    Abstract Objectives: Vacuolar H
    Materials and methods: Transient forebrain ischemia was induced by occlusion of both common carotid arteries for 5 min and animals were sacrificed at various time points after ischemia for immunohistochemical staining of ATP6V1B2 and measurements of pH and lactate levels in the hippocampus.
    Results: ATP6V1B2 immunoreactivity was found to be transiently increased in the hippocampal CA1 region and dentate gyrus 12-24 hr after ischemia when the pH and lactate levels were decreased. In addition, ATP6V1B2 immunoreactivity significantly increased in the hippocampal CA3 and dentate gyrus, regions relatively resistant to ischemic damage, 4 days after ischemia, when the NeuN-positive, mature neuron numbers were significantly decreased in the hippocampal CA1 region.
    Conclusion: These results suggest that ATP6V1B2 expression is transiently increased in the hippocampus following ischemia, which may be intended to compensate for ischemia-related dysfunction of ATP6V1B2 in the hippocampus.

    Abstract Selenium has been reported to induce the expression of some cytoprotective enzymes, which may account for its chemoprotective and chemopreventive effects. However, it remains largely unresolved whether these effects are exerted by selenium itself or mediated by its metabolite(s). In the present study, methylseleninic acid (MSeA), a monomethylated selenium, induced the expression of NAD(P)H:quinone oxidoreductase-1 (NQO-1) in human Chang liver cells. Expression of NQO-1 and other antioxidant/stress response genes is primarily regulated by the transcription factor NF-E2-related factor2 (Nrf2). Exposure of human Chang liver cells to MSeA (3 μM) increased nuclear translocation of Nrf2 and binding to antioxidant response elements. Silencing Nrf2 markedly reduced the MSeA-induced NQO-1 expression. In comparison with embryonic fibroblasts from Nrf2 wild-type mice, those from Nrf2 knockout mice failed to induce NQO-1 expression when treated with MSeA. Moreover, MSeA treatment enhanced ubiquitination of Keap1, but repressed Nrf2 ubiquitination. Pretreatment of cells with dithiothreitol abrogated the MSeA-induced NQO-1 expression, suggesting that MSeA causes Keap1 thiol modification. MSeA-induced NQO-1 upregulation was attenuated in cells harbouring the mutant Keap1 in which the cysteine 151 residue was replaced by serine. Oral administration of MSeA (1 mg/kg) by gavage to mice induced hepatic NQO-1 expression. Similar to MSeA, methylselenol generated from selenomethionine by methioninase activity induced NQO-1 expression. In conclusion, MSeA, the immediate precursor of methylselenol, upregulates the expression of NQO-1 via the Keap1-Nrf2 signaling. The above findings suggest that biological activities of selenium are dependent on the nature of the metabolites as well as the type of ingested selenium formulations.

    Abstract Testicular oxidative stress is one of the most common factors underlying male infertility. Welted thistle, Carduus crispus Linn., and its bioactive principles are attracting scientific interest in treating male reproductive dysfunctions. Here, the protective effects of apigenin isolated from C. crispus against oxidative damage induced by hydrogen peroxide (H 2 O 2 ) and dysregulation in spermatogenesis associated parameters in testicular sperm cells was investigated. Cell viabilities, ROS scavenging effects, and spermatogenic associated molecular expressions were measured by MTT, DCF-DA, Western blotting and real-time RT-PCR, respectively. A single peak with 100% purity of apigenin was obtained in HPLC conditions. Apigenin treated alone (2.5, 5, 10 and 20 µM) did not exhibit cytotoxicity, but inhibited the H 2 O 2 -induced cellular damage and elevated ROS levels significantly ( p < 0.05 at 5, 10 and 20 µM) and dose-dependently. Further, H 2 O 2 -induced down-regulation of antioxidant (glutathione S -transferases m5, glutathione peroxidase 4, and peroxiredoxin 3) and spermatogenesis-associated (nectin-2 and phosphorylated-cAMP response element-binding protein) molecular expression in GC-2spd cells were attenuated by apigenin at both protein and mRNA levels ( p < 0.05). In conclusion, our study showed that apigenin isolated from C. crispus might be an effective agent that can protect ROS-induced testicular dysfunctions.

    Abstract This study aimed to investigate the effect of H 2 O 2 addition, ozone feed rate, and UV addition on the change in the concentration of organics such as COD Mn , COD Cr , TOC, and PFCs in tertiary-treated effluent from a sewage treatment plant (STP) during the O 3 and UV/H 2 O 2 process. The degradation of organic pollutants from tertiary effluent is a significant challenge because biological treatment cannot degrade these recalcitrant pollutants. Therefore, the O 3 /UV/H 2 O 2 process was an effective method for treating recalcitrant organics. Several batch tests were conducted to investigate the direct UV photolysis, UV/H 2 O 2 , and ozone-based advanced oxidation process to degrade COD Mn , COD Cr , TOC, and PFCs. The chemical oxygen demand (COD) and total organic carbon (TOC) with UV irradiation showed 95% and 50% removal efficiency percentages under optimal conditions (initial pH = 6.7, H 2 O 2 dosage = 50 mg/L, ozone feed rate = 5.8 mg/L/min. Moreover, UV irradiation, with the addition of H 2 O 2 , and a sufficient dose of ozone, demonstrated the efficient removal of organic compounds by the indication of radical oxidation. (·OH) is the dominant mechanism. However, AOPs are not sufficient to fully treat the PFC compound; thus, additional procedures are required to degrade PFCs. In this study, the removal of organic recalcitrant contaminants and the change in added PFC concentration in tertiary-treated sewage were investigated by applying the ozone-based advanced oxidation process.

    Abstract Objective(s): Vacuolar H+-ATPase is a highly conserved enzyme that plays an important role in maintaining an acidic environment for lysosomal function and accumulating neurotransmitters in synaptic vesicles. In the present study, we investigated the time-dependent changes in the expression of vacuolar H+-ATPase V1B2 (ATP6V1B2), a major neuronal subtype of vacuolar H+-ATPase located in the hippocampus, after 5 min of transient forebrain ischemia in gerbils.     Abstract Abstract The whitebacked planthopper (WBPH), has become a devastating pest for rice crops, causes serious yield losses each year, and urgently needs biological control. Here, we developed a WBPH-resistant rice cultivar by overexpressing the OsF3H gene. A genetic functional analysis of the OsF3H gene confirmed its role in facilitating flavonoid contents and have indicated that the expression of the OsF3H gene is involved in regulation of the downstream genes (OsDFR and OsFLS) of the flavonoid pathway and genes (OsSLR1 and OsWRKY13) involved in other physiological pathways. OxF3H (OsF3H transgenic) plants accumulated significant amounts of the flavonols kaempferol (Kr) and quercetin (Qu) and the anthocyanins delphinidin and cyanidin, compared to the wild type, in response to the stress induced by WBPH. Similarly, OsF3H-related proteins were significantly expressed in OxF3H lines after WBPH infestation. The present study, indicated that the regulation of JA in OxF3H plants was suppressed due the overexpression of the OsF3H gene, which induced the expression of downstream genes related to anthocyanin. Similarly, the OsWRKY13 transcriptional factor was significantly suppressed in OxF3H plants during WBPH infestation. Exogenous application of Kr and Qu increased the survival rates of susceptible TN1 lines in response to WBPH, while decreased the survival rate of first instar WBPHs, indicating that both flavonols exhibit pesticide activity. Phenotypic demonstration also affirms that OxF3H plants show strong resistance to WBPH compared with wild type. Collectively, our result suggested that OsF3H overexpression led to the up-regulation of defense related genes and enhanced rice resistance to WBPH infestation.
