    Abstract As a low-cost demand-side management application, non-intrusive load monitoring (NILM) offers feedback on appliance-level electricity usage without extra sensors. NILM is defined as disaggregating loads only from aggregate power measurements through analytical tools. Although low-rate NILM tasks have been conducted by unsupervised approaches based on graph signal processing (GSP) concepts, enhancing feature selection can still contribute to performance improvement. Therefore, a novel unsupervised GSP-based NILM approach with power sequence feature (STS-UGSP) is proposed in this paper. First, state transition sequences (STS) are extracted from power readings and featured in clustering and matching, instead of power changes and steady-state power sequences featured in other GSP-based NILM works. When generating graph in clustering, dynamic time warping distances between STSs are calculated for similarity quantification. After clustering, a forward-backward power STS matching algorithm is proposed for searching each STS pair of an operational cycle, utilizing both power and time information. Finally, load disaggregation results are obtained based on STS clustering and matching results. STS-UGSP is validated on three publicly accessible datasets from various regions, generally outperforming four benchmarks in two evaluation metrics. Besides, STS-UGSP estimates closer energy consumption of appliances to the ground truth than benchmarks.

    Abstract Salivary gland hypofunction (SGH) caused by systemic disease, drugs, aging, and radiotherapy for head and neck cancer can cause dry mouth, which increases the risk of disorders such as periodontitis, taste disorders, pain and burning sensations in the mouth, dental caries, and dramatically reduces the quality of life of patients. To date, the treatment of SGH is still aimed at relieving patients’ clinical symptoms and improving their quality of life, and is not able to repair and regenerate the damaged salivary glands. Pluripotent stem cells (PSCs), including embryonic stem cells (ESCs), induced pluripotent stem cells (iPSCs), and extended pluripotent stem cells (EPSCs), are an emerging source of cellular therapies that are capable of unlimited proliferation and differentiation into cells of all three germ layers. In recent years, the immunomodulatory and tissue regenerative effects of PSCs, their derived cells, and paracrine products of these cells have received increasing attention and have demonstrated promising therapeutic effects in some preclinical studies targeting SGH. This review outlined the etiologies and available treatments for SGH. The existing efficacy and potential role of PSCs, their derived cells and paracrine products of these cells for SGH are summarized, with a focus on PSC-derived salivary gland stem/progenitor cells (SGS/PCs) and PSC-derived mesenchymal stem cells (MSCs). In this Review, we provide a conceptual outline of our current understanding of PSCs-based therapy and its importance in SGH treatment, which may inform and serve the design of future studies.

    Abstract Abstract RNA N6-methyladenosine (m6A) regulators play essential roles in diverse biological processes, including immune responses. Mounting evidence suggests that their dysregulation is intricately linked to numerous diseases. However, the role of m6A-associated genes in carotid atherosclerosis and their relationship with aging and immune cells remain unclear. Analyze the expression profiles of m6A-related genes in carotid atherosclerosis-related datasets. Based on the expression patterns of m6A-related genes, perform consistent clustering analysis of carotid atherosclerosis samples and investigate associated immune cell infiltration patterns and aging characteristics. Develop an m6A prediction model specific to carotid atherosclerosis and analyze the relationships between immune cells infiltration and aging features. The m6A methylation modification level exhibited a substantial decrease in early-stage carotid atherosclerosis samples compared to late-stage carotid atherosclerosis samples. Subsequently, two distinct m6A subtypes were defined through consensus clustering analysis, with the lower m6A modification level group showing associations with heightened immune cell infiltration and increased expression of aging-related genes. A model composed of five m6A-related genes was formulated, and the results indicated that this model possesses effective predictive and therapeutic capabilities for carotid atherosclerosis. Furthermore, the downregulation of YTHDC1 expression resulted in elevated expression of inflammatory factors and a decrease in the expression of the aging-related gene RGN. Single-cell data analysis suggests that the reduced expression of YTHDC1 may decrease the degradation of inflammation-related factors in macrophages, leading to a highly inflammatory state in the carotid artery wall. Furthermore, the sustained release of inflammatory factors may increase the expression of the aging-related gene RGN in vascular smooth muscle cells, further exacerbating the progression of atherosclerosis.     Abstract Updating the existing intensity–duration–frequency (IDF) curves (focusing on 1-24 h) to reduce the global warming risks are typically based on regional climate models (RCMs). However, uncertainties are known to be present in such updating and need quantifying, especially whether bias correction (BC) methods should be used or not is less understood. Furthermore, the updating is still challenging for flood-prone cities in Southeast Asia due to a lack of sub-daily gauge-based observations. Therefore, in this study, firstly, we investigate the uncertainties related to the use of the BC, and the choices of different RCMs, RCP scenarios, parameter estimation methods by using the ‘analysis of variance’ method. Secondly, we propose a framework for developing and updating IDF curves in the data-scarce city (with only daily gauge observations) in Cambodia (Phnom Penh City (PPC)) by utilizing the rainfall information from the nearby cities (Ho Chi Minh City (HCMC) and Can Tho City (CTC) in Vietnam), based on the scaling property of rainfall duration versus intensity. A comparison between the RCMs with and without BC reveals that BC can efficiently remove the model bias, and the changes of rainfall intensity with BC are consistent with the underlying physical mechanisms (i.e., where atmospheric circulation tends to drive a larger increase in rarer rainfall events than less rare ones), supporting the application of BC before developing IDF curves. Further quantification of different uncertainty sources reveals that RCMs account for the largest share of the total variance (41.0% and 47.9% in HCMC and CTC, respectively). It is followed by the BC, with 39.4% and 29.7% in the two cities, respectively. RCP scenarios and parameter estimation methods provide the comparable and lowest uncertainty. The projected IDF curves in PPC, developed based on the integrated rainfall information (i.e., daily rainfall in PPC and scaling properties in nearby cities), reveal an apparent upshift (e.g., 33.6–43.7% increase in 20–year rainfall intensity in the near future (2026–2045) under RCP 4.5) of the IDF curves compared with the curves in the historical period (1986–2005). However, the deep uncertainty in such projection requires us to adopt a dynamic adaption strategy that can be iteratively updated based on new information. The current framework on quantifying different uncertainty sources and updating the IDF curves for data-scarce cities can provide suggestions for similar studies in other regions globally.
