    Abstract Highly pathogenic emerging and reemerging viruses have serious public health and socioeconomic implications. Although conventional live virus research methods can more reliably investigate disease pathogenicity and evaluate antiviral products, they usually depend on high-level biosafety laboratories and skilled researchers; these requirements hinder in vitro assessments of efficacy, as well as efforts to test vaccines and antibody drugs. In contrast, pseudotyped viruses (i.e., single-round infectious viruses that mimic the membrane structures of various live viruses) are widely used in studies of highly pathogenic viruses because they can be handled in biosafety level 2 facilities. This chapter provides a concise overview of various aspects of pseudotyped virus technologies, including (1) exploration of the mechanisms of viral infection; (2) evaluation of the efficacies of vaccines and monoclonal antibodies based on pseudovirion-based neutralization assay; (3) assessment of antiviral agents (i.e., antibody-based drugs and inhibitors); (4) establishment of animal models of pseudotyped virus infection in vivo; (5) investigation of the evolution, infectivity, and antigenicity of viral variants and viral glycosylation; and (6) prediction of antibody-dependent cell-mediated cytotoxic activity.

    Abstract Artificial molecular photoswitches that can be reversibly controlled into different configurations by external light stimulation have broad application prospects in various fields, such as materials and chemical biology. Among them, the pyrrole hemithioindigo (PHT) photoswitch has a geometric structure similar to that of the fluorescent protein chromophore. What happens when the chromophore is replaced by PHT, and does it achieve similar functions to the original one? To answer these questions, we carried out ONIOM(QM/MM) and classical molecular dynamics studies on the photoisomerization mechanism and spectroscopic properties of PHT in the fluorescent protein. The results showed that in the protein environment, the fate of excited PHT is governed by the competition between fluorescence emission and hula-twist isomerization. Due to the strong steric hindrance effects caused by the interlacing residues in the protein that restrict the PHT conformation transformation, the cis-to-trans isomerization process of PHT needs to overcome a barrier of at least 4.9 kcal/mol; thus, fluorescence emission is more dominant in competition. It is also found that the intermolecular interaction between LYS67 and the carbonyl oxygen of PHT has a significant effect on the fluorescence emission. These results revealed the photochemical reaction mechanism of a light-driven molecular switch in the fluorescent protein and provided further theoretical support for the field of chemical biology.

    Abstract Tourism promotes the economic development of agro-cultural heritage sites while causing the problem of spatial alienation, so exploring the spatial alienation problems based on the integration of culture and tourism is of great significance for achieving the sustainable development of tourism in agro-cultural heritage sites. This study took the Yuanyang Hani Terrace agro-cultural heritage site as the research area, constructed the model of the spatial system of the agro-cultural heritage site based on the classical grounded theory, and analyzed the spatial characteristics of the agro-cultural heritage site. The results show that the spatial disorder of the agro-cultural heritage site is caused by the interaction of the imbalance of physical space, the fragmentation of cultural space and the complications of social space. Next, this study constructed the model for the spatial restructuring of the agro-cultural heritage site in the context of culture and tourism integration, with the integration of management systems as a guiding force, the integration of talent systems as a crucial force, the integration of agricultural, cultural and tourism resources as a core force, the integration of the cultural and tourism industries as a driving force, the integration of ecology and culture as a basic force, and the integration of digital scenes as an innovative force. This study expands and deepens the spatial theory of tourist destinations and the knowledge system of cultural heritage tourism, and provides a systematic implementation framework and management tools for the integration of culture and tourism and the high-quality development of agro-cultural heritage sites.

    Abstract In this paper, a stochastic SIB(Susceptible-Infected-Vibrios) cholera model with saturation recovery rate and Ornstein-Uhlenbeck process is investigated. It is proved that there is a unique global solution for any initial value of the model. Furthermore, the sufficient criterion of the stationary distribution of the model is obtained by constructing a suitable Lyapunov function, and the expression of probability density function is calculated by the same condition. The correctness of the theoretical results is verified by numerical simulation, and the specific expression of the marginal probability density function is obtained.

    Abstract Background: In the past few years, a burgeoning interest has emerged in applying gamification to promote desired health behaviors. However, little is known about the effectiveness of such applications in the HIV prevention and care continuum among men who have sex with men (MSM).
    Objective: This study aims to summarize and evaluate research on the effectiveness of gamification on the HIV prevention and care continuum, including HIV-testing promotion; condomless anal sex (CAS) reduction; and uptake of and adherence to pre-exposure prophylaxis (PrEP), postexposure prophylaxis (PEP), and antiretroviral therapy (ART).
    Methods: We comprehensively searched PubMed, Embase, the Cochrane Library, Web of Science, Scopus, and the Journal of Medical Internet Research and its sister journals for studies published in English and Chinese from inception to January 2024. Eligible studies were included when they used gamified interventions with an active or inactive control group and assessed at least one of the following outcomes: HIV testing; CAS; and uptake of and adherence to PrEP, PEP, and ART. During the meta-analysis, a random-effects model was applied. Two reviewers independently assessed the quality and risk of bias of each included study.
    Results: The systematic review identified 26 studies, including 10 randomized controlled trials (RCTs). The results indicated that gamified digital interventions had been applied to various HIV outcomes, such as HIV testing, CAS, PrEP uptake and adherence, PEP uptake, and ART adherence. Most of the studies were conducted in the United States (n=19, 73%). The most frequently used game component was gaining points, followed by challenges. The meta-analysis showed gamification interventions could reduce the number of CAS acts at the 3-month follow-up (n=2 RCTs; incidence rate ratio 0.62, 95% CI 0.44-0.88). The meta-analysis also suggested an effective but nonstatistically significant effect of PrEP adherence at the 3-month follow-up (n=3 RCTs; risk ratio 1.16, 95% CI 0.96-1.38) and 6-month follow-up (n=4 RCTs; risk ratio 1.28, 95% CI 0.89-1.84). Only 1 pilot RCT was designed to evaluate the effectiveness of a gamified app in promoting HIV testing and PrEP uptake. No RCT was conducted to evaluate the effect of the gamified digital intervention on PEP uptake and adherence, and ART initiation among MSM.
    Conclusions: Our findings suggest the short-term effect of gamified digital interventions on lowering the number of CAS acts in MSM. Further well-powered studies are still needed to evaluate the effect of the gamified digital intervention on HIV testing, PrEP uptake, PEP initiation and adherence, and ART initiation in MSM.

    Abstract Due to the advantages of its numerous modification sites, predictable structure, high thermal stability, and excellent biocompatibility, DNA is the ideal choice as a key component of biosensors. DNA biosensors offer significant advantages over existing bioanalytical techniques, addressing limitations in sensitivity, selectivity, and limit of detection. Consequently, they have attracted significant attention from researchers worldwide. Here, we exemplify four foundational categories of functional nucleic acids: aptamers, DNAzymes, i-motifs, and G-quadruplexes, from the perspective of the structure-driven functionality in constructing DNA biosensors. Furthermore, we provide a concise overview of the design and detection mechanisms employed in these DNA biosensors. Noteworthy advantages of DNA as a sensor component, including its programmable structure, reaction predictility, exceptional specificity, excellent sensitivity, and thermal stability, are highlighted. These characteristics contribute to the efficacy and reliability of DNA biosensors. Despite their great potential, challenges remain for the successful application of DNA biosensors, spanning storage and detection conditions, as well as associated costs. To overcome these limitations, we propose potential strategies that can be implemented to solve these issues. By offering these insights, we aim to inspire subsequent researchers in related fields.

    Abstract A transmission dynamics model with the logistic growth of cystic echinococcus in sheep was formulated and analyzed. The basic reproduction number was derived and the results showed that the global dynamical behaviors were determined by its value. The disease-free equilibrium is globally asymptotically stable when the value of the basic reproduction number is less than one; otherwise, there exists a unique endemic equilibrium and it is globally asymptotically stable. Sensitivity analysis and uncertainty analysis of the basic reproduction number were also performed to screen the important factors that influence the spread of cystic echinococcosis. Contour plots of the basic reproduction number versus these important factors are presented, too. The results showed that the higher the deworming rate of dogs, the lower the prevalence of echinococcosis in sheep and dogs. Similarly, the higher the slaughter rate of sheep, the lower the prevalence of echinococcosis in sheep and dogs. It also showed that the spread of echinococcosis has a close relationship with the maximum environmental capacity of sheep, and that they have a remarkable negative correlation. This reminds us that the risk of cystic echinococcosis may be underestimated if we ignore the increasing number of sheep in reality.
