Abstract Technologies in laboratory diagnostics are changing fast with progress in understanding and therapy of diseases. Unfortunately, new analyzers are often needed to be installed in a clinical laboratory to implement such techniques. The demand for new hardware is a bottleneck in improving the diagnostic services for many facilities with limited resources. In this regard, hemostasis laboratories take a slightly different position. Because many

Abstract Standard deep learning algorithms are implemented using floating-point real numbers. This presents an obstacle for implementing them on low-end devices which may not have dedicated floating-point units (FPUs). As a result, researchers in tinyML have considered machine learning algorithms that can train and run a deep neural network (DNN) on a low-end device using integer operations only. In this paper we propose PocketNN, a light and self-contained proof-of-concept framework in pure C++ for the training and inference of DNNs using only integers. Unlike other approaches, PocketNN directly operates on integers without requiring any explicit quantization algorithms or customized fixed-point formats. This was made possible by pocket activations, which are a family of activation functions devised for integer-only DNNs, and an emerging DNN training algorithm called direct feedback alignment (DFA). Unlike the standard backpropagation (BP), DFA trains each layer independently, thus avoiding integer overflow which is a key problem when using BP with integer-only operations. We used PocketNN to train some DNNs on two well-known datasets, MNIST and Fashion-MNIST. Our experiments show that the DNNs trained with our PocketNN achieved 96.98% and 87.7% accuracies on MNIST and Fashion-MNIST datasets, respectively. The accuracies are very close to the equivalent DNNs trained using BP with floating-point real number operations, such that accuracy degradations were just 1.02%p and 2.09%p, respectively. Finally, our PocketNN has high compatibility and portability for low-end devices as it is open source and implemented in pure C++ without any dependencies.

Abstract In order to build self-consistent personalized dialogue agents, previous research has mostly focused on textual persona that delivers personal facts or personalities. However, to fully describe the multi-faceted nature of persona, image modality can help better reveal the speaker's personal characteristics and experiences in episodic memory (Rubin et al., 2003; Conway, 2009). In this work, we extend persona-based dialogue to the multimodal domain and make two main contributions. First, we present the first multimodal persona-based dialogue dataset named MPCHAT, which extends persona with both text and images to contain episodic memories. Second, we empirically show that incorporating multimodal persona, as measured by three proposed multimodal persona-grounded dialogue tasks (i.e., next response prediction, grounding persona prediction, and speaker identification), leads to statistically significant performance improvements across all tasks. Thus, our work highlights that multimodal persona is crucial for improving multimodal dialogue comprehension, and our MPCHAT serves as a high-quality resource for this research.

Abstract Understanding how nanoparticles (NPs) interact with biological systems is important in many biomedical research areas. However, the heterogeneous nature of biological systems, including the existence of numerous cell types and multitudes of key environmental factors, makes these interactions extremely challenging to investigate precisely. Here, using a single-cell-based, high-dimensional mass cytometry approach, we demonstrated that the presence of protein corona has significant influences on the cellular associations and cytotoxicity of gold NPs for human immune cells, and those effects vary significantly with the types of immune cells and their subsets. The altered surface functionality of protein corona reduced the cytotoxicity and cellular association of gold NPs in most cell types (e.g., monocytes, dendritic cells, B cells, natural killer (NK) cells, and T cells) and those immune cells selected different endocytosis pathways such as receptor-mediated endocytosis, phagocytosis, and micropinocytosis. However, even slight alterations in the major cell type (phagocytic cells and non-phagocytic cells) and T cell subsets (e.g., memory and naive T cells) resulted in significant protein corona-dependent variations in their cellular dose of gold NPs. Especially, naive T killer cells exhibited additional heterogeneity than memory T killer cells, with clusters exhibiting distinct cellular association patterns in single-cell contour plots. This multi-parametric analysis of mass cytometry data established a conceptual framework for a more holistic understanding of how the human immune system responds to external stimuli, paving the way for the application of precisely engineered NPs as promising tools of nanomedicine under various clinical settings, including targeted drug delivery and vaccine development.

Abstract This study was conducted to investigate the effects of protease supplementation and different nutrient density of diets in growing-finishing pigs. A total of one hundred-eight crossbred growing pigs ([Landrace × Yorkshire] × Duroc) with an initial body weight (BW; 18.74 ± 3.46 kg) were used for 15 weeks. Pigs were randomly assigned to six dietary treatments with 6 replicates of 3 pigs per pen in a 3 × 2 factorial through the following arrangement: Three groups of protease (1, Basal diets; 2, Protease A: 125 mg/kg protease derived from

Abstract Increasing production and application of silver nanoparticles (Ag NPs) have raised concerns on their possible adverse effects on human health. However, a comprehensive understanding of their effects on biological systems, especially immunomodulatory responses involving various immune cell types and biomolecules (e.g., cytokines and chemokines), is still incomplete. In this study, a single-cell-based, high-dimensional mass cytometry approach is used to investigate the immunomodulatory responses of Ag NPs using human peripheral blood mononuclear cells (hPBMCs) exposed to poly-vinyl-pyrrolidone (PVP)-coated Ag NPs of different core sizes (i.e., 10-, 20-, and 40-nm). Although there were no severe cytotoxic effects observed,
