    Abstract Aim: For the successful approval and clinical prescription of insulin biosimilars, it is essential to show pharmacokinetic (PK) and pharmacodynamic (PD) bioequivalence to the respective reference products sourced from the European Union and the United States.
    Methods: Three phase 1, randomized, double-blind, three-period crossover trials compared single doses of the proposed biosimilar insulin analogues aspart (GL-Asp, n = 36), lispro (GL-Lis, n = 38) and glargine (GL-Gla, n = 113), all manufactured by Gan & Lee pharmaceuticals, to the respective EU- and US-reference products in healthy male participants (GL-Asp and GL-Lis) or people with type 1 diabetes (GL-Gla). Study participants received 0.2 U/kg (aspart and lispro) or 0.5 U/kg (glargine) of each treatment under automated euglycaemic clamp conditions. The clamp duration was 12 h (aspart and lispro) or 30 h (glargine). Primary PK endpoints were the total area under the PK curves (AUC
    Results: Bioequivalence to both EU- and US-reference products were shown for all three GL insulins. Least squares mean ratios for the primary PK/PD endpoints were close to 100%, and both 90% and 95% confidence intervals were within 80%-125% in all three studies. There were no noticeable differences in the safety profiles between test and reference insulins, and no serious adverse events were reported for the GL insulins.
    Conclusion: GL-Asp, GL-Lis and GL-Gla are bioequivalent to their EU- and US-reference products.

    Abstract A novel and stable cytoplasmic male sterility CMS line of tuber mustard has been bred by subsequent backcrosses for 10 years. Two specific markers atpA and orf220 were cloned and partially characterized in our previous study (Zhang et al. 2003). In this study, two new molecular markers, orf256 and orf305/orf324, have been isolated and identified. The orf256 gene size was found to be 825 bp in CMS line and a 1,357 bp in its maintainer line. Sequence analysis indicated that the orf256 gene was an entire coding sequence and downstream of the cox1 gene. Interestingly, the 906 bp fragment, which contains part of the sequence of orf222, nad5 and orf139 genes, was found to be inserted from the 451st bp of 5'-flank of the 1,357 bp fragment. In the same way, the orf324 gene was isolated from CMS line and orf305 gene from its maintainer line. Both of them are entire coding sequences, upstream from nad3 and rps12 gene, and co-transcribed with the nad3 and rps12 genes. In addition, two molecular markers, orf256 and orf324/orf305, have been successfully converted into the SCAR markers. Subsequently, ORF256, ORF324, ORF305 protein and ORF256-M-431 fragment are predicated to contain signal peptide sequences, and ORF220 was predicated to contain signal anchor sequence. RFLP analysis results revealed that all of the molecular markers exhibited polymorphisms. Northern blot analysis indicated that the expression level of these genes in CMS line is higher than that of the maintainer line. In the mass, all of these genes are expressed lower in the leaf than that of floral organs between the CMS line and its maintainer line. The difference in expression pattern of different mitochondrial specific marker genes suggests that the abundance of mitochondrial proteins is differentially regulated in the organ/tissue development in tuber mustard. Results of this study also provide some novel and useful clues to explore the biological function of these specific marker genes in the tuber mustard.

    Abstract This study was conducted to investigate the effects of soil properties on the heavy metal accumulation in flowering Chinese cabbage (Brassica campestris L. ssp. chinensis var. utilis Tsen et Lee) at the field scale. The concentrations of cadmium (Cd), mercury (Hg), and chromium (Cr) in topsoil and vegetable samples from Nanhai district of Foshan city in the Pearl River Delta (PRD) were analyzed. The results showed that 56.5% of the soil samples exceeded the grade II of the Chinese Soil Environmental Quality Standard (GB 15618-1995) for Hg concentrations, while 8.70% and 17.4% of the vegetable samples exceeded the criteria of the Chinese Safety Qualification of Agricultural Products (GB 18406.1-2001) for Cd and Hg concentrations, respectively. The calculated bio-concentration factor (BCF; i.e., the ratio of the metal concentration in the edible parts of flowering Chinese cabbage to that in soil) values were ranked as: Cd (0.1415) > Cr (0.0061) > Hg (0.0012) (p < 0.01), which demonstrated that Cd was easier to be accumulated in the edible parts of flowering Chinese cabbage than Hg and Cr. Furthermore, the following relationships between (bio-concentration factor) BCF values (BCFs) and soil physicochemical properties were concluded from our results: i) the mean BCFs of coarse-textured soils were higher than those of fine-textured soils; ii) the BCFs decreased with increasing soil pH; iii) the soils with high organic matter(OM) and Cation exchange capacity (CEC) have low BCFs, resulting from their high sorption capacities for Cd, Hg, and Cr. The stepwise linear multiple regression analyses showed that total metal concentrations and available calcium in soils were two main factors controlling the accumulation of Cd, Hg, and Cr in the flowering Chinese cabbage.

    Abstract Expression profiling was performed using cDNA-AFLP technology on floral buds of a genic male sterile line (A line) and a maintainer line (B line) in Chinese cabbage pak-choi (Brassica campestris L. ssp. chinensis Makino var. communis Tsen et Lee cv. Aijiaohuang). A differentially expressed cDNA fragment, BA18-T16, was obtained via selective amplification with A18/T16 primer pair. BA18-T16 was specifically associated with the maintainer line, and was expressed only in floral buds revealed by half-quantitative RT-PCR. The 3' end and 5' end sequences of BA18-T16 were obtained by 3' and 5' RACE amplification and the full cDNA,designated as BcMF3, is 2082 bp long containing a 1755 bp open reading frame. BcMF3 shares high homology with Bp19, a pectin methylesterase from Brassica rapa L. with 99% identity at nucleotide level and 85% identity at amino acid level. Suggesting that BcMF3 most likely encodes a pectin methylesterase in Chinese cabbage pak-choi.
