    Abstract Objective: To study in vivo mercury absorption and accumulation through repeated transdermal administration of Yuhong ointment containing calomel, in order to provide scientific evidences for clinical safe medication.
    Method: A total of 100 SD rats were randomly classified into five groups: the control group, the Yuhong ointment group, the double-concentration Yuhong Ointment group, the quadruple-concentration Yuhong ointment group and the 1.6% calomel group. The rats were treated with the dosage of 0.04 g . cm-2 by repeated transdermal administration for 2, 4 weeks. After the drug discontinuance for 4 weeks, the levels of mercury in blood, urine, and tissues of heart, liver, brain and kidney were determined, respectively.
    Result: Compared with the control group, the blood mercury level of the Yuhong ointment group show no obvious change after treatment for 4 weeks. However, the levels of mercury in blood and urine of other experimental groups increased significantly with time and the increase in dosage, and so did the level of mercury in major organ. At 4 weeks, all experimental groups showed increase in the content of mercury, and kidneys displayed the highest level, whereas brain displayed the lowest level After the drug discontinuance for 4 weeks, the mercury level in blood and urine of every dose group recovered to normal, with significant decline in the content of mercury in each organ.
    Conclusion: After transdermal administration in rats for 4 weeks, there was no obvious absorption of mercury in blood. Mercury was mainly accumulated in kidneys and excreted through urine. The results suggest that the patients' mercury content and kidney function indexes need to be monitored in long-term clinical use of Yuhong ointment.

    Abstract Objective: To develop a method for determining the content of mercury contained in Yuhong ointment.
    Method: The wet catalytic digestion method was adopted for the pretreatment, and the mercury content in Yuhong ointment was determined by hydride generation-atomic absorption spectrometry (HG-AAS).
    Result: The mercury showed a good linear relation in the range from 2 to 20 microg x L(-1), with the average recovery of 104.27% and RSD of 3.37%. The RSD for real sample repeated measurement was determined to be 8.4%. The mercury content in Yuhong ointment was detected in range from 0.7 to 1.5 mg x g(-1).
    Conclusion: The proposed method is accurate, highly reproducible and it can be used to control mercury content of Yuhong ointment.

    Abstract The ribosome is responsible for assembling proteins using 20 naturally occurring L-handed amino acids. However, incorporating non-natural amino acids into a protein is a challenging process needs improvement. In this study, we report a new possible approach to creating nonnatural peptides using ribozymes inspired by the peptidyl transfer center. These RNA scaffolds, which are approximately 100 nucleotides in length, bind to RNase T1 truncated tRNA-like chimeras and bring them into close proximity to facilitate peptide ligation.     Abstract In order to solve the research on the extraction of ceramic sculpture artwork patterns, and, in the process of image reproduction, the problem of too few feature points in the object image, the author proposes an image stitching algorithm that combines SIFT and MSER algorithms. After comprehensively analyzing the principles, advantages, and disadvantages of the current main image stitching methods, in terms of feature matching, based on the K-D tree search algorithm, the improved BBF algorithm is used to improve the search efficiency of feature points. In order to remove the possible cracks in the stitching process, an improved multiband fusion algorithm is used to seamlessly stitch the registered images. The results show that the feature points detected by the one-dimensional normal distribution algorithm are on average 0.1%, 0.5%, 1.7%, 4.4%, and 9.2%. The algorithm combining SIFT and MSER to extract feature points can reach 3.6%, 4.6%, 8.4%, 15%, and 19.1%. The experimental results show that the algorithm proposed by the author can extract more image feature points to facilitate later image registration. The image blur phenomenon in the original image fusion algorithm is solved, and a complete and clear two-dimensional plane pattern is finally obtained.

    Abstract Quality variables play a pivotal role in monitoring the performance of chemical production systems. However, certain critical quality variables cannot be measured online through instruments. In such scenarios, using soft sensors becomes imperative to enable real-time measurements, accurately reflecting the system's operational status. The development of high-performance soft sensors requires abundantly labeled samples. Nevertheless, the prolonged periods and substantial costs associated with acquiring quality variable data pose challenges in obtaining sufficient labeled samples. Therefore, this paper proposes a regression generative adversarial network to generate virtual samples. The proposed method considers the mapping relationship between auxiliary and target variables while learning the data distribution. Moreover, the importance-weighted autoencoder is introduced to enhance the training stability of the generative model. The virtual samples, selected by using the similarity measurement algorithm, are incorporated into the training set. This inclusion addresses the diminished predictive performance of soft sensors when labeled samples are insufficient. The soft sensor employed in the anaerobic digestion process serves as a case study to illustrate the efficacy of the proposed generative method. Experimental results validate that the virtual samples generated by the proposed method exhibit greater proximity to the actual samples compared to those of other methods. Furthermore, integrating virtual samples into the training process of the long short-term memory-based soft sensor yields a 21.03% reduction in root-mean-square error compared with that of using the original training set alone.

    Abstract The ribosome is a large ribonucleoprotein complex that assembles proteins processively along mRNA templates. The diameter of the ribosome is approximately 20 nm to accommodate large tRNA substrates at the A-, P- and E-sites. Consequently, the ribosome dynamics are naturally de-phased quickly. Single molecule method can detect each ribosome separately and distinguish inhomogeneous populations, which is essential to reveal the complicated mechanisms of multi-component systems. We report the details of a smFRET method based on the Nikon Ti2 inverted microscope to probe the ribosome dynamics between the ribosomal protein L27 and tRNAs. The L27 is labeled at its unique Cys 53 position and reconstituted into a ribosome that is engineered to lack L27. The tRNA is labeled at its elbow region. As the tRNA moves to different locations inside the ribosome during the elongation cycle, such as pre- and post- translocation, the FRET efficiencies and dynamics exhibit differences, which have suggested multiple subpopulations. These subpopulations are not detectable by ensemble methods. The TIRF-based smFRET microscope is built on a manual or motorized inverted microscope, with home-built laser illumination. The ribosome samples are purified by ultracentrifugation, loaded into a home-built multi-channel sample cell and then illuminated via an evanescent laser field. The reflection laser spot can be used to achieve feedback control of perfect focus. The fluorescence signals are separated by a motorized filter-turret and collected by two digital CMOS cameras. The intensities are retrieved via the NIS-Elements software.
