    Abstract In welchem Umfang werden technische Materialeffizienzinnovationen in der Produktion adoptiert, mit denen bestehende Materialeinsparungspotenziale ausgeschöpft werden können? Und welche Motive, Hemmnisse und betriebsspezifische Faktoren beeinflussen diese Adoptionsentscheidung? Auf Basis empirischer Untersuchungen anhand der Erhebung Modernisierung der Produktion werden Befähiger für eine materialeffiziente Produktion identifiziert. Das Fraunhofer-Institut für System- und Innovationsforschung ISI analysiert Entstehung und Auswirkungen von Innovationen. Es erforscht die kurz- und langfristigen Entwicklungen von Innovationsprozessen und die gesellschaftlichen Auswirkungen neuer Technologien und Dienstleistungen. Auf dieser Grundlage stellt es seinen Auftraggebern aus Wirtschaft, Politik und Wissenschaft Handlungsempfehlungen und Perspektiven für wichtige Entscheidungen zur Verfügung. Seine Expertise liegt in der breiten wissenschaftlichen Kompetenz sowie einem interdisziplinären und systemischen Forschungsansatz.

    Abstract The prognosis for many patients with acute myeloid leukemia (AML) is poor, mainly due to disease relapse driven by leukemia stem cells (LSCs). Recent studies have highlighted the unique metabolic properties of LSCs, which might represent opportunities for LSC-selective targeting. LSCs characteristically have low levels of reactive oxygen species (ROS), which apparently result from a combination of low mitochondrial activity and high activity of ROS-removing pathways such as autophagy. Due to this low activity, LSCs are highly dependent on mitochondrial regulatory mechanisms. These include the anti-apoptotic protein BCL-2, which also has crucial roles in regulating the mitochondrial membrane potential, and proteins involved in mitophagy. Here we review the different pathways that impact mitochondrial activity and redox-regulation, and highlight their relevance for the functionality of both HSCs and LSCs. Additionally, novel AML therapy strategies that are based on interference with those pathways, including the promising BCL-2 inhibitor Venetoclax, are summarized.

    Abstract Previous studies have demonstrated that highly narcissistic individuals perceive themselves as grandiose and devaluate and sometimes overvalue others. These results are mainly based on behavioural data, but we still know little about the neural correlates underlying, such as perceptional processes. To this end, we investigated event-related potential components (ERP) of visual face processing (P1 and N170) and their variations with narcissism. Participants (N = 59) completed the Narcissistic Admiration and Rivalry Questionnaire and were shown pictures of their own face, a celebrity's face, and a stranger's face. Variations of P1 and N170 with Admiration and Rivalry were analysed using multilevel models. Results revealed moderating effects of both narcissism dimensions on the ERP components of interest. Participants with either high Admiration or low Rivalry scores showed a lower P1 amplitude when viewing their own face compared with when viewing a celebrity's face. Moreover, the Self-Stranger difference in the N170 component (higher N170 amplitude in the Self condition) was larger for higher Rivalry scores. The findings showed, for the first time, variations of both narcissism dimensions with ERPs of early face processing. We related these effects to processes of attentional selection, an expectancy-driven perception, and the mobilisation of defensive systems. The results demonstrated that by linking self-report instruments to P1 and N170, and possibly to other ERP components, we might better understand self- and other-perception in narcissism.

    Abstract Eine möglichst energieeffiziente und damit auch kostengünstige Produktion - welches Unternehmen wünscht sich dies nicht? In nahezu allen Bereichen des Verarbeitenden Gewerbes wird eine effiziente Energienutzung immer wichtiger und in vielen Fällen sogar zum entscheidenden Wettbewerbsvorteil gegenüber konkurrierenden Unternehmen - gerade in Zeiten steigender Energiepreise. Doch eine energieeffiziente Fertigung entsteht nicht von heute auf morgen und bedarf neben dem Einsatz technischer Effizienzlösungen auch strategischer Managementwerkzeuge und geeigneter Qualifikationsmaßnahmen für die betroffenen Beschäftigten. Die folgenden Auswertungen mit Daten aus der Erhebung Modernisierung der Produktion 2015 können Unternehmen neue Wege aufzeigen, wie insbesondere der Einsatz von Energiemanagementsystemen und das Angebot von formellen oder informellen Qualifikationsmöglichkeiten eine effiziente Energienutzung im Betriebsalltag unterstützen und somit entscheidende Wettbewerbsvorteile ermöglichen.

    Abstract Hemophagocytic lymphohistiocytosis (HLH) is a rare but often fatal hyperinflammatory syndrome caused by an inborn or acquired error of immunity. In adults, the underlying immunodeficiency generally arises alongside severe infections, malignancies, autoimmune diseases, and immunosuppressive treatment. To analyze risk factors and outcome in adults, we conducted a multicenter retrospective study. A total of 62 adult (age ≥18 years) patients met at least one of the following inclusion criteria: (1) ≥5 of 8 HLH-2004 criteria, (2) HScore ≥ 200 plus 4 HLH-2004 criteria, or (3) mutation compatible with an HLH diagnosis. Most patients (65%) were male, and the median age at diagnosis was 53.5 years (range, 19-81 years). All patients were assigned to 4 etiologic subgroups based on their most likely HLH trigger. The survival probability of the 4 etiologic subgroups differed significantly (P = .004, log-rank test), with patients with an underlying malignancy having the worst clinical outcome (1-year survival probability of 21%). The parameters older age, malignant trigger, elevated serum levels of aspartate transferase, creatinine, international normalized ratio, lactate dehydrogenase, sCD25, and a low albumin level and platelet count at treatment initiation were significantly (P < .1) associated with worse overall survival in the univariate Cox regression model. In multivariate analysis, sCD25 remained the only significant prognostic factor (P = .005). Our results suggest that sCD25 could be a useful marker for the prognosis of patients with HLH that might help to stratify therapeutic interventions.

    Abstract CITED2 (CBP/p300-interacting-transactivator-with-an-ED-rich-tail 2) is a regulator of the acetyltransferase CBP/p300 and elevated CITED2 levels are shown in a number of acute myeloid leukemia (AML). To study the in vivo role of CITED2 in AML maintenance, AML cells were transduced with a lentiviral construct for RNAi-mediated knockdown of CITED2. Mice transplanted with CITED2-knockdown AML cells (n=4) had a significantly longer survival compared to mice transplanted with control AML cells (P<0.02). In vitro, the reduction of CITED2 resulted in increased p53-mediated apoptosis and CDKN1A expression, whereas BCL2 levels were reduced. The activation of p53 upon CITED2 knockdown is not a direct consequence of increased CBP/p300-activity towards p53, since no increased formation of CBP/p300/p53 complexes was demonstrated and inhibition of CBP/p300-activity could not rescue the phenotype of CITED2-deficient cells. Instead, loss of CITED2 had an inhibitory effect on the AKT-signaling pathway, which was indicated by decreased levels of phosphorylated AKT and altered expression of the AKT-pathway regulators PHLDA3 and SOX4. Notably, simultaneous upregulation of BCL2 or downregulation of the p53-target gene PHLDA3 rescued the apoptotic phenotype in CITED2-knockdown cells. Furthermore, knockdown of CITED2 led to a decreased interaction of p53 with its inhibitor MDM2, which results in increased amounts of total p53 protein. In summary, our data indicate that CITED2 functions in pathways regulating p53 activity and therefore represents an interesting target for AML therapy, since de novo AML cases are characterized by an inactivation of the p53 pathway or deregulation of apoptosis-related genes.

    Abstract Für die Industrie ist eine ressourceneffiziente Produktion ein wichtiges Thema. Grund dafür ist vor allem das Kosteneinsparungspotenzial. Dennoch könnten heute immer noch laut eigener Einschätzung von Industriebetrieben durchschnittlich 14 Prozent des Energiebedarfs und 6 Prozent des Materialverbrauchs in der Produktion eingespart werden. Zur Ausschöpfung dieser Effizienzpotenziale dienen technische Lösungen, wie Steuerungskonzepte zur Maschinenabschaltung oder der Einsatz neuer technologischer Fertigungsverfahren. Die vorliegenden Analysen zeigen jedoch, dass auch Managementinstrumente eine wichtige flankierende Rolle bei der nachhaltigen Verankerung von Energie- und Ressourceneffizienz im Betrieb spielen. Sie tragen zur Sensibilisierung von Beschäftigten und damit auch zur Identifikation weiterer Einsparpotenziale bei. Aufgrund ihrer vergleichsweise geringen Kosten sind solche organisatorischen Instrumente insbesondere für KMU ein niedrigschwelliger, aber dennoch wirkungsvoller Einstieg in eine ressourceneffiziente Produktion
