Abstract Despite extensive characterization of the state and function of haematopoietic stem cells (HSCs), the use of transcription factors to define the HSC population is still limited. We show here that the HSC population in mouse bone marrow can be defined by the distinct expression levels of Spi1 and Gata1. By using a double fluorescence knock-in mouse model, PGdKI, in which the expression levels of PU.1 and GATA-1 are indicated by the expression of GFP and mCherry, respectively, we uncover that the HSCs with lymphoid and myeloid repopulating activity are specifically enriched in a Lin

Abstract Editorial: How to deal with climate change, how to mitigate or even reverse it, maybe the hottest scientific topic of the 21st century. Do you know that a Chinese scientist and his team contributed to climate change control by reducing PM2.5 in diesel car exhaust by 35-40%? That scientist is Prof. Lin Li, a Fellow of the Royal Academy of Engineering and founder of the Laser Processing Research Centre at The University of Manchester, United Kingdom. Of course, this is only one example of Professor Li's scientific achievements. As a pioneer of microsphere super-resolution lens, his team, in collaboration with Singapore colleagues, broke the optical diffraction limit in optical microscopic imaging, making real-time observation of biological viruses without interference possible. He also used lasers to synthesize new nanomaterials which kill drug-resistant bacteria while remaining harmless to healthy human cells, which led to the development and breakthrough of related research fields. We are much honored to have Professor Lin Li for an exclusive interview in which he recalls his years of scientific research experience and talks about the future development trend of laser material processing.

Abstract Objective: To propose and validate a novel and comprehensive classification of lumbosacral spondylolysis and spondylolisthesis in the pediatric population according to surgical managements.
Methods: Cases of pediatric lumbosacral (L5/S1 level) spondylolysis and spondylolisthesis were reviewed. Lumbar Japanese Orthopedics Association score, health-related quality of life, spondylolisthesis, lumbosacral disc degeneration, and dysplastic features were included for evaluation of outcomes after surgery. We also conducted a reliability study to assess the classification.
Results: A classification containing 4 types and a tentative treatment algorithm were proposed: type 1, direct pars repair without segmented fusion, for which the pedicle screw-U rod system is suitable; type 2, simple L5-S1 fusion; type 3, simple L4-S1 fusion; and type 4, simple L5-S2 fusion. All types of spondylolisthesis were suggested to achieve complete reduction and decompression. A total of 162 patients were included and we also reviewed the outcomes of patients in each type. All patients achieved satisfied outcomes. The reliability study showed that the average interobserver agreement was 89.5% (range, 85.2%-92.6%), with κ value of 0.862 (range, 0.802-0.901). Intraobserver agreement ranged from 88.9% to 92.6%, with an average κ value of 0.868 (range, 0.835-0.901).
Conclusions: In our study, we proposed and validated a novel comprehensive classification of pediatric lumbosacral spondylolysis and spondylolisthesis. The pedicle screw-U rod system provided more benefits in the treatment of simple lumbosacral spondylolysis, and fusion from L5 to S2 combined with S2 screw would also benefit patients with high-degree spondylolisthesis.

Abstract: ... areas in Lin’an, southeastern China using logistic regression (LR), decision tree (DT), and random ...

Abstract Landslides are a common geodynamic phenomenon that cause substantial life and property damage worldwide. In the present study, we developed models to evaluate landslide susceptibility in forest-covered areas in Lin’an, southeastern China using logistic regression (LR), decision tree (DT), and random forest (RF) techniques. In addition to conventional landslide-related natural and human disturbance factors, factors describing forest cover, including forest type (two plantations (hickory and bamboo) and four natural forests (conifer, hardwood, shrub, and moso bamboo) and understory vegetation conditions, were included as predictors. Model performance was evaluated based on true-positive rate, Kappa value, and area under the ROC curve using a 10-fold cross-validation method. All models exhibited good performance with measures of ≥0.70, although the LR model was relatively inferior. The key predictors were forest type, understory vegetation height (UVH), normalized differential vegetation index (NDVI) in summer, distance to road (DTRD), and maximum daily rainfall (MDR). Hickory plantations yielded the highest landslide probability, while conifer and hardwood forests had the lowest values. Bamboo plantations had probability results comparable to those of natural forests. Using the RF model, areas with a shorter UVH (<1.2 m), a lower NDVI (<0.70), a heavier MDR (>115 mm), or a shorter DTRD (<500 m) were predicted to be landslide-prone. Information on forest cover is essential for predicting landslides in areas with rich forest cover, and conversion from natural forests to plantations could increase landslide risk. Across the study areas, the northwestern part was the most landslide-prone. In terms of landslide prevention, the RF model-based map produced the most accurate predictions for the “very high” category of landslide. These results will help us better understand landslide occurrences in forest-covered areas and provide valuable information for governments in designing disaster mitigation.
