    Abstract Screening for prostate cancer (PCa) using prostate-specific antigen (PSA) is common. This study aimed to identify potential nutritional variables associated with PSA levels and develop a PCa risk prediction model. A total of 5,725 participants from the 2001-2010 National Health and Nutrition Examination Survey (NHANES) database were recruited and divided into discovery and validation sets. Three machine learning (ML) algorithms were performed in discovery set to select features associated with a high-risk PSA level. Age, red blood cell (RBC), blood cholesterol, blood lead, and prognostic nutritional index (PNI) were overlapping important contributors among the three ML models. Furthermore, the relationship between the selected variables and the high-risk PSA level was verified using weighted logistic regression models and restricted cubic spline (RCS) curves. After adjustment, age and blood lead were significantly positively associated with high-risk PSA, while PNI was significantly negatively associated with high-risk PSA. A nomogram including age, RBC, blood cholesterol, and PNI was constructed for predicting the PCa risk according to PSA levels, and the area under the receiver operating characteristic curve (AUROC) of discovery and validation sets was 0.755 and 0.756, respectively. The constructed nomogram may be used for targeted PSA testing and cancer screening in community healthcare.

    Abstract Purpose: We aim to explore the predictive values of homeobox gene A-antisense transcript 3 (HOXA-AS3), cystatin 6 (CST6), and chromobox homolog 4 (CBX4) expressions in cancer tissues for the recurrence of early colon cancer after surgery.
    Patients and methods: A total of 136 patients who received surgery from January 2020 to January 2021 were enrolled and followed up for 24 months to observe the recurrence after surgery, based on which they were assigned into recurrence and non-recurrence groups. All patients underwent a histopathological examination on admission.
    Results: The recurrence group had a lower degree of differentiation as well as a higher HOXA-AS3 level and CST6 and CBX4 expression scores than those of the non-recurrence group (P<0.05). HOXA-AS3 level, CST6 expression score, and CBX4 expression score were risk factors for the recurrence of early colon cancer after surgery [odds ratio (OR)>1, P<0.05]. The receiver operating characteristic curve analysis showed that the areas under the curves of HOXA-AS3 level, CST6 expression score, CBX4 expression score, and their combination for predicting recurrence were 0.909 [95% confidence interval (95% CI): 0.785-1.000], 0.819 (95% CI: 0.690-0.948), 0.794 (95% CI: 0.663-0.926), and 0.942 (95% CI: 0.882-1.000), respectively.
    Conclusion: The expressions of HOXA-AS3, CST6, and CBX4 in cancer tissues have close correlations with the recurrence of early colon cancer after surgery and are thus of high predictive values.

    Abstract Clear cell renal cell carcinoma (ccRCC) is characterized by high heterogeneity and recurrence rates, posing significant challenges for stratification and treatment. Basement membrane-related genes (BMGs) play a crucial role in tumor initiation and progression. Clinical and transcriptomic data of ccRCC patients were extracted from TCGA and GEO databases. We employed univariate regression and LASSO-Cox stepwise regression analysis to construct a BMscore model based on BMGs expression level. A nomogram combining clinical features and BMscore was constructed to predict individual survival probabilities. Further enrichment analysis and immune-related analysis were conducted to explore the enriched pathways and immune features associated with BMGs. High-risk individuals predicted by BMscore exhibited poorer overall survival, which was consistent with the validation dataset. BMscore was identified as an independent risk factor for ccRCC. Functional analysis revealed that BMGs were related to cell-matrix and tumor-associated signaling pathways. Immune profiling suggests that BMGs play a key role in immune interactions and the tumor microenvironment. BMGs serve as a novel prognostic predictor for ccRCC and play a role in the immune microenvironment and treatment response. Targeting the BM may represent an alternative therapeutic approach for ccRCC.
