    Abstract In this paper, a scheme to realize encryption and digital identity authentication at the same time is proposed for enhancing the physical-layer security of point-to-point optical links (PPOL). Exploiting identity code encrypted by the key as authentication information effectively resists passive eavesdropping attacks in fingerprint authentication. The proposed scheme theoretically realizes secure key generation and distribution (SKGD) by phase noise estimation of the optical channel and the generation of identity codes with good randomness and unpredictability by the four-dimensional (4D) hyper-chaotic system. The local laser, erbium doped fiber amplifier (EDFA), and public channel provide the entropy source of uniqueness and randomness to extract symmetric key sequences for legitimate partners. The simulation conducted in a quadrature phase shift keying (QPSK) PPOL system over 100km standard single mode fiber verify successfully that 0.95Gbit/s error-free SKGD. The unpredictability and high sensitivity to the initial value and control parameters of the 4D hyper-chaotic system provide a huge space of ~10

    Abstract Objective: Abundant neutrophils have been identified in both ruptured and unruptured intracranial aneurysm (IA) domes, with their function and clinical implication being poorly characterized.
    Materials and methods: We employed single-cell RNA sequencing (scRNA-Seq) datasets of both human and murine model, and external bulk mRNA sequencing datasets to thoroughly explore the features and functional heterogeneous of neutrophils infiltrating the IA dome.
    Results: We found that both unruptured and ruptured IA dome contain a substantial population of neutrophils, characterized by FCGR3B, G0S2, CSF3R, and CXCR2. These cells exhibited heterogeneity in terms of function and differentiation. Despite similar transcriptional activation, neutrophils in IA dome expressed a repertoire of gene programs that mimicked transcriptomic alterations observed from bone marrow to peripheral blood, showing self-similarity. In addition, the recruitment of neutrophils in unruptured IA was primarily mediated by monocytes/macrophages, and once ruptured, both neutrophils, and a specific subset of inflammatory smooth muscle cells (SMCs) were involved in the process. The receiver operator characteristic curve (ROC) analysis indicated that distinct neutrophil subclusters were associated with IA formation and rupture, respectively. By reviewing current studies, we found that neutrophils play a detrimental role to IA wall integrity through secreting specific ligands, ferroptosis driven by ALOX5AP and PTGS2, and the formation of neutrophil extracellular traps (NETs) mediated by PADI4.
    Interpretation: This study delineated the biology and potential clinical implications of neutrophils in IA dome and provided a reliable basis for future researches.

    Abstract Aptamers are single-stranded oligonucleotides that bind to their targets via specific structural interactions. To improve the properties and performance of aptamers, modified nucleotides are incorporated during or after a selection process such as systematic evolution of ligands by exponential enrichment (SELEX). We summarize the latest modified nucleotides and strategies used in modified (mod)-SELEX and post-SELEX to develop modified aptamers, highlight the methods used to characterize aptamer-target interactions, and present recent progress in modified aptamers that recognize different targets. We discuss the challenges and perspectives in further advancing the methodologies and toolsets to accelerate the discovery of modified aptamers, improve the throughput of aptamer-target characterization, and expand the functional diversity and complexity of modified aptamers.

    Abstract Aptamers are single-stranded oligonucleotides that recognize their target of interest via specific structural interactions. To improve the properties and performance of aptamers, modified nucleotides are introduced during or after selection processes referred to as the systematic evolution of ligands by exponential enrichment (SELEX). Herein, we summarize the latest modified nucleotides and strategies used in modified (mod)-SELEX and post-SELEX to develop modified aptamers, highlight the approaches commonly used to characterize modified aptamer-target interactions, and present recent progress in modified aptamers recognizing different targets. We discuss the challenges and perspectives to further advance the methodologies and toolsets capable of speeding up the discovery of modified aptamers, improving the throughput of aptamer-target characterization, and expanding the functional diversity and complexity of modified aptamers.

    Abstract G-quadruplexes (G4) are special nucleic acid structures with diverse conformational polymorphisms. Selective targeting of G-quadruplex conformations and regulating their biological functions provide promising therapeutic intervention. Despite the large repertoire of G4-binding tools, only a limited number of them can specifically target a particular G4 conformation. Here, we introduce a novel method, G4-SELEX-Seq and report the development of the first L-RNA aptamer, L-Apt12-6, with high binding selectivity to parallel G4 over other nucleic acid structures. Using parallel dG4 c-kit 1 as an example, we demonstrate the strong binding affinity between L-Apt12-6 and c-kit 1 dG4 in vitro and in cells, and notably report the applications of L-Apt12-6 in controlling DNA replication and gene expression. Our results suggest that L-Apt12-6 is a valuable tool for targeting parallel G-quadruplex conformation and regulating G4-mediated biological processes. Furthermore, G4-SELEX-Seq can be used as a general platform for G4-targeting L-RNA aptamers selection and should be applicable to other nucleic acid structures.
