    Abstract Information relating to errors in evapotranspiration (ET) products, including satellite-derived ET products, is critical to their application but often challenging to obtain, with a limited number of flux towers available for the sufficient validation of measurements. Triple collocation (TC) methods can assess the inherent uncertainties of the above ET products using just three independent variables as a triplet input. However, both the severity with which the variables in the triplet violate the assumptions of zero error correlations and the corresponding impact on the error estimation are unknown. This study proposed a cross-correlation analysis approach to discover the optimal triplet of satellite-derived ET products with regard to providing the most reliable error estimation. All possible triple collocation solutions for the same product were first evaluated by the extended triple collocation (ETC), among which the optimum was selected based on the correlation between ETC-based and in-situ-based error metrics, and correspondingly, a statistic experiment based on ranked triplets demonstrated how the optimal triplet was valid for all pixels of the product. Six popular products (MOD16, PML_V2, GLASS, SSEBop, ERA5, and GLEAM) that were produced between 2003 to 2018 and which cover China’s mainland were chosen for the experiment, in which the error estimates were compared with measurements from 23 in-situ flux towers. The findings suggest that (1) there exists an optimal triplet in which a product as an input of TC with other collocating inputs together violate TC assumptions the least; (2) the error characteristics of the six ET products varied significantly across China, with GLASS performing the best (median error: 0.1 mm/day), followed by GLEAM, ERA5, and MOD16 (median errors below 0.2 mm/day), while PML_V2 and SSEBop had slightly higher median errors (0.24 mm/day and 0.27 mm/day, respectively); and (3) removing seasonal variations in ET signals has a substantial impact on enhancing the accuracy of error ...

    Abstract Background: This paper aimed to identify the key genes and potential mechanisms of renal fibrosis, and provide methods of evaluation and new ideas for the early diagnosis and treatment of renal fibrosis.
    Methods: The GSE102515 dataset was searched from the Gene Expression Omnibus (GEO) database was searched, the differential genes were screened out, and the down-regulated and up-regulated genes were identified. Enrichment analysis of differential genes in the development of renal fibrosis was carried out using the DAVID database, differential genes were analyzed using the STRING database, and Cytoscape software was used for visual processing.
    Results: Eighteen up-regulated genes and ten down-regulated genes were screened. Differential genes are mainly involved in the integrin-mediated signaling pathway and mitotic sister chromatid binding, etc. We found that the molecular functions (MFs) of the differential genes are phospholipid binding and regulatory region DNA binding, etc. Moreover, the cellular components (CCs) of the differential genes are mainly related to low-density lipoprotein (LDL) particles and nuclei. The survey also collected data on students’ perceptions through their evaluation of and satisfaction with current online learning.Setting In response to the educational challenges created by COVID-19, medical schools in China have adopted formal online courses for students.Participants The questionnaire was sent to 225 329 students, of whom 52.38% (118 080/225 329) replied, with valid data available for 44.18% (99 559/225 329).Methods Pearson correlations and t-tests were used to examine the relationship between familiarity and PU. Multiple linear regression and logistic regression analyses were used to determine the impact of prior learning experiences and its interactions with gender, area, learning phase and academic performance on students’ perceptions.Results Students’ PU had a significant positive correlation with their familiarity with online learning modes (p<0.01). Students’ evaluation of and satisfaction with their current online education were positively associated with their familiarity (β=0.46, 95% CI 0.45 to 0.48, p<0.01; OR 1.14, 95% CI 1.13 to 1.14, p<0.01) with and PU (β=3.11, 95% CI 2.92 to 3.30, p<0.01; OR 2.55, 95% CI 2.37 to 2.75, p<0.01) of online learning. Higher learning phases, in which clinical practices are crucial, and high academic performance led to lower evaluation and satisfaction scores.

    Abstract Background: While CAR-T therapy has successfully treated haematological malignancies, it has proved sub-optimal for solid tumours. The main limitation is the inability of CAR-T cells to infiltrate and then proliferate within tumours.
    Method: We co-expressed IL-7 and PH20, a type of hyaluronidase, with CAR targeting GPC3 (G3CAR-7 × 20). We test the anti-tumour ability in vitro and in vivo. Moreover the capacity of infiltration and proliferation of G3CAR-7 × 20 was measured.
    Result: We found (G3CAR-7 × 20) exhibited better proliferation in vivo and in vitro than G3CAR, reduced the level of apoptosis after stimulation by tumour cells, and maintained the memory phenotype of CAR-T cells. G3CAR-7 × 20 also increased the ability of CAR-T cells to infiltrate tumour tissue.
    Conclusion: co-expressed IL-7 and PH20 may significantly enhance the efficacy of targeted GPC3 CAR-T cells in solid tumours treatment.

    Abstract Circular RNAs (circRNAs) have been demonstrated to play important roles in cancer progress. However, the roles in hepatocellular carcinoma (HCC) are still unclear. Here, we found has_circRNA_001306 (circ_1306) was up-regulated in HCC tissues and cell lines. Knockdown the expression circ_1306 significantly suppressed HCC cell proliferation and induced the cell apoptosis in vitro and in vivo. Furthermore, we identified circ_1306 could up-regulate the expression of CDK16 by sponging miR-584-5p. The expression of miR-584-5p was decreased, and the expression of CDK16 was increased in HCC tissues and cell lines. Meanwhile, either knockdown of miR-584-5p or overexpression of CDK16 could suppress the HCC cell proliferation. In vivo, overexpression of miR-584-5p or knockdown of circ_1306 could inhibit the expression of CDK16, and suppress tumour growth. Altogether, our findings suggested that circ_1306 could promoter HCC progress by miR-584-5p/CDK16 axis, which provided a novel marker for HCC diagnosis and treatment.

    Abstract Discrete fracture models are used for investigating precise processes of groundwater flow in fractured rocks, while a disc-shaped parallel-plates model for a single fracture is more reasonable and efficient for computational treatments. The flow velocity has a large spatial differentiation which is more likely to produce non-linear flow and additional head losses on and nearby intersections in such shaped fractures, therefore it is necessary to understand and quantify them. In this study, both laboratory experiments and numerical simulations were performed to investigate the total head loss on and nearby the intersections as well as the local head loss exactly on the intersections, which were not usually paid sufficient attention or even ignored. The investigation results show that these two losses account for 29.17%-84.97% and 0-73.57% of the entire total head loss in a fracture, respectively. As a result, they should be necessarily considered for groundwater modeling in fractured rocks. Furthermore, both head losses become larger when aperture and flow rate increase and intersection length decreases. Particularly, the ratios of these two head losses to the entire total head loss in a fracture could be well statistically explained by power regression equations with variables of aperture, intersection length, and flow rates, both of which achieved high coefficients of determination. It could be feasible through this type of study to provide a way on how to adjust the groundwater head from those obtained by numerical simulations based on the traditional linear flow model. Finally, it is practicable and effective to implement the investigation approach combining laboratory experiments with numerical simulations for quantifying the head losses on and nearby the intersections between disc-shaped fractures.

    Abstract Changping Plain, located in the northwest of Beijing, has become an important groundwater recharge area for the Beijing Plain and an important source for the urban water supply. In this study, groundwater samples were collected during the dry and wet seasons in 2015 from 24 monitoring wells distributed in Changping Plain. A Piper-Tri-linear diagram, a Schoeller diagram, a Gibbs diagram, and the isotope technique were used to investigate the temporal and spatial variations in the concentrations of groundwater hydrochemicals and the sources of groundwater recharge. The results indicated: 1) seasonal variations in the concentrations of , Ca2+, and Na+ were significant; the spatial variations of these ions were more dramatic in the dry season than in the wet season due to the dilution effect of precipitation; 2) Most groundwater samples had a HCO3-Ca-Mg based hydrochemical type and a few had a HCO3-Na-K based hydrochemical type; the hydrochemical type tended to evolve from HCO3-Ca-Mg based to HCO3-Na-K based in some monitoring wells that showed distinct seasonal variation; 3) the groundwater in the study area originated mainly from atmospheric precipitation, and it is affected by evaporation and concentration processes.
