Abstract Background: Burnout among emergency health care professionals is well-described, especially during the COVID-19 pandemic. Prevention interventions, such as mindfulness, focus on the management of stress.
Objective: To evaluate the effects of the FIRECARE program (a mindfulness intervention, supplemented by heart coherence training and positive psychology workshops) on burnout, secondary stress, compassion fatigue, and mindfulness among advanced life support ambulance staff of the Paris Fire Brigade.
Materials and methods: We used a non-randomized, two-group quasi-experimental study design with a waitlist control and before-and-after measurements in each group. The intervention consisted of six, once-weekly, 2.5-h sessions that included individual daily meditation and cardiac coherence practice. The study compared intervention and waitlist control groups, and investigated baseline, post-program, and 3-month follow-up change on burnout (measuring using the ProQOL-5 scale) and mindfulness (measuring using the FMI scores). Baseline burnout (measured using the ProQOL-5) was evaluated and used in the analysis.
Results: Seventy-four 74 participants volunteered to participate; 66 were included in the final analysis. Of these, 60% were classified as suffering from moderate burnout, the 'burnout cluster'. A comparison of intervention and waitlist control groups found a decrease in the burnout score in the burnout cluster (
Conclusion: FIRECARE may be a useful approach to preventing and reducing burnout among prehospital caregivers.

Title translation Physical principles of optical biopsy.
Abstract The purpose of this presentation is to explain the physical principles underlying the three main methods used to obtain images of living tissues at the cellular scale. In confocal microscopy, the tissue of interest is illuminated and scanned through a confocal aperture; a lateral resolution close to 1 microm can be obtained with high numerical aperture. Multiphoton microscopy uses a high-power short-pulse laser with instantaneous irradiance sufficient to excite fluorescence in a very small focal volume. The concentration of natural tissue fluorophores is too low to obtain an adequate signal, so exogenous fluorophores have to be added, either locally or through the body. These fluorophores can be conjugated to a variety of biomolecules that target specific disease processes, thereby increasing diagnostic specificity. Finally, OCT (optical coherence tomography) provides high-resolution images of entire tissue volumes by using a broadband source and an interferometric configuration; the depth of field and lateral resolution are both on the micrometer scale. These methods allow images to be obtained at the cellular level, but image contrast and stability require specific adjustment for their significance to be established with respect to conventional biopsy methods.

Abstract The 90-kDa heat shock proteins (Hsp90) are important for stress tolerance, for newly synthesised protein folding and for the growth of various organisms. Participation of Hsp90 in the development of Apicomplexa, notably in Plasmodium falciparum and Toxoplasma gondii, has been proven. In this work, the importance of Hsp90 for Eimeria tenella, which is responsible for avian caecal coccidiosis, was studied. Our results show that E. tenella Hsp90 (EtHsp90) expression increases during infection. Immunofluorescence microscopy studies reveal a dispersed localisation of EtHsp90 during the first schizogony. Moreover, EtHsp90 is secreted by sporozoites as early as 5min after addition of FCS in a temperature-dependent manner. By using staurosporine, we invalidated the hypothesis that EtHsp90 might be a micronemal protein. Then, EtHsp90 was detected in a parasitophorous vacuole membrane. This result suggests the importance of EtHsp90 for intracellular growth of the parasite. Inhibition of EtHsp90 function using specific antibodies and geldanamicin attenuates the capacity of E. tenella to invade and grow in the host cell.
