    Abstract Riverbed sediments are dynamic freshwater environments colonized by a great diversity of microorganisms which play important roles in supporting freshwater ecosystem by performing a vast array of metabolic functions. Recent evidence generated by HTS approaches has revealed that the structure of sediment microbial communities is influenced by natural seasonal variations in water such as temperature or streamflow as well by disturbances caused by local human activities. Here, a spatiotemporal analysis of sediment microbial distribution from São Francisco River headwaters section was conducted using Illumina 16S rRNA-V4 region amplicon sequencing in order to accomplish three major goals: (i) to investigate whether the diversity and composition of bacterial communities accessed in riverbed sediments vary in response to distinct land-use activities; (ii) to estimate whether the diversity patterns vary between the dry and wet seasons; and (iii) to evaluate whether the diversity of bacterial metabolic functions, predicted by PICRUSt2 approach, varies similarly to the estimated taxonomic diversity. Our findings revealed that bacterial communities in the sediment show differences in diversity and taxonomic composition according to the anthropic activities performed in the local environment. However, the patterns in which this taxonomic diversity is spatially structured show differences between the dry and wet seasons. On the other hand, the most changes in predicted bacterial metabolic functions were verified between sediment samples accessed in portions of the river located in protected and unprotected areas. Our findings contributed with new evidence about the impact of typical land-use practices conducted in countryside landscapes from developing countries on riverbed bacterial communities, both in their taxonomic and functional structure.

    Abstract Sustainable use of freshwater resources for human civilization needs requires the assessment and monitoring of freshwater health, and bacterial communities from riverbed sediments have been shown to be susceptible to chronic anthropogenic disturbances in freshwater ecosystems. Here, we took advantage of the occurrence of well-recognized adjacent sections from the Upper São Francisco River basin with well-recognized levels of anthropogenic activity intensity to test the applicability of sediment bacterial communities as bioindicators of impacts on freshwater ecosystems. We applied 16S amplicon sequencing to estimate the diversity and composition of bacterial communities from 12 sampling sites across the Upper São Francisco River basin, classified as being of no, low, or high intensity of anthropogenic activities, and used diversity metrics and LEfSe to compare the patterns of community structure. Our results revealed that accessed sediment environments associated with land areas with a high intensity of anthropogenic activities presented the lowest levels of community diversity, and the bacterial community compositions of these environments were significantly different from the other sampled areas. Our findings can be considered a source of evidence for the usefulness of bacterial community-based approaches as a tool for diagnosis and monitoring of ecosystem health in areas of vulnerable freshwater environments, and can even be incorporated into regular water quality programs.

    Abstract The International Conference on Viral Hepatitis 2016 brought exciting news on the treatment of viral hepatitis. The conference was mainly focused on the most recent estimates of burden for HBV and HCV; the current gaps and prospects for regional and global HCV eradication; the use of HCV treatment as prevention; and the management of difficult-to-cure hepatitis C patients, including individuals who fail on direct-acting antivirals, people who inject drugs, and those with decompensated cirrhosis or renal insufficiency. Special patient populations, such as children, pregnant women, HIV-coinfected and persons with acute hepatitis C, were addressed separately. Data from both clinical trials and real-world experience were discussed. Further debates focused on hepatic conditions that may alter the management and outcome of viral hepatitis, such as fatty liver disease, liver transplantation, and hepatocellular carcinoma.

    Abstract Cachexia is a devastating, multifactorial and often irreversible systemic syndrome characterized by substantial weight loss (mainly of skeletal muscle and adipose tissue) that occurs in around 50-80% of patients with cancer. Although this condition mainly affects skeletal muscle (which accounts for approximately 40% of total body weight), cachexia is a multi-organ syndrome that also involves white and brown adipose tissue, and organs including the bones, brain, liver, gut and heart. Notably, cachexia accounts for up to 20% of cancer-related deaths. Cancer-associated cachexia is invariably associated with systemic inflammation, anorexia and increased energy expenditure. Understanding these mechanisms is essential, and the progress achieved in this area over the past decade could help to develop new therapeutic approaches. In this Review, we examine the currently available evidence on the roles of both the tumour macroenvironment and microenvironment in cancer-associated cachexia, and provide an overview of the novel therapeutic strategies developed to manage this syndrome.

    Abstract Despite the growing body of research on chronic pain in adults with osteogenesis imperfecta (OI), there is still a lack of comprehensive understanding of the influence of psychological factors on pain experienced by individuals with this condition. This study aims to delve into the correlation between personality traits and various aspects of pain, such as frequency, intensity, appraisal, and coping mechanisms, in a significant sample of adults with OI. Additionally, the investigation seeks to identify whether certain personality profiles may be more susceptible to chronic pain within this specific population. A descriptive cross-sectional study was conducted on a sample of 418 adults diagnosed with OI. Participants completed an online survey that assessed sociodemographic and clinical variables, pain parameters, personality traits, pain appraisal, and coping strategies. Subsequently, descriptive, correlational, cluster and comparative analyses were performed. Up to 83% of the participants reported experiencing pain on a regular basis. Regarding personality dimensions, moderate scores were obtained, with no significant differences compared to the general population. Neuroticism emerged as the trait showing the most robust relationships with the evaluated variables. It positively correlated with pain intensity, frequency, and the perception of pain as threatening (P < .001). Conversely, higher levels of extraversion were associated with a reduction in pain and its threatening perception (P < .001). Finally, the cluster analysis revealed a personality profile that showed greater vulnerability in pain adaptation, characterized by high levels of neuroticism and low levels of extraversion, agreeableness, and conscientiousness. Chronic pain is prevalent in adults with OI. Personality dimensions maintain a significant relationship with this pain, acting as vulnerability or protective factors. Consequently, specific personality profiles are associated with poorer adaptation. Understanding these profiles would allow for a deeper comprehension of the pain experience in adults with OI.

    Abstract Introduction: The increasing prevalence of high-intensity sports activities, notably the burgeoning popularity of CrossFit, underscores the contemporary significance of such physical pursuits. The discernible protective impact of branched-chain amino acids on muscle fatigue and injuries is emerging as a noteworthy area of investigation.
