    Abstract The observed discharge, an important input for flood forecasting systems, can significantly affect the accuracy of forecasting results. Since the reservoir inflow is not measured directly but calculated based on the observed reservoir stage and the reservoir outflow, it always contains gross errors causing some inflow to be outliers. In this study, a robust error estimation method for real-time flood forecasting has been developed for reservoirs’ data processing. The method differs from the conventional flood forecasting method, in that it represses the gross errors by a robust loss function based on the real error distribution of measurements. Furthermore, a fluctuation coefficient has been proposed to quantify the degree of fluctuation of a jagged reservoir inflow. The performance of the method is evaluated by both synthetic data and real cases. By using floods generated synthetically by an ideal model in which the true values and errors are known, the method is shown to be efficient as theoretically expected. The results show that the method is efficient and universally applicable in different cases. And the degree of forecast improvement is positively related to the fluctuation coefficient of the floods. The more severe the fluctuation of the flood hydrograph, the more robust the method. HIGHLIGHTS A robust error estimation method for real-time flood forecasting in reservoirs is developed.; A fluctuation coefficient is proposed to quantify the degree of fluctuation of a jagged reservoir inflow.; The method is demonstrated to be efficient by an ideal model and 10 reservoir basins of different characteristics.; The performance of the proposed method is positively related to the fluctuation coefficient of the floods.;

    Abstract TurboID-based proximity labeling coupled to mass spectrometry (PL-MS) has emerged as a powerful tool for mapping protein-protein interactions in both plant and animal systems. Despite advances in sensitivity, PL-MS studies can still suffer from false negatives, especially when dealing with low abundance bait proteins and their transient interactors. Protein-level enrichment for biotinylated proteins is well developed and popular, but direct detection of biotinylated proteins by peptide-level enrichment and the difference in results between direct and indirect detection remain underexplored. To address this gap, we compared and improved enrichment and data analysis methods using TurboID fused to SPY, a low-abundance O-fucose transferase, using an AAL-enriched SPY target library for cross-referencing. Our results showed that MyOne and M280 streptavidin beads significantly outperformed antibody beads for peptide-level enrichment, with M280 performing best. In addition, while a biotin concentration ≤ 50 μM is recommended for protein-level enrichment in plants, higher biotin concentrations can be used for peptide-level enrichment, allowing us to improve detection and data quality. FragPipe's MSFragger protein identification and quantification software outperformed Maxquant and Protein Prospector for SPY interactome enrichment due to its superior detection of biotinylated peptides. Our improved washing protocols for protein-level enrichment mitigated bead collapse issues, improving data quality, and reducing experimental time. We found that the two enrichment methods provided complementary results and identified a total of 160 SPY-TurboID-enriched interactors, including 60 previously identified in the AAL-enriched SPY target list and 100 additional novel interactors.     Abstract Direct analysis of amyloid proteins in human plasma will promote rapid screening of brain amyloidosis, the earliest pathological signature of Alzheimer's disease. We developed a microflow liquid chromatography-targeted mass spectrometry assay for quantitation of four intact β-amyloid proteins starting from 1 mL of human plasma samples. This method showed 90% accuracy for predicting brain amyloid using plasma Aβ42/Aβ40 values from 36 cognitively normal individuals in a prospective clinical study (raw data deposited in MassIVE, Data set ID MSV000087451). Our method may contribute to early diagnosis of Alzheimer's disease.

    A Deep Learning Framework for Retrieving Global Sea Surface Wind Speed Using Spaceborne GNSS-R Data

    Abstract Spaceborne Global Navigation Satellite System Reflectometry (GNSS-R) is a new remote sensing technology that uses GNSS signals reflected from the Earth’s surface to estimate geophysical parameters. Because of its unique advantages such as high temporal and spatial resolutions, low observation cost, wide coverage and all-weather operation, it has been widely used in land and ocean remote sensing fields. Ocean wind monitoring is the main objective of the recently launched Cyclone GNSS (CYGNSS). In previous studies, wind speed was usually retrieved using features extracted from delay-Doppler maps (DDMs) and empirical geophysical model functions (GMFs). However, it is a challenge to employ the GMF method if using multiple sea state parameters as model input. Therefore, in this article, we propose an improved deep learning network framework to retrieve global sea surface wind speed using spaceborne GNSS-R data, named GloWS-Net. GloWS-Net considers the fusion of auxiliary information including ocean swell significant wave height (SWH), sea surface rainfall and wave direction to build an end-to-end wind speed retrieval model. In order to verify the improvement of the proposed model, ERA5 and Cross-Calibrated Multi-Platform (CCMP) wind data were used as reference for extensive testing to evaluate the wind speed retrieval performance of the GloWS-Net model and previous models (i.e., GMF, fully connected network (FCN) and convolutional neural network (CNN)). The results show that, when using ERA5 winds as ground truth, the root mean square error (RMSE) of the proposed GloWS-Net model is 23.98% better than that of the MVE method. Although the GloWS-Net model and the FCN model have similar RMSE (1.92 m/s), the mean absolute percentage error (MAPE) of the former is improved by 16.56%; when using CCMP winds as ground truth, the RMSE of the proposed GloWS-Net model is 2.16 m/s, which is 20.27% better than the MVE method.
