    Abstract Prehistoric human cultural sites along the eastern coast of Fujian Province are important records for studying the origin and spread of the Austronesian civilization. The Huangqiyu site is the first Tanshishan culture site to be unearthed in the Min River estuary area and was excavated by scientific archeology. It has great significance for further research of the Tanshishan culture and the coupled relationship between prehistoric human activities and environmental changes in the Fujian coastal region. The chronology of archeological sites is an important foundation for prehistoric archeology. In this study, the burial age of the Huangqiyu site in Lianjiang County was analyzed using the Optically Stimulated Luminescence (OSL) dating technique on the basis of Accelerator Mass Spectrometry (AMS) 14C dating. In the field, the T2 section of the Huangqiyu site was selected for sampling, and the single-aliquot regeneration dose method of medium-grained quartz was used to assess OSL chronology in the laboratory. The results showed that the OSL burial age of the Huangqiyu site ranges from approximately 8.5 ± 0.5 to 4.4 ± 0.2 ka, corresponding to the sedimentary/cultural strata of the early Middle Holocene. The OSL age of sample 2019118, which was consistent with the AMS 14C age, was 4.4 ± 0.2 ka, corresponding roughly to the Tanshishan culture. The age of sample 2019117 was 5.4 ± 0.3 ka, which may be an overestimation because the equivalent dose of this sample showed a weakly skewed distribution with a tail at the high value end, indicating that there was a small amount of sundry particles and that the drying was insufficient. This problem may have led to a bias in the measured equivalent dose and, in turn, to overestimation of the age of the sample. The age of sample 2019119, which was affected by the sampling environment, was 8.5 ± 0.5 ka, possibly indicating the maximum age of the bottom boundary of the site. Archeological studies have found that the age indicated by the pottery features excavated from the cultural layer at the bottom of the Huangqiyu site is most likely earlier than the Tanshishan culture, or possibly that of the lower strata of the Tanshishan culture. Overall, the ages of samples 2019117 and 2019119 are different from the AMS 14C ages, but these are consistent with the archeological characteristics of the site. By comparing the geomorphological and environmental evolution process of the Fuzhou Basin, where the Huangqiyu site is located, with the regional prehistoric human activities, it was found that the site corresponds to the warm climate of 6 500-4 000 a B P and relatively a high sea level period. Many prehistoric remains were developed in the Fuzhou Basin during this period, and the prosperous Tanshishan culture and the lower Tanshishan culture emerged in the region. At the end of the Neolithic age, the abrupt cold event of 4.2-4.0 ka, the drastic change of climate and environment in the tropical and subtropical regions on both sides of the Pacific Ocean at 4 000-3 000 a B P, and the invasion of the northern Wu-Yue culture may explain the disappearance of the follow-up culture. Because of the limited precision of the regional sea-level change curve, it is insufficient to discuss sea-land change and the coupled relationship with human activities. Relevant studies need to strengthen these aspects in the future.

    Abstract Background: Epidemiologic evidence has demonstrated a correlation between ankylosing spondylitis and psychiatric disorders. However, little is known about the common genetics and causality of this association. This study aimed to investigate the common genetics and causality between ankylosing spondylitis (AS) and psychiatric disorders.
    Methods: A two-sample Mendelian Randomization (MR) analysis was carried out to confirm causal relationships between ankylosing spondylitis and five mental health conditions including major depressive disorder (MDD), anxiety disorder (AXD), schizophrenia (SCZ), bipolar disorder (BIP), and anorexia nervosa (AN). Genetic instrumental variables associated with exposures and outcomes were derived from the largest available summary statistics of genome-wide association studies (GWAS). Bidirectional causal estimation of MR was primarily obtained using the inverse variance weighting (IVW) method. Other MR methods include MR-Egger regression, Weighted Median Estimator (WME), Weighted Mode, Simple Mode, and Mendelian randomization pleiotropy residual sum and outlier (MR-PRESSO). Sensitivity analyses are conducted to estimate the robustness of MR results.
    Results: The findings suggest that AS may be causally responsible for the risk of developing SCZ (OR = 1.18, 95% confidence interval = (1.06, 1.31), P = 2.58 × 10
    Conclusion: Our study provides fresh insights into the relationship between AS and psychiatric disorders (SCZ and AN). Furthermore, it may provide new clues for risk management and preventive interventions for mental disorders in patients with AS.

    Abstract Background: A nomogram is a valuable and easily accessible tool for individualizing cancer prognosis. This study aims to establish and validate two prognostic nomograms for long-term overall survival (OS) and cancer-specific survival (CSS) in non-metastatic nasopharyngeal carcinoma (NPC) patients and to investigate the treatment options for the nomogram-based risk stratification subgroups.
    Methods: A total of 3959 patients with non-metastatic NPC between 2004 and 2015 were identified from the Surveillance, Epidemiology, and End Results (SEER) database. The patients were randomly allocated to the training and validation cohorts in a 7:3 ratio. Prognostic nomograms were constructed to estimate OS and CSS by integrating significant variables from multivariate Cox regression employing a backward stepwise method. We examined the correlation indices (C-index) and areas under the curves (AUC) of time-dependent receiver operating characteristic curves to assess the discriminative ability of our survival models. The comprehensive enhancements of predictive performance were evaluated with net reclassification operating improvement (NRI) and integrated discrimination improvement (IDI). Reliability was validated using calibration plots. Decision curve analysis (DCA) was used to estimate clinical efficacy and capability. Finally, the nomogram-based risk stratification system used Kaplan-Meier survival analysis and log-rank tests to examine differences between subgroups.
    Results: The following independent parameters were significant predictors for OS: sex, age, race, marital status, histological type, median household income, AJCC stage tumor size, and lymph node size. Except for the race variables mentioned above, the rest were independent prognostic factors for CSS. The C-index, AUC, NRI, and IDI indicated satisfactory discriminating properties. The calibration curves exhibited high concordance with the exact outcomes. Moreover, the DCA demonstrated performed well for net benefits. The prognosis significantly differed between low- and high-risk patients (p < 0.001). In a treatment-based stratified survival analysis in risk-stratified subgroups, chemotherapy benefited patients in the high-risk group compared to radiotherapy alone. Radiotherapy only was recommended in the low-risk group.
    Conclusions: Our nomograms have satisfactory performance and have been validated. It can assist clinicians in prognosis assessment and individualized treatment of non-metastatic NPC patients.

    Abstract Nucleic acids are considered as promising materials for developing exquisite nanostructures from one to three dimensions. The advances of DNA nanotechnology facilitate ingenious design of DNA nanostructures with diverse shapes and sizes. Especially, the algebraic topological framework nucleic acids (ATFNAs) are functional DNA nanostructures that engineer guest molecules (e. g., nucleic acids, proteins, small molecules, and nanoparticles) stoichiometrically and spatially. The intrinsic precise properties and tailorable functionalities of ATFNAs hold great promise for biological applications, such as cell recognition and immunotherapy. This Perspective highlights the concept and development of precisely assembled ATFNAs, and outlines the new frontiers and opportunities for exploiting the structural advantages of ATFNAs for biological applications.

    Abstract Aims: The study aims to provide comprehensive evidence for the selection of agents in type 2 diabetes mellitus (T2DM) patients with cardiovascular risk and summarize the lasted evidence for the cardiovascular effects of sodium glucose cotransporter-2 inhibitor (SGLT2i) in patients with heart failure (HF).
    Methods and results: Several online databases were searched. All studies that explored the cardiovascular effects of SGLT2i or glucagon-like peptide 1 receptor agonist (GLP1-RA) were screened and reviewed. A total of 38 studies were included. Compared with GLP1-RA, the use of SGLT2i significantly reduced the risk of cardiovascular death [risk ratio (RR) = 0.59; 95% confidence interval (CI), 0.44-0.58], hospitalization of heart failure (HHF) (RR = 0.77; 95% CI, 0.74-0.80), death from any cause (RR = 0.64; 95% CI, 0.60-0.68), and myocardial infarction (MI) (RR = 0.81; 95% CI, 0.76-0.87). However, SGLT2i significantly increased the risk of stroke (RR = 1.10; 95% CI, 1.04-1.17). Compared with the control group, SGLT2i treatment reduced the risk of cardiovascular death by 14% (RR = 0.86; 95% CI, 0.79-0.94), HHF by 25%, and death from any cause by 9% in patients with HF, regardless of diabetes status.
    Conclusions: SGLT2i is associated with a lower risk of cardiovascular death, HHF, death from any cause, and MI in patients with T2DM compared with GLP1-RA. In addition, SGLT2i brought more benefits with respect to the effects of cardiovascular death, HHF, and death from any cause in patients with HF, regardless of diabetes status.

    Abstract There are nearly 1.15 million new cases of colon cancer, as well as 586,858 deaths from colon cancer worldwide in 2020. The aim of this study is to reveal whether ZMIZ1 can control the fate of colon cancer cells and the mechanism by which it functions. Specific shRNA transfection was used to knock down the expression of ZMIZ1 in colon cancer cell lines (HCT116 and HT29), and cell proliferation was detected using EdU and CCK-8 reagents, apoptosis by flow cytometry, and autophagy by western blot. The interaction of ZMIZ1 and SIRT1 was analyzed. Knockdown of ZMIZ1 significantly inhibited autophagy and proliferation, and induced apoptosis of HCT116 and HT29 cells. The mRNA level of SIRT1 was not affected by ZMIZ1 knockdown, but the protein level of SIRT1 was significantly decreased and the protein level of the SIRT1-specific substrate, acetylated FOXO3a, was reduced. Immunoprecipitation assays identified the interaction between SIRT1 and ZMIZ1 in HCT116 and HT29 cells. ZMIZ1 increased intracellular ubiquitination of SIRT1. Knockdown or pharmacological inhibition of SIRT1 neutralized the effects of ZMIZ knockdown on proliferation, autophagy and apoptosis in HCT116 and HT29 cells. ZMIZ1 may control the fate of colon cancer cells through the SIRT1/FOXO3a axis. Targeting ZMIZ1 would be beneficial for the treatment of colon cancer.
