    Abstract Background: In recent years, with a large number of rural people going out to work, the mental problems of left-behind children have been widely concerned by people from all walks of life.
    Objective: The purpose is to improve the teaching quality of music education of left-behind children and solve their unique mental problems.
    Methods: Based on the questionnaire survey, the mental state of left-behind children is studied, and the mental problems of left-behind children are intervened based on musical practice classes. First, the left-behind children's mental status is studied. Second, through the questionnaire, the left-behind children's music classroom status is studied and analyzed. Then, the musical classroom intervention experiment is designed to prevent and adjust the left-behind children's mental problems.
    Results: The results show that more than 90% of left-behind children spend less than one month with their parents every year. Most of the left-behind children are supervised by their grandparents, and 68% of them have been accompanied by their grandparents for more than nine months. Due to the lack of parents' company, most of the left-behind children are lack of self-confidence and timid and tend to care about others' opinions. Musical practice classes have a good effect on the regulation and prevention of left-behind children's mental problems.
    Conclusions: This study uses a questionnaire survey method to explore the new form of left-behind children's music classroom, preventing and regulating left-behind children's mental problems, which provides a reference for the research in related fields.

    Abstract Non-alcoholic fatty liver disease (NAFLD) is a chronic liver disease worldwide. Numerous evidence has demonstrated that metabolic reprogramming serves as a hallmark associated with an elevated risk of NAFLD progression. Selenoprotein W (SelW) is an extensively expressed hepatic selenoprotein that plays a crucial role in antioxidant function. Here, we first demonstrated that SelW is a significantly distinct factor in the liver tissue of NAFLD patients through the Gene Expression Omnibus (GEO) database. Additionally, loss of SelW alleviated hepatic steatosis induced by a high-fat diet (HFD), and was accompanied by the regulation of metabolic and inflammatory pathways as verified by transcriptomic analysis. Moreover, co-immunoprecipitation (CO-IP), liquid chromatography-tandem mass spectrometry (LC-MS), laser scanning confocal microscopy (LSCM) and molecular docking analysis were subsequently implemented to identify Pyruvate Kinase M2 (PKM2) as a potential interacting protein of SelW. Meanwhile, SelW modulated PKM2 translocation into the nucleus to trigger transactivation of the HIF-1α, in further mediating mitochondrial apoptosis, eventually resulting in mitochondrial damage, ROS excessive production and mtDNA leakage. Additionally, mito-ROS accumulation induced the activation of the NLRP3 inflammasome-mediated pyroptosis, thereby facilitating extracellular leakage of mtDNA. The escaped mtDNA then evokes the cGAS-STING signaling pathway in macrophage, thus inducing a shift in macrophage phenotype. Together, our results suggest SelW promotes hepatocyte apoptosis and pyroptosis by regulating metabolic reprogramming to activate cGAS/STING signaling of macrophages, thereby exacerbating the progression of NAFLD.

    Abstract Real-life events, behaviors, and interactions produce sequential data. An important but rarely explored problem is to analyze those nonoccurring (also called negative) yet important sequences, forming negative sequence analysis (NSA). A typical NSA area is to discover negative sequential patterns (NSPs) consisting of important nonoccurring and occurring elements and patterns. The limited existing work on NSP mining relies on frequentist and downward closure property-based pattern selection, producing large and highly redundant NSPs, nonactionable for business decision-making. This work makes the first attempt for actionable NSP discovery. It builds an NSP graph representation, quantifies both explicit occurrence and implicit nonoccurrence-based element and pattern relations, and then discovers significant, diverse, and informative NSPs in the NSP graph to represent the entire NSP set for discovering actionable NSPs. A DPP-based NSP representation and actionable NSP discovery method, EINSP, introduces novel and significant contributions to NSA and sequence analysis: 1) it represents NSPs by a determinantal point process (DPP)-based graph; 2) it quantifies actionable NSPs in terms of their statistical significance, diversity, and strength of explicit/implicit element/pattern relations; and 3) it models and measures both explicit and implicit element/pattern relations in the DPP-based NSP graph to represent direct and indirect couplings between NSP items, elements, and patterns. We substantially analyze the effectiveness of EINSP in terms of various theoretical and empirical aspects, including complexity, item/pattern coverage, pattern size and diversity, implicit pattern relation strength, and data factors.

    Abstract Introduction: Hypertriglyceridemia and its derivatives are independent predictors of diabetes mellitus type 2 (T2DM). However, the relationship between triglyceride concentrations within the normal range and the incidence of T2DM remains to be clarified. This study investigated the potential relationship between variations in plasma triglyceride levels within the normal range and T2DM onset using data from a longitudinal study of health and retirement in China.
    Methods: Between, 2010 and, 2016, we conducted a retrospective cohort study involving 36,441 individuals with normal triglyceride levels. Using a Cox proportional hazards regression model, we examined the connection between normal triglyceride levels and T2DM incidence. We employed this method with smooth curve fitting to investigate potential nonlinear associations. Subgroup analyses were performed based on age, sex, body mass index, smoking and drinking status, hypertension, and family history of diabetes.
    Results: A significant linear relationship was observed between normal triglyceride levels and the incidence of T2DM. The hazard ratio for T2DM in individuals with normal triglycerides was 1.81 (95% confidence interval: 1.39, 2.36); P<0.001). Kaplan-Meier analysis further demonstrated a prospective association between the higher tertiles of normal triglyceride levels and the development of T2DM (P<0.001). Subgroup analysis revealed a stronger positive correlation between normal triglyceride levels in females and the risk of T2DM.
    Discussion: An increase in triglyceride levels within the normal range is related to a continuous increase in the incidence of T2DM in the general population. These findings show that almost everyone can benefit from reducing triglyceride levels, further emphasizing the importance of lifestyle changes in the general population.

    Abstract Laparoscopic and robotic surgery is a challenge to the surgeon's hand-eye coordination ability, which requires constant practice. Traditional mentor training is gradually shifting to simulation training based on various models. Laparoscopic and robotic bilioenteric anastomosis is an important and difficult operation in hepatobiliary surgery. We constructed and optimized the reusable modular 3D-printed models of choledochal cyst. The aim of this study was to verify the ability of this optimized model to distinguish between surgeons with different levels of proficiency and the benefits of repeated practice. A total of 12 surgeons with different levels participated in the study. Operation completion time and OSATS score were recorded. The model was validated by Likert scale. Surgeons were shown the steps and contents before performing laparoscopic or robotic bilioenteric anastomosis using the model. Surgeons with different levels of experience showed different levels when performing laparoscopic bilioenteric anastomosis on this model. Repeated training can significantly shorten the time of laparoscopic bilioenteric anastomosis and improve the operation scores of surgeons with different levels of experience. At the same time, preliminary results have shown that the performance of surgeons on the domestic robotic platform was basically consistent with their laparoscopic skills. This model may distinguish surgeons with different levels of experience and may improve surgical skills through repeated practice. It is worth noting that in order to draw more reliable conclusions, more subjects should be collected and more experiments should be done in the future.
