Abstract Scholars have used the experience economy to analyze the behavior of tourists. However, in the field of intangible cultural heritage (ICH) tourism, the relationship between the experience economy and the behavior intention of tourists has not been studied. Scholars also point out that the relationship between the four dimensions of the experience economy is not static, and that aesthetic experience may be predictive of other dimensions. This study uses aesthetic experience as the starting point and constructs a theoretical model that includes the experience economy, the memories of ICH tourists, the perception of authenticity, and behavioral intentions. Qiong Opera, part of China's national intangible cultural heritage, is used as a scenario in which to conduct empirical research. The results show that education, entertainment, and escape play a mediator role in the relationship between aesthetics and memory; memory plays a complete mediator role in the relationship between education, entertainment, escape, and behavioral intention; and authenticity plays a moderator role in the relationship between education, entertainment, escape, and memory. This study introduces the experience economy into ICH tourism. While expanding the application field of experience economy theory, it also provides theoretical and management inspiration for ICH tourism development.

Abstract Our previous study found that Qiong-Yu-Gao (QYG), a traditional Chinese medicine formula derived from Rehmanniae Radix, Poria, and Ginseng Radix, has protective effects against cisplatin-induced acute kidney injury (AKI), but the underlying mechanisms remain unknown. In the present study, the potential role of gut microbiota in the nephroprotective effects of QYG was investigated. We found that QYG treatment significantly attenuated cisplatin-induced AKI and gut dysbiosis, altered the levels of bacterial metabolites, with short-chain fatty acids (SCFAs) such as acetic acid and butyric acid increasing and uremic toxins such as indoxyl sulfate and

Abstract Salt stress, causing serious loss on crop productions, is one of the most important environmental stresses throughout the world. The aim of this study is to select salt-tolerant traditional rice resources collected from Lei-Qiong area of South China and investigate their physiological performances and biochemical regulations during salt stress response, together with two well-known international varieties, Nona Bokra (salt-tolerant sample) and IR29 (salt-sensitive sample). After comprehensive analyses, we discovered that two Lei-Qiong traditional salt-tolerant rice samples showed less growth inhibition by salt stress during both germination and seedling stage, in comparison with other rice samples. Moreover, there were less chlorosis symptoms in these two kinds of salt tolerant rice under salt stress, corresponding to their better water-holding capacity. We measured malondialdehyde and proline contents, and activities of CAT and POD of seedlings treated with 100 mM NaCl for 5 dand 10 d, respectively. Interestingly, less cellular membrane damage and stronger antioxidant enzyme system were found in the two Lei-Qiong rice samples. Our study suggests that traditional rice landrace growing onshore of Lei-Qiong area in China possesses good salt-tolerant capacity, which could be attributed to their efficient antioxidant enzyme system.

Abstract Cisplatin is a highly effective anti-cancer chemotherapeutic agent; however, its clinical use is severely limited by serious side effects, of which nephrotoxicity is the most important. In this study, we investigated whether Qiong-Yu-Gao (QYG), a popular traditional Chinese medicinal formula described 840 years ago, exhibits protective effects against cisplatin-induced renal toxicity. Using a mouse model of cisplatin-induced renal dysfunction, we observed that pretreatment with QYG attenuated cisplatin-induced elevations in blood urea nitrogen and creatinine levels, ameliorated renal tubular lesions, reduced apoptosis, and accelerated tubular cell regeneration. Cisplatin-mediated elevations in tumor necrosis factor alpha (TNF-α) mRNA, interleukin-1 beta (IL-1β) mRNA, and cyclooxygenase-2 (COX-2) protein in the kidney were also significantly suppressed by QYG treatment. Furthermore, QYG reduced platinum accumulation in the kidney by decreasing the expression of copper transporter 1 and organic cation transporter 2. An in vivo study using implanted Lewis lung cancer cells revealed that concurrent administration of QYG and cisplatin did not alter the anti-tumor activity of cisplatin. Our findings suggest that the traditional Chinese medicinal formula QYG inhibits cisplatin toxicity by several mechanisms that act simultaneously, without compromising its therapeutic efficacy. Therefore, QYG may be useful in the clinic as a protective agent to prevent cisplatin-induced nephrotoxicity.

Abstract Objectives: More and more evidences show that circular RNAs (circRNAs) can be used as miRNA sponge to regulate the drug resistance of malignancies, including melanoma. However, how exosomal circRNAs participate in the therapeutic resistance of melanoma remains ambiguous.
Methods: Vemurafenib-resistant A375 cells were cultured and then the circRNA profile of exosomes from the parental A375 and A375-resistant cells were sequenced. Transmission electron microscopy (TEM), exogenous nanoparticle tracking analysis (NTA) and Western Blot assays were leveraged to confirm the successful collection of exosomes from A375 and A375R cells. Another five published RNA-seq data and microRNA-seq data, and seven miRNA databases were collected to construct a competing endogenous RNA (ceRNA) network. Comprehensive bioinformatic analysis was adopted to identify key molecules related to the drug resistance, including multiscale embedded gene co-expression network analysis (MEGENA). Then, qRT-PCR, cell viability and colony formation were used to estimate the function of hub circRNAs. The role of has_circ_0001005 in vivo was verified via xenograft assay. The Tumor online Prognostic analyses Platform (ToPP) was leveraged to develop the has_circ_0001005-related prognostic models for melanoma patients based on TCGA data.
Results: Compared with parental cells, hsa_circ_0001005 expression was significantly increased in resistant cells and their exosomes. The elevated level of hsa_circ_0001005 was related to the poor clinical prognosis of melanoma patients. Hsa_circ_0001005 found in melanoma was mainly secreted by drug-resistant cells as exosomes. Exosomal hsa_circ_0001005 activated multiple canonical pathways related to drug resistance through sponging four miRNAs, thus suppressing the drug sensitivity of melanoma. Knocking down hsa_circ_0001005 in vitro, we found that the inhibition of hsa_circ_0001005 could hinder the clone formation of melanoma. Further in vivo animal experiments suggested that suppression of hsa_circ_0001005 can increase the sensitivity to Vemurafenib of melanoma cells. Finally, we also constructed the functional regulatory ceRNA network and prognostic risk models for hsa_circ_0001005, and further survival analysis reveals that the regulatory network and prognostic risk models obviously affected the prognosis of melanoma patients.
Conclusions: This study confirmed that the level of hsa_circ_0001005 in exosomes is the key factor affecting drug resistance of melanoma, which provides a new potential therapeutic target for melanoma patients.
