    Abstract Objective: To investigate the expression and clinical significances of HGFA, Matriptase, HAI-1 and HAI-2 in patients with acute myeloid leukemia (AML).
    Methods: The bone marrow samples from 91 AML patients, 41 AML patients in complete remission, and 32 normal controls were collected. Real time fluorescence quantitative RT-PCR (qRT-PCR) was used to detect the mRNA expressions levels of HGFA, Matriptase, HAI-1, HAI-2 . The expressions of these genes were compared among AML untreated group, the complete remission group and the healthy control group. The correlation of their expression with clinical characteristics was analyzed.
    Results: The level of HGFA in the AML untreated group was higher than that in the healthy control group(P<0.05), while the HAI-2 mRNA level was lower than that in the healthy control group(P<0.05). The mRNA levels of HAI-1 and Matriptase were not changed significantly in all groups. The HAI-2 mRNA expression level was significantly lower in the high white blood cell group (P<0.05).
    Conclusion: The abnormal activation of HGF/c-Met signaling system in AML may result from the increase of HGFA expression and the decrease of HAI-2 expression of the upstream regulatory factors.

    Abstract Climate change impacts crop productivity as atmospheric conditions and water supply change, particularly in intensive cropping areas. This study used the validated AquaCrop Model, which was run with downscaled daily climate data produced by SDSM and CanESM2. The changes in the potential grain yield of winter wheat and summer maize and water budget during the cropping seasons were estimated for the Huang-Huai-Hai Plain (3H Plain) under RCP4.5 and RCP8.5 scenarios. The results show that the potential yield of winter wheat is increasing with similar spatial patterns in the 2030s, 2050s, and 2080s, with much of the increase is distributed in Shandong and northeastern parts of Henan. During the winter wheat growth period, the water budget deficit will likely improve from −210 mm in the 2030s to −202 mm in 2080s under RCP4.5 and from −206 mm in the 2030s to −191 mm in 2080s under RCP8.5 across the 3H Plain. The water budget during the winter wheat period will continue to be in deficit in the north 3H Plain and improvements are estimated mostly in the lower southern areas of the Plain. The summer maize potential yield is estimated to increase from the baseline period, but yields will decrease by 0.81%, 1.19%, and 2.10% in the 2030s, 2050s, and 2080s, respectively, under RCP8.5 compared to RCP4.5. During the summer maize growth period, the water budget is also estimated to improve from 109 mm in 2030s to 126 mm in 2080s under RCP4.5 and 107 mm in the 2030s to 163 mm in 2080s under RCP8.5. This increase is mainly estimated in the central and south of the 3H Plain. The estimated ETc of winter wheat shows no significant decrease, while the reduction of 6 mm and 13 mm for summer maize is observed under RCP4.5 and RCP8.5, respectively. The study provides scientific evidence to devise adaptation and mitigation climate change strategies for agricultural productivity and water resource management.

    Abstract Biochar can significantly enhance soil organic carbon (SOC) and crop yield, and it is therefore the preferred material for soil improvement in medium-low yield fields. In this study, a field experiment was designed to explore the impacts of biochar application on SOC content and fraction composition. Results indicated that incorporation of biochar into soil increased the SOC content by 26.9%-65.3% in the surface layer (0-10 cm) and 30.3%-63.0% in the subsurface layer (10-20 cm) of soil, while water-soluble organic carbon (WSOC) of the two layers was increased by 2.2-40.0% and 2.3-39.8%, respectively. Microbial biomass carbon decreased under conventional nitrogen treatments and increased with biochar addition under increased nitrogen application.     Abstract The Huang-huai-hai River Basin is one of the most economically developed areas, but is also heavily impacted by drought and flood disasters. Research on the precipitation feature of the Huang-huai-hai River Basin is of great importance to the further discussion of the cause of flood disaster. Based on the selected meteorological stations of the study area from 1961-2016, the inverse distance weighting method was used to get daily precipitation grid data. Interannual variation of precipitation intensity and cover area of different precipitation classes was analyzed. The generalized extreme-value distribution method was used to analyze the spatial distribution of extreme precipitation. The results show that: (1) decrease of accumulated precipitation in light precipitation year and moderate precipitation year might be the reason why the precipitation in the whole basin decreased, but the coefficient of variation (CV) of different classes of precipitation and precipitation days does not change significantly; (2) since the cover area of precipitation > 50 mm and precipitation intensity both decreased, the extreme precipitation of the whole basin may be decreasing; (3) extreme precipitation mainly occurred in the loess plateau in the northeast of Huang-huai-hai River Basin, Dabieshan in the middle of Huang-huai-hai River Basin and other areas.

    Abstract Matriptase is ectopically expressed in neoplastic B-cells, in which matriptase activity is enhanced by negligible expression of its endogenous inhibitor, hepatocyte growth factor activator inhibitor (HAI)-1. HAI-1, however, is also involved in matriptase synthesis and intracellular trafficking. The lack of HAI-1 indicates that other related inhibitor, such as HAI-2, might be expressed. Here, we show that HAI-2 is commonly co-expressed in matriptase-expressing neoplastic B-cells. The level of active matriptase shed after induction of matriptase zymogen activation in 7 different neoplastic B-cells was next determined and characterised. Our data reveal that active matriptase can only be generated and shed by those cells able to activate matriptase and in a rough correlation with the levels of matriptase protein. While HAI-2 can potently inhibit matriptase, the levels of active matriptase are not proportionally suppressed in those cells with high HAI-2. Our survey suggests that matriptase proteolysis might aberrantly remain high in neoplastic B-cells regardless of the levels of HAI-2.

    Abstract Objective: To investigate the clinical significance of dynamic monitoring of VEGF, SECAD, HCY and TNF-α in serum levels in patients with gastric cancer. Methods: A total of 94 patients with gastric cancer treated in our hospital from February 2013 to September 2014 were selected as the subjects. Sixty healthy subjects were selected as control group. VEGF, SE-CAD, HCY and TNF-α in serum levels were measured by cyclic enzyme method and enzyme-linked immunosorbent assay (ELISA). The correlation between the serum index and prognosis was analyzed. Results: The levels of Hcy, VEGF, SE-CAD and TNF-α in serum of gastric cancer group were significantly higher than those in control group. 1 month after surgery, levels of Hcy, VEGF, SE-CAD and TNF-α were still higher than those in control group. 3 months after surgery, Hcy, SE-CAD, TNF-α levels reached to the normal range, while VEGF was still higher than the normal range. The levels of Hcy, VEGF, SE-CAD and TNF-α in relapse group were significantly higher than those in non-recurrence group after 3 years of follow-up. Conclusion: The results show that the changes of serum Hcy, VEGF, SECAD and TNF-α level in patients with gastric cancer before and after operation were related to the disease condition and prognosis. The dynamic monitoring of the changes of Hcy, VEGF and TNF-α was helpful to the observation and prognosis.

    Abstract Biochar can significantly enhance soil organic carbon (SOC) and crop yield, and it is therefore the preferred material for soil improvement in medium-low yield fields. In this study, a field experiment was designed to explore the impacts of biochar application on SOC content and fraction composition. Results indicated that incorporation of biochar into soil increased the SOC content by 26.9%–65.3% in the surface layer (0–10 cm) and 30.3%–63.0% in the subsurface layer (10–20 cm) of soil, while water-soluble organic carbon (WSOC) of the two layers was increased by 2.2–40.0% and 2.3–39.8%, respectively. Microbial biomass carbon decreased under conventional nitrogen treatments and increased with biochar addition under increased nitrogen application. The C:N value increased with biochar application, while the water-soluble C:N value of soil applied with 30 t ha⁻¹ biochar was lower than that of soil applied with 15 t ha⁻¹ biochar, both in the two tested soil layers. Wheat yield is evidently correlated with SOC, with the correlation coefficients of 0.919 and 0.952 in the surface and subsurface soil layers (P < 0.01), respectively. Particularly, increasing fulvic and humic acid-like compounds of WSOC promoted the bioavailability of nutrient elements, thereby increasing the crop yields. Therefore, biochar application is an effective means to fertilize middle-low yield soils through increasing SOC sequestration and nutrient reserves, or adjusting soil C:N value to a proper range, thereby reducing nutrient loss and increasing wheat yield.
