    Abstract Motivation: The Polygenic Score (PGS) Catalog is a recently established open database of published polygenic scores that, to date, has collected, curated and made available 721 polygenic scores from over 133 publications. The PGS Catalog REST API is the only method allowing programmatic access to this resource.
    Results: Here, we describe quincunx, an R package that provides the first client interface to the PGS Catalog REST API. quincunx enables users to query and quickly retrieve, filter and integrate metadata associated with polygenic scores, as well as polygenic scoring files in tidy table format.
    Availability and implementation: quincunx is freely available under an MIT License, and can be accessed from https://github.com/maialab/quincunx.

    Abstract Motivation: The National Human Genome Research Institute Catalog of Published Genome-Wide Association Studies (GWAS) Catalog has collected, curated and made available data from over 7100 studies. The recently developed GWAS Catalog representational state transfer (REST) application programming interface (API) is the only method allowing programmatic access to this resource.
    Results: Here, we describe gwasrapidd, an R package that provides the first client interface to the GWAS Catalog REST API, representing an important software counterpart to the server-side component. gwasrapidd enables users to quickly retrieve, filter and integrate data with comprehensive bioinformatics analysis tools, which is particularly critical for those looking into functional characterization of risk loci.

    Abstract TiO₂, ZnO, and SnO₂ metal oxides were synthesized by the sol-gel method and heterojunctions were fabricated by combining TiO₂ with either ZnO or SnO₂ in a 1:1 ratio using mechanochemical ball milling process. The ball milling process promotes phase transition of TiO₂ from anatase to rutile and yields ternary heterojunction of the type TiO₂(A)/TiO₂(R)/ZnO and TiO₂(A)/TiO₂(R)/SnO₂ (A-anatase and R-rutile). These ternary heterojunctions were characterized by various analytical techniques and its photocatalytic efficiency is evaluated using 4-Chloro Phenol as a model compound under UV and solar light. The enhanced catalytic activity of TiO₂(A)/TiO₂(R)/ZnO heterojunction is attributed to the formation of Ti³⁺-Vₒ defect states which leads to the efficient charge carrier separation. During the ball milling process severe crystal deformation takes place in TiO₂ and ZnO lattices by creating crystal lattice distortion which leads to the formation of defects due to valency mismatch between Ti⁴⁺ and Zn²⁺. A mechanistic pathway is proposed for the enhanced photocatalytic activity of the ternary heterojunctions.

    Abstract The plant species Acmella oleracea L. is used in the north of Brazil for the treatment of a range of illnesses, such as tuberculosis, flu, cough, and rheumatism and as an anti-inflammatory agent; besides, hydroethanolic formulations with this species are popularly used as a female aphrodisiac agent. However, currently, there are no studies performed evaluating its effect on embryonic development. Hence, this research aimed to evaluate the effects of the hydroethanolic extract of A. oleracea (EHFAo) on the reproductive performance (parental) and embryonic development (F1 generation) of zebrafish, at concentrations of 50, 100, and 200 μg/L. Histopathology of parental gonads after 21 days of exposure to EHFAo reveals few alterations in the ovaries and testes, not impairing the reproduction; an increase of eggs deposition was observed in animals treated with EHFAo at the highest concentrations. Nevertheless, concerning the embryonic development of F1, teratogenic effects were observed including tail deformation, cardiac and yolk edema, scoliosis, and growth retardation; these alterations were more prominent in the groups born from progenitors exposed to the highest concentrations (100 and 200 μg/L.); but only the occurrence of yolk and cardiac edema had a statistically significant difference when compared to the control group. The chromatographic analysis shows that spilanthol (affinin) was the primary compound found in the EHFAo. Hence, in silico assessment was performed to evaluate the pharmacokinetic and toxicological properties of this molecule and 37 metabolites derived from it. Overall, our data show that the treatment caused no detrimental changes in progenitors regarding their gonads or fertility but caused some potentially teratogenic activity in embryos, which may be due to the action of spilanthol’s metabolites M3, M6, M7, M8, M16, M28, and M31.

    Abstract The rpoB gene encoding for beta subunit of RNA polymerase is a target of mutations leading to rifampicin resistant (Rif(r)) phenotype of bacteria. Here we have characterized rpoB/Rif(r) system in Pseudomonas aeruginosa and Pseudomonas putida as a test system for studying mutational processes. We found that in addition to the appearance of large colonies which were clearly visible on Rif selective plates already after 24h of plating, small colonies grew up on these plates for 48 h. The time-dependent appearance of the mutant colonies onto selective plates was caused by different levels of Rif resistance of the mutants. The Rif(r) clusters of the rpoB gene were sequenced and analyzed for 360 mutants of P. aeruginosa and for 167 mutants of P. putida. The spectrum of Rif(r) mutations characterized for P. aeruginosa grown at 37 degrees C and that characterized for P. putida grown at 30 degrees C were dissimilar but the differences almost disappeared when the mutants of both strain were isolated at the same temperature, at 30 degrees C. The strong Rif(r) phenotype of P. aeruginosa and P. putida was accompanied only with substitutions of these residues which belong to the putative Rif-binding pocket. Approximately 70% of P. aeruginosa mutants, which were isolated at 37 degrees C and expressed weak Rif(r) phenotype, contained base substitutions in the N-terminal cluster of the rpoB gene. The differences in the spectra of mutations at 30 degrees C and 37 degrees C can be explained by temperature-sensitive growth of several mutants in the presence of rifampicin. Thus, our results imply that both the temperature for the growth of bacteria and the time for isolation of Rif(r) mutants from selective plates are critical when the rpoB/Rif(r) test system is employed for comparative studies of mutagenic processes in Pseudomonas species which are conventionally cultivated at different temperatures.

    Abstract Objective: to describe a solution enabling geolocation of hospital admissions (AIH), processed on the Brazilian National Health System's Hospital Information System.
    Methods: in order to spatialize AIHs an R language script was written, based on the microdatasus and CepR packages; the script was applied to identify all AIHs in Goiás state in the year 2015; after downloading and pre-processing the data, the procedure for AIH spatialization was detailed.
    Results: of the 361,213 AIHs processed, we were able to retrieve 24,220 different ZIP codes (CEPs); from this set of ZIP codes, 23,910 (98.7%) were geolocated; these geolocated ZIP codes enabled spatialization of 97.7% of AIHs processed for the state of Goiás.
    Conclusion: it is possible to spatialize AIHs with a high success rate; the method detailed in this paper opens a new range of possibilities for the design of evaluation studies, formulation of policies and planning of health care actions.

    Abstract We developed a computer program for life table analysis using the open source, free software programming environment R. It is useful to quantify chronic nonlethal effects of treatments on arthropod populations by summarizing information on their survival and fertility in key population parameters referred to as fertility life table parameters. Statistical inference on fertility life table parameters is not trivial because it requires the use of computationally intensive methods for variance estimation. Our codes present some advantages with respect to a previous program developed in Statistical Analysis System. Additional multiple comparison tests were incorporated for the analysis of qualitative factors; a module for regression analysis was implemented, thus, allowing analysis of quantitative factors such as temperature or agrochemical doses; availability is granted for users, once it was developed using an open source, free software programming environment. To illustrate the descriptive and inferential analysis implemented in lifetable.R, we present and discuss two examples: 1) a study quantifying the influence of the proteinase inhibitor berenil on the eucalyptus defoliator Thyrinteina arnobia (Stoll) and 2) a study investigating the influence of temperature on demographic parameters of a predaceous ladybird, Hippodamia variegata (Goeze).

    Abstract With the evolution of bacterial resistance over conventional antimicrobials and the development of new antimicrobials, the search for new compounds of natural origin has intensified, since plants with therapeutic potential constitute a source of new biologically active compounds, encouraging the development of new therapeutic options. The objective of this study was to evaluate the antibacterial and anti-adherent activity of the ethanolic extract of Praxelis clematidea (Griseb.) R.M. King & Robinson (Asteraceae) on strains of Staphylococcus aureus, Streptococcus pneumoniae, Klebsiella pneumoniae, Pseudomonas aeruginosa and Enterococcus faecalis. Tests were carried out to determine the MIC and MBC by means of broth microdilution in 96-well plates, and to determine the MICA, a technique with inclined tubes in the presence of 5% sucrose was used. It was observed that the ethanolic extract of P. clematidea has strong bactericidal activity on K. pneumoniae, and moderate bactericidal activity on S. pneumoniae and E. faecalis. Being able to inhibit adherence to K. pneumoniae strains. Thus, the ethanolic extract of P. clematidea proves to be effective as an antimicrobial in the control and prevention of infections by S. pneumoniae, K. pneumoniae and E. faecalis. It also has an effective anti-adherent effect on K. pneumoniae strains.
