Abstract Chromatin modifiers are emerging as major determinants of many types of cancers, including Anaplastic Large Cell Lymphomas (ALCL), a family of highly heterogeneous T-cell lymphomas for which therapeutic options are still limited. HELLS is a multifunctional chromatin remodeling protein that affects genomic instability by participating in the DNA damage response. Although the transcriptional function of HELLS has been suggested, no clues on how HELLS controls transcription are currently available. In this study, by integrating different multi-omics and functional approaches, we characterized the transcriptional landscape of HELLS in ALCL. We explored the clinical impact of its transcriptional program in a large cohort of 44 patients with ALCL. We demonstrated that HELLS, loaded at the level of intronic regions of target promoters, facilitates RNA Polymerase II (RNAPII) progression along the gene bodies by reducing the persistence of co-transcriptional R-loops and promoting DNA damage resolution. Importantly, selective knockdown of HELLS sensitizes ALCL cells to different chemotherapeutic agents, showing a synergistic effect. Collectively, our work unveils the role of HELLS in acting as a gatekeeper of ALCL genome stability providing a rationale for drug design.

Abstract Total hip arthroplasty (THA) is one of the most common surgical procedures in orthopedics; however, it is subjected to different kinds of failures, one of them being dislocation. Many different prosthetic designs have been developed to overcome this problem, such as dual mobility coupling. The main purpose of this article is to determine whether there are differences regarding the revision surgery of unstable THA comparing the risk of failure between dual mobility cup (DMC) implants, standard implants, and among different head sizes. A registry-based population study has been conducted by analyzing data collected by the Emilia Romagna Registry of Orthopedic Prosthetic Implants (RIPO), including a total of 253 implants failed for dislocation and instability that were operated on by cup revision surgery between 2000 and 2019. The selected population has been divided into two groups based on the insert type: standard and DMC. The age at revision surgery was significantly lower in the standard cup group with respect to DMC (

Abstract In this article, we present a link budget analysis for Global Navigation Satellite System (GNSS) signals scattered off the sea surface in arbitrary acquisition geometries. The aim of our study is to investigate the reliability of the Geometrical Optics (GO) scattering model, which accurately describes sea surface scattering at and near the specular reflection direction, in properly modeling the sea surface return in far-from-specular acquisition geometries, which are of interest for maritime surveillance purposes and where GO is expected to fail. To this end, we adopted the recent Bistatic Anisotropic Polarimetric Two-Scale Model (BA-PTSM), which revealed good agreement with advanced scattering models, such as the second-order Small Slope Approximation (SSA2), regardless of the acquisition geometry, with the advantage of a reduced computational complexity. Numerical results have been derived for both circular polarization channels and for both spaceborne and airborne GNSS-Reflectometry (GNSS-R). It has been shown that, as long as conventional GNSS-R processing is assumed, GO can be safely adopted for simulation and analysis of spaceborne GNSS-R data regardless of the acquisition geometry and sea state, whereas more accurate scattering models, e.g., BA-PTSM, should be used for airborne receivers in far-from-specular configurations.

Abstract This study investigated the effects of germinated millet flour on adipogenesis, insulin resistance, glucose tolerance and thyroid function in Wistar rats fed with a high-fat high-fructose diet (HFHF). The experiment was divided into two phases. Phase 1: control group, which received an AIN-93M diet (n = 10) and HFHF group (n = 20), which received a diet rich in saturated fat (31%) and fructose (20%), for eight weeks. Phase 2: intervention: the control group maintained the AIN-93M diet (n = 10) and the HFHF group was divided into two groups: the HFHF (n = 10) and the germinated millet group (n = 10), for 10 weeks. The germinated millet flour maintained (p > 0, 05) the plasma levels of thyroid hormones, increased (p < 0.05) the insulin receptor (INSR) mRNA expression, protein kinase B (AKT) mRNA expression and the phospho-AKT1 protein concentration, phosphofructokinase (PFK) mRNA, pyruvate kinase (PK) mRNA and activated protein kinase (AMPK) mRNA expression, and the brown adipose tissue and reduced (p < 0.05) the glucose triglyceride index (TyG), glucose, insulin, HOMA-IR and hypercorticosteronemia, compared to the HFHF group. These effects contributed to reduce the gluconeogenesis, hyperinsulinemia and adiposity. Thus, germinated millet flour is a good alternative for modulating the adipogenesis and glucose metabolism, without interfering with the thyroid hormones, in rats with an insulin resistance condition with a high-fat high-fructose diet.

Abstract The revised IPSS (IPSS-R) was developed aiming at a better prognostication, taking into account patients treated with best supportive care. We herein validated this model on the basis of data from 1314 patients who received BSC only as well as patients who underwent induction chemotherapy (n=214) or allogeneic transplantation (n=167). We could demonstrate a clear distinction of the IPSS-R risk categories with regard to survival and risk of AML evolution in all patient cohorts. When comparing IPSS-R, IPSS, WHO prognostic scoring system (WPSS) and Duesseldorf score, the best results regarding the ability to predict survival were obtained by the IPSS-R.

Abstract Given a set of points on the plane and an assignment of values to them, an optimal linear partition is a division of the set into two subsets which are separated by a straight line and maximally contrast with each other in the values assigned to their points. We present a method for inspecting and rating all linear partitions of a finite set, and a package of three functions in the R language for executing the computations. One function is for finding the optimal linear partitions and corresponding separating lines, another for graphically representing the results, and a third for testing how well the data comply with the linear separability condition. We illustrate the method on possible data from a psychophysical experiment (concerning the size-weight illusion) and compare its performance with that of linear discriminant analysis and multiple logistic regression, adapted to dividing linearly a set of points on the plane.

Abstract A search for anomalous production of events with three or more isolated leptons and bottom-quark jets produced in pp collisions at √s=8 TeV is presented. The analysis is based on a data sample corresponding to an integrated luminosity of 19.5 fb(-1) collected by the CMS experiment at the LHC in 2012. No excess above the standard model expectations is observed. The results are interpreted in the context of supersymmetric models with signatures that have low missing transverse energy arising from light top-squark pair production with R-parity-violating decays of the lightest supersymmetric particle. In two models with different R-parity-violating couplings, top squarks are excluded below masses of 1020 GeV and 820 GeV when the lightest supersymmetric particle has a mass of 200 GeV.
