    Abstract Sudden cardiac arrest (SCA) consisting of ventricular fibrillation and ventricular tachycardia considered as shockable rhythms is a life-threatening heart disease, which is treated efficiently by the automated external defibrillator (AED). This work proposes a novel design of the SAA, which includes a k-nearest neighbors model and a subset of 8 features extracted from the ECG segments, for the SCA diagnosis on the electrocardiogram (ECG) signal. These features are addressed as the most productive subset among 31 input features based on the evaluation of the feature correlation. The recursive feature elimination algorithm combined with the Boosting model and wise-patient fivefold cross-validation method is adopted for the calculation of the average feature importance, which shows the degree of feature correlation, to construct various input feature subsets. Moreover, component feature combinations known as the representatives of the input feature subsets with an enormous level of correlation and independence are transformed from the input subsets by the principal component analysis method. The wise-patient fivefold cross-validation procedure is used for the evaluation of these component feature combinations on the validation set. The proposed SAA is certainly efficient for SCA detection with a small number of the extracted feature and relatively high diagnosis performance such as accuracy of 99.52%, sensitivity of 97.69%, and specificity of 99.91%.

    Abstract Approaches for profiling protease substrates are critical for defining protease functions, but remain challenging tasks. We combine genetic code expansion, photocrosslinking and proteomics to identify substrates of the mitochondrial (mt) human caseinolytic protease P (hClpP). Site-specific incorporation of the diazirine-bearing amino acid DiazK into the inner proteolytic chamber of hClpP, followed by UV-irradiation of cells, allows to covalently trap substrate proteins of hClpP and to substantiate hClpP's major involvement in maintaining overall mt homeostasis. In addition to confirming many of the previously annotated hClpP substrates, our approach adds a diverse set of new proteins to the hClpP interactome. Importantly, our workflow allows identifying substrate dynamics upon application of external cues in an unbiased manner. Identification of unique hClpP-substrate proteins upon induction of mt oxidative stress, suggests that hClpP counteracts oxidative stress by processing of proteins that are involved in respiratory chain complex synthesis and maturation as well as in catabolic pathways.

    Abstract Objectives: The aim of this study is to provide an updated overview of the oral health status and associated risk factors in maritime settings.
    Methods: We systematically searched PubMed, Ovid Embase, Web of Science, CINAHL and SCOPUS from January 2010 to April 2023. Two independent reviewers extracted the data. The quality of included studies was assessed using relevant assessment tools.
    Results: A total of 260 records were found in the initial search; 24 articles met the inclusion criteria. Most studies had descriptive design, and only two randomized controlled trials were found. The main oral health issues noted are oral cancer, dental caries, periodontal diseases, oral mucosal lesions, and dental emergency. Male seafarers have higher risk of oral cancers in the tongue, lips, and oral cavity while oral mucosal lesions are more prevalent among fishermen.
    Conclusions: Dental caries and periodontal diseases are prevalent in both seafarers and fishermen. The consumption of tobacco, alcohol, fermentable carbohydrate, and poor oral hygiene are risk factors that affect the oral health status at sea. The occurrence of oral diseases in maritime setting requires more attention of researchers and authorities to develop strategies to tackle these issues.

    Abstract Summary: This review describes recent research that has advanced our understanding of the role of immune cells in the tumor microenvironment (TME) using advanced 3D in vitro models and engineering approaches. The TME can hinder effective eradication of tumor cells by the immune system, but immunotherapy has been able to reverse this effect in some cases. However, patient-to-patient variability in response suggests that we require deeper understanding of the mechanistic interactions between immune and tumor cells to improve response and develop novel therapeutics. Reconstruction of the TME using engineered 3D models allows high-resolution observation of cell interactions while allowing control of conditions such as hypoxia, matrix stiffness, and flow. Moreover, patient-derived organotypic models are an emerging tool for prediction of drug efficacy. This review highlights the importance of modeling and understanding the immune TME and describes new tools for identifying new biological targets, drug testing, and strategies for personalized medicine.
