Abstract Objective: To investigate the protective effect of Jinkui Shenqi Pills (JSP) against cyclophosphamide-induced testis injury (TI) and its anti-oxidation mechanism in mice.
Methods: Thirty male mice were equally divided into a blank control, a TI model control and a JSP treatment group. The mice in the JSP treatment group were treated intragastrically with JSP and the blank controls with normal saline at 1.2 g/kg qd for 7 days, and then the animals in both the TI model control and JSP treatment groups were injected intraperitoneally with cyclophosphamide at 50 mg/kg, once a week, for 35 days, to induce testis injury. After modeling, all the mice were weighed and sacrificed, followed by detection of the serum T content, measurement of the testis weight, examination of semen parameters in the caudad epididymis, and determination of the levels of super oxide dismutase (SOD) and malondialdehyde (MDA) in the testis tissue and the expressions of relevant genes by qRT-PCR.
Results: The mice of the TI model control group, compared with the blank controls, showed significant decreases in the body weight ([34.63 ± 1.92] vs [48.32 ± 1.64] g, P<0.05), testis weight ([80.00 ± 3.90] vs [140.00 ± 6.10] mg, P<0.05), testicular organ coefficient ([0.22 ± 0.01] vs [0.31 ±0.03]%, P<0.05), sperm motility ([48.66 ± 8.08]% vs [89.33 ± 4.04]%, P<0.05), sperm concentration ([28.42 ± 5.26] vs [77.67 ± 8.73] ×10⁶/ml, P<0.05), and levels of serum T ([8.75 ± 0.96] vs [21.75 ± 1.71] pg/ml, P<0.05) and SOD ([140.82 ± 10.08] vs [358.52 ± 40.41] U/mg prot, P<0.05), but remarkable increases in the sperm deformity rate ([37.33 ± 2.08] vs [15.33±1.53]%, P<0.05) and MDA level ([54.89±6.09] vs [30.21±2.17] nmol/ng prot, P<0.05). The mice of the JSP treatment group, in comparison with the TI model controls, exhibited markedly increased body weight ([39.80±2.89] vs [34.63±1.92]g, P<0.05), testis weight ([130.00 ± 11.00] vs [80.00 ± 3.90] mg, P<0.05), testicular organ coefficient ([0.28 ± 0.01] vs [0.22 ± 0.01]%, P<0.05), sperm motility ([76.00 ± 5.29]% vs [48.66 ± 8.08]%, P<0.05), sperm concentration ([56.08 ± 4.29] vs [28.42 ± 5.26] ×10⁶/ml, P<0.05), and levels of serum T ([15.50 ± 1.29] vs [8.75 ± 0.96] pg/ml, P<0.05) and SOD ([206.59 ± 16.38] vs [140.82 ± 10.08] U/mg prot, P<0.05), but decreased sperm deformity rate ([25.01 ± 2.99]% vs [37.33 ± 2.08]%, P<0.05) and MDA level ([35.84 ± 3.61] vs [54.89 ± 6.09] nmol/ng prot, P<0.05). The mRNA expressions of NOQ-1, Nrf2 and HO-1 in the testis tissue were significantly lower and that of Caspase-3 remarkably higher in the TI model control than in the blank control group (P<0.05), while those of Nrf2 and HO-1 significantly higher and that of Caspase-3 markedly lower in the JSP treatment group than in the TI model controls (P<0.05). Histopathological images displayed reduced layers of spermatogenic cells in the seminiferous tubules, complete exfoliation of the spermatogenic cells in some of the tubules and decreased number of sperm cells in the TI model controls, which were all found normal in the JSP treatment group.
Conclusions: Jinkui Shenqi Pills can effectively inhibit cyclophosphamide-induced testis injury, which may be related to its effect of regulating the gene expression of the Nrf2 signaling pathway and enhancing the activity of antioxidant enzymes.

Abstract The aim of the study was to investigate the effects and underlying mechanism of JKSQP in a rat model of asthma with kidney-yang deficiency (KYD). Materials and Methods. Hydrocortisone (HYD) was used to establish the rat model of KYD; rats were then sensitized and challenged with ovalbumin (OVA). JKSQP was administered to OVA-challenged rats, and the changes in signs and symptoms of KYD were observed. The leukocyte number and subpopulations in bronchoalveolar lavage fluid (BALF) were counted and the cells were stained with Wright–Giemsa dye. Serum adrenocorticotropic hormone (ACTH), corticosterone (CORT), corticotropin-releasing hormone (CRH), total immunoglobulin E (IgE), and OVA-specific IgE levels were determined using relevant enzyme-linked immunosorbent assays (ELISA) kits. Results. JKSQP not only reversed the phenomenon of KYD but also significantly inhibited the number of leukocyte and eosinophils in the BALF, increasing the level of interferon (IFN)-γ and decreasing the levels of interleukin-4 (IL-4) and IgE in the serum compared with the OVA-challenged groups. Conclusions. Taken together, the antiasthma effects of JKSQP were likely mediated by the enhancement of the function of the hypothalamic-pituitary-adrenal axis and the reversal of T helper 1/2 imbalance.

Abstract On the base of establishing the fingerprint of JinKui ShenQi pills, the ultraviolet spectra-mass spectrometry/mass spectrometry, method was used to identify the fingerprint. Seperation was performed on the Symmetry Shield RP18 (5 microm, 4. 6 mm X 15 mm) analytical column with mobile phase consisting of 1% acetic acid and acetonitrile with gradient elute at the flow rate of 1.0 mL x min(-1), and the ultraviolet detection wavelength was set at 248 nm. Using the above-mentioned chromatographic condition, the fingerprint of different samples was established and the same fingerprint was defined. The fingerprints of different samples were compared with similarity evaluation software published by Pharmacopeia committee codex (2004A). The mass spectrograph with API-ESI ionization source was used, setting the flow rate at 0.5 mL x min(-1) after splitting stream. The pressure of atomization room was 50 Psi, the flow rate of dry gas was 9.0 L x min(-1), the capillary voltage was 4 kV, and the transmission voltage was 70 V. The negative scanner mode was chosen, scan scope was 100-2000, using ion trap to analyze quasimolecular ion peak and the selected fragment ion, and TIC chromatography and second order mass chromatogram were recorded. The major constituents among in JinKui ShenQi pills from different origins were separated well by HPLC. Although there was difference among different origins, they showed nineteen identical characteristic absorption bands. Three fingerprints chemical compositions such as loganin, cinnamal and paeonol were identified based on the retention time and ultraviolet spectra of standard preparation. According to their ultraviolet spectra, molecular weight and fragmentation information, ten peaks in the fingerprint were identified by ultraviolet spectroscopy-mass, spectrometry/massg spectrometry. They are 1,2,3-tri-O-galloyl-glucose, loganin, paeoniflorin, 1,2,3,6-tetro-O-galloyl-glucose, soya-cerebroside, cornuside, and PGG, benzoyl-oxypaeoniflorin. The result showed that the presented fingerprint of JinKui ShenQi pills contained plenty of information quite valuable for the quality evaluation as well as chemical characterization of JinKui ShenQi pills.

Abstract Objective: To inquire into the cerebral gene change of effect of Jingui Shenqiwan and Youguiwan for animal model of kidney-yang asthenia caused by excessive physical and sexual activities, which may study the effect mechanism of the medicine with syndrome disproved therapeutic efficacy of drugs used.
Method: Male mice of Kunming species, weight 35-40 g, and female weight 28-35 g were randomly divided into four groups: control group, model group and treatment groups of Jigui Shenqiwan and Youguiwan in which there were ten male mice, fifteen ones, ten ones and ten ones, respectively. All of them were fed normally, and poured into their stomach with 0.5 mL of distill water for each mouse in the control group and model group, and with 0.5 mL suspension of the drugs (including 1.1 g x kg(-1) drug) for each one in the treatment groups every day. The mice in the model group and treatment groups were kept by means of each male mouse with six female mice in the same cage, and all male mice swam until they gradully submerged and were scooped up from water once everyday for lasting four weeks to induce the kidney-yang asthenia with excessive physical and sexual activities. Animals' manifestation such as fearing cold, activity and responses, mouse' fur and so on were observed. The brain gene were detected with the mouse brain gene chip of 36K Mouse genome array made by Capital Bio Corp. Beijing, China, and the differential expression gene were screened according to the ratio equal to or above 2 and equal to or below 0.5 with the related fluorescent intensity comparing the two groups, which could be further verified in the light of partly differential expression gene with qRT-PCR.
Result: The mouse model of kidney-yang asthenia in the model group was successfully induced by way of excessive physical and sexual activities. There were twenty-three genes among up-regulated genes in the model group versus control group but down-regulated genes in the treatment groups versus model group, chiefly including the genes association with inflammation/immunization, neurotransmissions/ signal transduction and so on. There were six genes among the down-regulated genes in the model group versus control group but upregulated in the treatment groups versus model group, mainly involving the related genes of cellular cycle and structure, neurotransmissions/signal transduction, transcription and et al.
Conclusion: Jigui Shenqiwan and Youguiwan may make it markedly up-regulated to notably down-regulated genes of hormone and melanin-concentrating hormone (MCH) for model of mouse of kidney-yang asthenia, and promote cellular proliferation, which can inquire into effect mechanism of the drug in genetic level at the same time.

Abstract Purpose: The present study aimed to develop a nomogram to predict the prognosis of patients with secondary bone tumors in the intensive care unit to facilitate risk stratification and treatment planning.
Methods: We used the MIMIC IV 2.0 (the Medical Information Mart for Intensive Care IV) to retrieve patients with secondary bone tumors as a study cohort. To evaluate the predictive ability of each characteristic on patient mortality, stepwise Cox regression was used to screen variables, and the selected variables were included in the final Cox proportional hazard model. Finally, the performance of the model was tested using the decision curve, calibration curve, and receiver operating characteristic (ROC) curve.
Results: A total of 1028 patients were enrolled after excluding cases with missing information. In the training cohort, albumin, APSIII (Acute Physiology Score III), chemotherapy, lactate, chloride, hepatic metastases, respiratory failure, SAPSII (Simplified Acute Physiology Score II), and total protein were identified as independent risk factors for patient death and then incorporated into the final model. The model showed good and robust prediction performance.
Conclusion: We developed a nomogram prognostic model for patients with secondary bone tumors in the intensive care unit, which provides effective survival prediction information.
