Abstract Normal-tension glaucoma is a form of optic nerve degeneration that is characterized by loss of retinal ganglion cells independent of eye pressure elevation. In this issue of the JCI, Pan et al. report the discovery in a Japanese family of a mutation in the METTL23 gene, which encodes a histone arginine methyltransferase that causes normal-pressure glaucoma in haploinsufficiency. Inherited as an autosomal dominant condition, METTL23 deficiency revealed an important function in the regulation of pS2 and the downstream NF-κB signaling pathway, which has previously been linked to glaucomatous optic nerve degeneration. These findings are the first direct link between defective epigenetic regulatory machinery and genetic forms of optic nerve degeneration.

Abstract Apoptotic colopathy is a pattern of injury not frequently associated with Crohn's disease (CD) despite its association with medications used in CD management. We describe a patient with CD on methotrexate who underwent a diagnostic colonoscopy for abdominal pain and diarrhea, with biopsies demonstrating apoptotic colopathy. After methotrexate discontinuation, repeat colonoscopy demonstrated resolution of apoptotic colopathy in addition to diarrhea improvement.

Abstract Medical professionals such as physicians and veterinarians are responsible for appropriate antimicrobial prescription (AMP) and use. Although seemingly straightforward, the factors influencing antibiotic prescription, a category of antimicrobials, are complex. Many studies have been conducted in the past two decades on this subject. As a result, there is a plethora of empirical evidence regarding the factors influencing clinicians' AMP practices.
Aim: A systematic review of AMR studies on AMP was conducted, condensing findings according to a combination of the Knowledge, Attitude, and Practice (KAP) and Capacity, Opportunity, Motivation-Behavior (COM-B) models. Review findings were then synthesized and analyzed for policy implementation according to the Consolidated Framework for Implementation Research (CFIR).
Design and methodology: A systematic literature review was conducted according to PRISMA guidelines to identify peer-reviewed papers indexed in pre-determined medical science, social sciences, and humanities databases that apply the KAP model in their investigations. Antimicrobial prescription factors were compared and contrasted among low- and middle-income countries (LMICs) and high-income countries (HICs).
Findings: The KAP model is a heuristic and structured framework for identifying and classifying respondents' knowledge. However, other than medical knowledge, factors that influence prescription decision-making can be expanded to include attitudes, perception, personal affinities, professional circumstances, relational pressure, and social norms.

Abstract Urban forests provide multiple ecosystem services for city dwellers, amongst which improving public health via mitigating mental stresses and providing attractive spaces for diverse physical activities has attracted increasing attention from scholars and policy makers within the context of the ongoing COVID-19 pandemic as well as urgently-needed post-pandemic urban transformation towards healthy cities. This short communication summarizes existing empirical evidence pertinent to the linkage between urban forests and public health maintenance and improvement, highlights three underlying mechanisms, i.e., physiological, psychological, and immunological pathways, and outlines practical implications for the establishment and management of urban forests as a strategy for planning healthy cities.
