and summarizes the attractive features of the latest research. The key points are briefly described as follows. (1) Analytical systems have been constructed based on fluorescence regulation, and nuclease-assisted and enzyme-free amplification strategies have been extensively adopted with fluorescent DNA-MNCs for amplified analysis. (2) DNA-MNCs may play more than one role (emitter, quencher, or catalyst) in ECL-based analytical systems. (3) Apart from antibacterial activity, DNA-MNCs also possess apparent catalytic capability, such as enzyme-like activity (i.e., nanozymes), which has been applied in colorimetric systems. (4) Reversibly regulating the catalytic activity of DNA-MNCs has been attained with DNA systems. It is believed that through in-depth investigation of the relationship between atomic structure and property, more novel DNA-MNCs will be explored and applied in the future.

    Abstract: Reported here is a new Cu(ii) compound with the formula of {Cu4(μ 2-Cl)6(μ 4-O)(4-NH2-py)4}n (1, 4-NH2-py = 4-amino-pyridine). The analysis of X-ray diffraction shows that the complex 1 indicates a 0D separated skeleton with {Cu4(μ 2-Cl)6(μ 4-O)} ... ...

    Abstract Reported here is a new Cu(ii) compound with the formula of {Cu4(μ 2-Cl)6(μ 4-O)(4-NH2-py)4}n (1, 4-NH2-py = 4-amino-pyridine). The analysis of X-ray diffraction shows that the complex 1 indicates a 0D separated skeleton with {Cu4(μ 2-Cl)6(μ 4-O)} tetranuclear cluster. The interactions of H-bond between the amino groups and μ 2-Cl anions from adjacent 0D cluster further extended this 0D cluster into a 3D supramolecular framework. In addition, this compound has excellent photocatalytic activities for the degradation of methyl blue under UV radiation. The cyclic testing results indicate that complex 1 exhibits good recyclability, as it can be repeated for as many as five cycles with no noticeable loss of performance, revealing its potential application in pollutant degradation within the environmental field.

    Abstract Road disease detection is challenging due to the the small proportion of road damage in target region and the diverse background,which introduce lots of domain information.Besides, disease categories have high similarity,makes the detection more difficult. In this paper, we propose a new LDBFSS(Latent Domain Background Feature Separation and Suppression) network which could perform background information separation and suppression without domain supervision and contrastive enhancement of object features.We combine our LDBFSS network with YOLOv5 model to enhance disease features for better road disease detection. As the components of LDBFSS network, we first design a latent domain discovery module and a domain adversarial learning module to obtain pseudo domain labels through unsupervised method, guiding domain discriminator and model to train adversarially to suppress background information. In addition, we introduce a contrastive learning module and design k-instance contrastive loss, optimize the disease feature representation by increasing the inter-class distance and reducing the intra-class distance for object features. We conducted experiments on two road disease detection datasets, GRDDC and CNRDD, and compared with other models,which show an increase of nearly 4% on GRDDC dataset compared with optimal model, and an increase of 4.6% on CNRDD dataset. Experimental results prove the effectiveness and superiority of our model.

    Abstract Intelligent transportation system combines advanced information technology to provide intelligent services such as monitoring, detection, and early warning for modern transportation. Intelligent transportation detection is the cornerstone of many intelligent traffic services by identifying task targets through object detection methods. However existing detection methods in intelligent transportation are limited by two aspects. First, there is a difference between the model knowledge pre-trained on large-scale datasets and the knowledge required for target task. Second, most detection models follow the pattern of single-source learning, which limits the learning ability. To address these problems, we propose a Multi Self-supervised Pre-fine-tuned Transformer Fusion (MSPTF) network, consisting of two steps: unsupervised pre-fine-tune domain knowledge learning and multi-model fusion target task learning. In the first step, we introduced self-supervised learning methods into transformer model pre-fine-tune which could reduce data costs and alleviate the knowledge gap between pre-trained model and target task. In the second step, we take feature information differences between different model architectures and different pre-fine-tune tasks into account and propose Multi-model Semantic Consistency Cross-attention Fusion (MSCCF) network to combine different transformer model features by considering channel semantic consistency and feature vector semantic consistency, which obtain more complete and proper fusion features for detection task. We experimented the proposed method on vehicle recognition dataset and road disease detection dataset and achieved 1.1%, 5.5%, 4.2% improvement compared with baseline and 0.7%, 1.8%, 1.7% compared with sota, which proved the effectiveness of our method.

    Abstract Adenosine triphosphate (ATP) is regarded as the "energy currency" in living cells, so real-time quantification of content variation of intracellular ATP is highly desired for understanding some important physiological processes. Due to its single-molecule readout ability, nanopipette sensing has emerged as a powerful technique for molecular sensing. In this study, based on the effect of targeting-aptamer binding on ionic current, and fluorescence resonance energy transfer (FRET), we reported a dual-signal readout nanopipette sensing system for monitoring ATP content variation at the subcellular level. In the presence of ATP, the complementary DNA-modified gold nanoparticles (cDNAs-AuNPs) were released from the inner wall of the nanopipette, which leads to sensitive response variations in ionic current rectification and fluorescence intensity. The developed nanopipette sensor was capable of detecting ATP in single cells, and the fluctuation of ATP content in the differentiation of dental pulp stem cells (DPSCs) was further quantified with this method. The study provides a more reliable nanopipette sensing platform due to the introduction of fluorescence readout signals. Significantly, the study of energy fluctuation during cell differentiation from the perspective of energy metabolism is helpful for differentiation regulation and cell therapy.
