Abstract Stem cells maintain adult tissue homeostasis under physiological conditions. Uncontrolled stem cell proliferation will lead to tumorigenesis. How stem cell proliferation is precisely controlled is still not fully understood. Phosphorylation of Yun is essential for ISC proliferation. Yun is essential for the proliferation of normal and transformed intestinal stem cells. Our mass spectrometry and biochemical data suggest that Yun can be phosphorylated at multiple residues in vivo. Interestingly, we show that the phosphorylation among these residues is likely interdependent. Furthermore, phosphorylation of each residue in Yun is important for its function in ISC proliferation regulation. Thus, our study unveils the important role of post-translational modification of Yun in stem cell proliferation.

Abstract Background: The symptoms of overactive bladder (OAB) are highly bothersome to patients. In behavioral therapy, traditional pelvic floor muscle training (PFMT) can be boring and monotonous, which can cause poor compliance. It's important to establish a new safe and effective crotch-pelvic floor functional reconstruction training method [Yun-type pelvic floor optimal training (Yun-type training)] for OAB patients.
Methods: This randomized, double-blind, crossover trial recruited a total of 150 women with mild to moderate OAB. Patients were randomly assigned to receive Yun-type training (n=83) or PFMT (n=67). The intervention group was first given Yun-type training for 6 weeks, then switched to PFMT for 6 weeks after 2 weeks of elution. Patients in the control group were first treated with PFMT for 6 weeks, and changed to the use of Yun-type training for 6 weeks after 2 weeks of elution. The primary endpoint was overactive bladder symptom scores (OABSS) after 6 and 14 weeks of Yun-type training and PFMT.
Results: Yun-type training could significantly improve OABSS compared with PFMT after 6 and 14 weeks (P<0.001). Yun-type training was associated with an improvement in urine, urgency, urge urinary incontinence (UUI), urogenital distress inventory-6 (UDI-6), patient's perception of bladder condition (PPBC), voiding volume (VV), incontinence impact questionnaire-7 (IIQ-7), female sexual function index (FSFI), sexual desire, arousal, pain, satisfaction, pelvic floor muscle tone, type I muscle strength, type II muscle strength, and sex partner satisfaction as compared with PFMT after 6 and 14 weeks. Moreover, nocturia, maximum flow rate (Q
Conclusions: Yun-type training could yield additional benefits on OAB symptoms, sexual function, and sex partner satisfaction as compared with PFMT. The superior sexual function resulting from Yun-type training can be explained by the fact that it can contract deep and superficial muscle layers, resulting in increased vaginal wall pressure and blood flow, which further improves FSFI and sex partner satisfaction. It could lead to a new non-invasive treatment for OAB patients.

Abstract The shallow neural network (SNN) is a popular algorithm in atmospheric parameters retrieval from microwave remote sensing. However, the deep neural network (DNN) has a stronger nonlinear mapping capability compared to SNN and has great potential for applications in microwave remote sensing. The Microwave Humidity and Temperature Sounder (Beijing, China, MWHTS) onboard the Fengyun-3 (FY-3) satellite has the ability to independently retrieve atmospheric temperature and humidity profiles. A study on the application of DNN in retrieving atmospheric temperature and humidity profiles from MWHTS was carried out. Three retrieval schemes of atmospheric parameters in microwave remote sensing based on DNN were performed in the study of bias correction of MWHTS observation and the retrieval of the atmospheric temperature and humidity profiles using MWHTS observations. The experimental results show that, compared with SNN, DNN can obtain better bias-correction results when applied to MWHTS observation, and can obtain higher retrieval accuracy of temperature and humidity profiles in all three retrieval schemes. Meanwhile, DNN shows higher stability than SNN when applied to the retrieval of temperature and humidity profiles. The comparative study of DNN and SNN applied in different atmospheric parameter retrieval schemes shows that DNN has a more superior performance.

Abstract The vertical evolution of the cloud effective radius ( R e ) reflects the precipitation-forming process. Based on observations from the first Chinese next-generation geostationary meteorological satellites (FY-4A, Feng Yun 4), we established a new method for objectively obtaining the vertical temperature vs. R e profile. First of all, R e was calculated using a bispectral lookup table. Then, cloud clusters were objectively identified using the maximum temperature gradient method. Finally, the R e profile in a certain cloud was then obtained by combining these two sets of data. Compared with the conventional method used to obtain the R e profile from the subjective division of a region, objective cloud-cluster identification establishes a unified standard, increases the credibility of the R e profile, and facilitates the comparison of different R e profiles. To investigate its performance, we selected a heavy precipitation event from the Integrative Monsoon Frontal Rainfall Experiment in summer 2018. The results showed that the method successfully identified and tracked the cloud cluster. The R e profile showed completely different morphologies in different life stages of the cloud cluster, which is important in the characterization of the formation of precipitation and the temporal evolution of microphysical processes.
