    Abstract Cistanche deserticola Y. C. Ma (CD), known as "desert ginseng", has been found to have hepatoprotective effect. This research aimed to investigate the quality control and its alleviating effect on alcoholic liver injury in mice. In this study, for the first time, a sensitive and efficient ultra-high-performance liquid chromatography with quadrupole ion-trap mass spectrometry (UPLC-Q-TRAP/MS) method was developed to rapidly characterize nine representative phenylethanoid glycosides (PhGs) in the CD extract within 14 min, offering a reference for the quality control standard of this plant. In addition, we found that the CD extract significantly inhibited the weight loss, decreased the liver index, and attenuated excessive lipid deposition, inflammatory and oxidative stress in the mice liver. With the help of the high-throughput lipidomics technique, we discovered that CD markedly reversed 17 lipid metabolites and their involved linoleic acid, arachidonic acid and glycerophospholipid metabolic pathways. As these metabolites are mainly associated with lipid metabolism and liver damage, we further used molecular biological tests to found that CD could regulate the upstream genes and proteins of the lipid metabolism pathway, including adenosine 5'-monophosphate-activated protein kinase (AMPK), sterol regulatory element binding protein-1c (SREBP-1c), fatty acid synthase (FAS), and peroxidase proliferators activate receptors α (PPARα). In conclusion, this study elucidates the modulatory effects of CD on lipid metabolism disorders in alcoholic fatty liver from holistic system and provides a reference for further research and development of CD as a therapeutic agent.

    Abstract CRISPR/Cas9-based cytosine base editors (CBEs) and adenine base editors (ABEs) can efficiently mediate C-to-T/G-to-A and A-to-G/T-to-C substitutions, respectively; however, achieving base transversions (C-to-G/C-to-A and A-to-T/A-to-C) is challenging and has been rarely studied in plants. Here, we constructed new plant C-to-G base editors (CGBEs) and new A-to-Y (T/C) base editors and explored their base editing characteristics in rice. First, we fused the highly active cytidine deaminase evoFENRY and the PAM-relaxed Cas9-nickase variant Cas9n-NG with rice and human uracil DNA N-glycosylase (rUNG and hUNG), respectively, to construct CGBE-rUNG and CGBE-hUNG vector tools. The analysis of five NG-PAM target sites showed that these CGBEs achieved C-to-G conversions with monoallelic editing efficiencies of up to 27.3% in T

    Abstract A chemical investigation of the endophytic fungus Diaporthe destruens from the Hernandiaceae plant Illigera orbiculata C. Y. Wu collected from southern Yunnan Province, China, led to the isolation of six undescribed compounds, including two azaphilone analogs, which are a pair of epimers (13R-hydroxy-chermesinone A and 13S-hydroxy-chermesinone A); a pyrrole derivative (1-(4-(methoxymethyl)-1H-pyrrol-3-yl)ethan-1-one); an isoindolone derivative (4-hydroxy-6-methoxyisoindolin-1-one); a benzylbenzene derivative (destruensine A) and a conjectural fragment of polyketide ((2R,4R)-2-(methoxymethyl)pentane-1,4-diol) along with nine known compounds. Their structures were elucidated by spectroscopic methods and HRESIMS, and the absolute configurations were further confirmed by electronic circular dichroism (ECD) and chemical derivatization. The antimicrobial activities, anti-acetylcholinesterase activities, antiproliferation, and NO production inhibitory effects of compounds 1-15 were evaluated.

    Abstract Cistanche deserticola Y.C. Ma has long been used for medical purposes in China. It mainly grows in the Chinese provinces of Inner Mongolia, Ningxia, Gansu, and Xinjiang, and the species in the Alxa region of northwest China, have the most distinct qualities. To explain the geoherbalism quality and geographical distribution of C. deserticola, we sampled 65 wild plants in Alxa, determined their echinacoside and acteoside content, and assessed the relationship between the ecological environment and quality of C. deserticola through maximum entropy modeling and geographic information system. We identified the areas suitable for the growth of high-quality C. deserticola species. The regionalization analysis of growth suitability showed that the most influential ecological factors for the growth of C. deserticola are soil type, annual sunshine duration, altitude, temperature seasonality (standard deviation ×100), vegetation type, sunshine duration in the growing season, mean precipitation in August and mean temperature in July. The most suitable areas for growing C. deserticola are southeast of Ejin Banner, central Alxa Right Banner, and north of Alxa Left Banner. The regionalization analysis of quality suitability showeds that the most influential ecological factors for glycosides in C. deserticola are sunshine duration in June, average precipitation in May, and average temperature in March, and the best-quality C. deserticola grows in Dalaihob Town, Ejin Banner. Upon inspection, the result of the experiment reached a high accuracy of 0.994, which indicates that these results are consistent with the actual distribution of C. deserticola in Alxa. The results of this study may serve as a scientific basis for site selection of artificial planting bases for C. deserticola.

This method is a highly attractive alternative to the approach of traditional formylation.

Here, we constructed new plant C-to-G base editors (CGBEs) and new A-to-Y (T/C) base editors and explored their base editing characteristics in rice. First, we fused the highly active cytidine deaminase evoFENRY and the PAM-relaxed Cas9-nickase variant Cas9n-NG with rice and human uracil DNA N-glycosylase (rUNG and hUNG), respectively, to construct CGBE-rUNG and CGBE-hUNG vector tools. The analysis of five NG-PAM target sites showed that these CGBEs achieved C-to-G conversions with monoallelic editing efficiencies of up to 27.3% in T 0 rice, with major byproducts being insertion/deletion mutations. Moreover, for the A-to-Y (C or T) editing test, we fused the highly active adenosine deaminase TadA8e and the Cas9-nickase variant SpGn (with NG-PAM) with Escherichia coli endonuclease V (EndoV) and human alkyladenine DNA glycosylase (hAAG), respectively, to generate ABE8e-EndoV and ABE8e-hAAG vectors. An assessment of five NG-PAM target sites showed that these two vectors could efficiently produce A-to-G substitutions in a narrow editing window; however, no A-to-Y editing was detected. Interestingly, the ABE8e-EndoV also generated precise small fragment deletions in the editing window from the 5′-deaminated A base to the SpGn cleavage site, suggesting its potential value in producing predictable small-fragment deletion mutations. Overall, we objectively evaluated the editing performance of CGBEs in rice, explored the possibility of A-to-Y editing, and developed a new ABE8e-EndoV tool, thus providing a valuable reference for improving and enriching base editing tools in plants.

    Abstract Ethnopharmacological relevance: Curcuma wenyujin is a multifunctional medicinal plant belonging to the ginger family (Zingiberaceae). It has been used to treat blood stasis, promote the flow of qi, dredge the meridians, and relieve pain for more than 1500 years. Its raw rhizomes, steamed rhizomes, and steamed roots constitute three herbal medicines currently listed in the Chinese Pharmacopoeia: pian-jiang-huang (), wen-e-zhu () and wen-yu-jin (), respectively.
    Aim of the review: The aim of this review was to comprehensively summarize the traditional use, phytochemistry, and pharmacology of C. wenyujin in order to provide theoretical support for its further investigation and utilization.
    Materials and methods: Multiple databases (Scifinder, CNKI, Web of Science, PubMed, Google Scholar, and Baidu Scholar) were searched. Some information was also obtained from the literatures on traditional Chinese medicine.
    Results: A total of 169 compounds have been isolated from C. wenyujin so far. Sesquiterpenoids are the major constituents and are crucial chemotaxonomic markers. Modern pharmacological studies and clinical trials have demonstrated that the extracts or active compounds from C. wenyujin have anti-inflammatory, antitumor, antioxidant, antibacterial, antiviral, and hepatoprotective properties.
    Conclusions: Until now, significant progress has been witnessed in phytochemistry and pharmacology of C. wenyujin. Some traditional uses of C. wenyujin have been supported by modern pharmacological studies. However, the establishment of quality control standards and additional clinical studies are warranted.
