    Abstract Purpose: Breast cancer is one of the leading causes of tumor death worldwide in female, and the five-year overall survival of breast cancer patients remains poor. It is an urgent need to seek novel target for its treatment. Synaptotagmin 13 (SYT13) is a synaptic vesicle transporting protein that regulates the malignant phenotypes of various cancers. However, its role in breast cancer is still unclear. The current study aimed to investigate the effects of SYT13 on the progression of breast cancer.
    Methods: Twenty-five pairs of breast cancer tissues and non-tumor tissues were obtained to assess the expression of SYT13. We manually modified the expression of SYT13 in MCF-7 and MDA-MB-231 cells. CCK-8 assay, EdU staining, and cell cycle analysis were carried out to measure the proliferated ability of cells. Annexin V/PI and TUNEL assays were used to detect the apoptotic ability of cells. Wound healing and transwell assays were employed to evaluate the migrated and invasive ability of breast cancer cells.
    Results: The results revealed that the mRNA and protein levels of SYT13 were higher in breast cancer tissues and cell lines. Knockdown of SYT13 inhibited the cell proliferation and induced cell cycle arrest in G1 phase of MCF-7 cells by downregulating cyclin D1 and CDK4, as well as upregulating p21. The migration and invasion of MCF-7 cells were repressed by the loss of SYT13 via the gain of E-cadherin and the loss of vimentin. Overexpression of SYT13 in MDA-MB-231 cells led to the opposite effects. Silencing of SYT13 induced the apoptosis ability of MCF-7 cells by the upregulation of bax and the downregulation of bcl-2. Moreover, we found that SYT13 depletion suppressed the FAK/AKT signaling pathway. PF573228 (a FAK inhibitor) and MK2206 (an AKT inhibitor) reversed the SYT13 overexpression-induced promotion of proliferation, migration, and invasion of MDA-MB-231 cells.
    Conclusion: The results indicated that SYT13 promoted the malignant phenotypes of breast cancer cells by the activation of FAK/AKT signaling pathway.

    Abstract Climatic comfort, which refers to the comfort of the human body's thermal sensations, is important for the human habitat. Although considerable efforts have been provided to examine changes in climatic comfort response to global warming from a partial perspective, the trajectory shift in past and future climatic comfort conditions in China's mainland based on uniform indicators has not been revealed. The spatiotemporal pattern of climatic comfort over historical and future periods was investigated in this study, using China's mainland as an example. The temperature‒humidity index was adopted on the basis of homogenised meteorological station observations and high-resolution climate model simulations to analyse the trends of comfort/discomfort days from 1960 to 2017 and project changes in climatic comfort under representative concentration pathway scenarios in the late 21st century (2071–2100). Results show a substantial decrease in cold-uncomfortable days and a moderate increase in comfortable and warm-uncomfortable days from 1960 to 2017. In the late 21st century, the signals of increasing warm-uncomfortable and decreasing cold-uncomfortable days are projected to enhance significantly while the direction of changes in comfortable days exhibits a north‒south divergence. The uneven changes in warm- and cold-uncomfortable days and an overall decrease in comfortable days in the late 21st century dominate the future trends in climatic comfort in the densely populated southeast half of China. Effective measures taken for adapting to and mitigating global climate warming can considerably avoid the adverse impact of the projected change.

    Abstract The monophasic variant of Salmonella enterica serovar Typhimurium with the antigenic formula 1,4,[5],12:i:- is one of the most common pathogenic bacteria causing global food-borne outbreaks. However, the research on molecular characteristics and evolution of monophasic S. typhimurium in China is still lacking. In the current study, 59 monophasic S. typhimurium strains were isolated from food animals and food products in South China between 2011 and 2018. A total of 87.5 % of monophasic S. typhimurium isolates were grouped into one independent clade with other monophasic S. typhimurium strains in China distinct from other countries by phylogenomic analysis. These isolates possess variable genotypes, including multiple ARGs on plasmid IncHI2, diverse evolutions at the fljAB locus, and virulence factors. Our results suggest that the monophasic S. typhimurium isolates currently circulating in China might be an independent epidemic subtype.
