    Abstract TatD DNase, a key enzyme in vertebrates and invertebrates, plays a pivotal role in various physiological processes. Dugesia japonica (D. japonica), a flatworm species, has remarkable regenerative capabilities and possesses a simplified immune system. However, the existence and biological functions of TatD DNase in D. japonica require further investigation. Here, we obtained the open reading frame (ORF) of DjTatD and demonstrated its conservation. The three-dimensional structure of DjTatD revealed its active site and binding mechanism. To investigate its enzymological properties, we overexpressed, purified, and characterized recombinant DjTatD (rDjTatD). We observed that DjTatD was primarily expressed in the pharynx and its expression could be significantly challenged upon stimulation with lipopolysaccharide, peptidoglycan, gram-positive and gram-negative bacteria. RNA interference results indicated that both DjTatD and DjDN2s play a role in pharyngeal regeneration and may serve as functional complements to each other.     Abstract Targeted protein degradation (TPD) technologies, particularly proteolysis-targeting chimeras (PROTACs), have emerged as a significant advancement in drug discovery. However, several hurdles - such as the difficulty of identifying suitable ligands for traditionally undruggable proteins, poor solubility and impermeability, nonspecific biodistribution, and on-target off-tissue toxicity - present challenges to their clinical applications. Aptamers are promising ligands for broad-ranging molecular recognition. Utilizing aptamers in TPD has shown potential advantages in overcoming these challenges. Here, we provide an overview of recent developments in aptamer-based TPD, emphasizing their potential to achieve targeted delivery and their promise for the spatiotemporal degradation of undruggable proteins. We also discuss the challenges and future directions of aptamer-based TPD with the goal of facilitating their clinical applications.

    Abstract In this study, dense anticorrosion magnesium-aluminum layered double hydroxide (MgAl-LDH) films were prepared for the first time by introducing a cationic surfactant tetradecyltrimethyl ammonium bromide (TTAB) in the process of in situ hydrothermal synthesis of Mg-Al LDH films on an AZ31 magnesium alloy.     Abstract Mitochondrial myopathy is a group of multi-system diseases in which mitochondrial DNA (mtDNA) or nuclear DNA (nDNA) defects lead to structural and functional dysfunction of mitochondria. The clinical manifestations of mitochondrial myopathy are complex and varied, and the testing for mtDNA and nDNA is not widely available, so misdiagnosis or missed diagnosis is common. Chronic progressive external ophthalmoplegia (CPEO) is a common type of mitochondrial myopathy, which is characterized by blepharoptosis. Here we report a 38-year-old female with mitochondrial myopathy presented with chronic numbness and weakness of the limbs, accompanied by blepharoptosis that was recently noticed. Laboratory and head magnetic resonance imaging (MRI) examinations showed no obvious abnormalities. Muscle and nerve biopsies showed characteristic ragged red fibers (RRFs) and large aggregates of denatured mitochondria. Testing for mtDNA and nDNA showed a known mutation c.2857C>T (p.R953C) and a novel variant c.2391G>C (p.M797I) in the polymerase gamma (

    Abstract The attitude closed-loop control of the parallel platform in the working space needs to determine the relationship between the pose of the top moving platform and the length of each mechanical arm, that is, the kinematics problem of the parallel platform. In this study, the kinematics model of the six-degree-of-freedom parallel platform was established. The kinematics forward solution algorithm based on Newton–Raphson iteration was studied. The kinematics forward solution method usually adopts a numerical solution, which often needs multiple iterations, and the algorithm has a poor real-time performance. In order to improve the real-time performance of the parallel platform control system, a multivariate polynomial regression kinematics forward solution algorithm is proposed in this paper. Moreover, by combining the multivariate polynomial regression with the Newton iterative method, we obtained an efficient solution algorithm with controllable solution accuracy. The effectiveness of the proposed method was verified by simulation tests and physical tests.

    Abstract The distributed edge storage system can store data collected at the edge of the network in a decentralised manner, with low latency, high security, and flexibility. Traditional edge-distributed storage systems only consider one single factor, such as node capacity, when storing data, ignoring network and storage node load conditions that affecting the system's read/write performance. At the same time, it could be more scalable in the widely used wireless terminal application scenarios. To tackle these challenges, this paper proposes an innovative software-defined edge storage architecture based on SDN (Software-Defined Networking) and SMB (Server Message Block) protocols, A data storage node selection algorithm that integrates the network state and storage node load state is designed based on multi-attribute decision model, and a system prototype is realised in conjunction with 5G wireless communication technology. Experimental results demonstrate significant improvements in the performance of high-load write operations compared to traditional edge-distributed storage systems. The proposed wireless distributed edge storage system also demonstrates superior scalability and adaptability, effectively addressing the challenge of limited system scalability and improving compatibility with edge scenarios in mobile applications. In addition, it results in cost savings in hardware deployment and presents a promising advancement in edge storage technology.
