    Abstract The reparative potential of cardiac Lin-KIT+ (KIT) cells is influenced by their population, but identifying their markers is challenging due to changes in phenotype during in vitro culture. Resolving this issue requires uncovering cell heterogeneity and discovering new subpopulations. Single-cell RNA sequencing (scRNA-seq) can identify KIT cell subpopulations, their markers, and signaling pathways. We used 10× genomic scRNA-seq to analyze cardiac-derived cells from adult mice and found 3 primary KIT cell populations: KIT1, characterized by high-KIT expression (KITHI), represents a population of cardiac endothelial cells; KIT2, which has low-KIT expression (KITLO), expresses transcription factors such as KLF4, MYC, and GATA6, as well as genes involved in the regulation of angiogenic cytokines; KIT3, with moderate KIT expression (KITMOD), expresses the cardiac transcription factor MEF2C and mesenchymal cell markers such as ENG. Cell-cell communication network analysis predicted the presence of the 3 KIT clusters as signal senders and receivers, including VEGF, CXCL, and BMP signaling. Metabolic analysis showed that KIT1 has the low activity of glycolysis and oxidative phosphorylation (OXPHOS), KIT2 has high glycolytic activity, and KIT3 has high OXPHOS and fatty acid degradation activity, indicating distinct metabolic adaptations of the 3 KIT populations. Through the systemic infusion of KIT1 cells in a mouse model of myocardial infarction, we observed their involvement in promoting the formation of new micro-vessels. In addition, in vitro spheroid culture experiments demonstrated the cardiac differentiation capacity of KIT2 cells.

    Abstract Under the circumstances of global warming and rapid urbanization, damage caused by urban inundation are becoming increasingly severe, attracting the attention of both researchers and governors. The accurate simulation of urban inundation is essential for the prevention of inundation hazards. In this study, a 1D pipe network and a 2D urban inundation coupling model constructed by InfoWorks ICM was used to simulate the inundation conditions in the typical urbanized area in the north of Lin’an. Two historical rainfall events in 2020 were utilized to verify the modeling results. The spatial–temporal variation and the causes of urban inundation under different designed rainfalls were studied. The results were as follows: (1) The constructed model had a good simulation accuracy, the Nash–Sutcliffe efficiency coefficient was higher than 0.82, R 2 was higher than 0.87, and the relative error was ±20%. (2) The simulation results of different designed rainfall scenarios indicated that the maximum inundation depth and inundation extent increased with the increase in the return period, rainfall peak position coefficient, and rainfall duration. According to the analysis results, the urban inundation in Lin’an is mainly affected by topography, drainage network (spatial distribution and pipe diameter), and rainfall patterns. The results are supposed to provide technical support and a decision-making reference for the urban management department of Lin’an to design inundation prevention measures.

    Abstract TSP is a classical NP-hard combinatorial optimization problem with many practical variants. LKH is one of the state-of-the-art local search algorithms for the TSP. LKH-3 is a powerful extension of LKH that can solve many TSP variants. Both LKH and LKH-3 associate a candidate set to each city to improve the efficiency, and have two different methods, $\alpha$-measure and POPMUSIC, to decide the candidate sets. In this work, we first propose a Variable Strategy Reinforced LKH (VSR-LKH) algorithm, which incorporates three reinforcement learning methods (Q-learning, Sarsa, Monte Carlo) with LKH, for the TSP. We further propose a new algorithm called VSR-LKH-3 that combines the variable strategy reinforcement learning method with LKH-3 for typical TSP variants, including the TSP with time windows (TSPTW) and Colored TSP (CTSP). The proposed algorithms replace the inflexible traversal operations in LKH and LKH-3 and let the algorithms learn to make a choice at each search step by reinforcement learning. Both LKH and LKH-3, with either $\alpha$-measure or POPMUSIC, can be significantly improved by our methods. Extensive experiments on 236 widely-used TSP benchmarks with up to 85,900 cities demonstrate the excellent performance of VSR-LKH. VSR-LKH-3 also significantly outperforms the state-of-the-art heuristics for TSPTW and CTSP.

    Abstract Background: Inflammatory bowel disease (IBD) is a chronic and recurrent inflammatory disorder in gastrointestinal tract. Shen Ling Bai Zhu San (SLBZS), which has a long history of use in Traditional Chinese Medicine (TCM), has been widely used to treat gastrointestinal diseases. The isolated fractions of TCM have also been proved to possess an important potential for treating diseases, which are due to their effective components.
    Purpose: In this study, we examined the possibility that SLBZS and its isolated active fractions may prevent DSS-induced colitis, and investigated the potential mechanisms by regulating genetic profile of colon.
    Methods: Colitis mice were induced by 2.5% DSS for 7 days, and then SLBZS and different SLBZS extracts were administrated to protect the mice for 7 days. Body weight, diarrhea, bleeding in stool, colon length, spleen weight, cytokines of serum and colon and pathology of colon were assessed. The level of Ginsenoside Rg1, Re and Rb1 in different SLBZS extracts and qualitative analysis of n-butanol extract of SLBZS (S-Nb) was performed by HPLC and LC-MS, respectively. And the effects of S-Nb on the transcriptome in colitis were investigated.
    Results: Our results showed that SLBZS and S-Nb significantly regained body weight, reduced DAI, splenomegaly and the length of colon and attenuated histological damage of the colon. Meanwhile, SLBZS and S-Nb markedly reduced the levels of TNF-α, IL-1β and IL-6 and increased the level of IL-10 in serum and colon. These effects may be associated with the high levels of Ginsenoside Rg1, Re and Rb1 and rich variety of compounds in S-Nb including 6 ginsenosides, glycyrrhizin, L-tryptophan, and so on. Transcriptome analysis revealed that S-Nb selectively regulated 103 differentially expressed genes (DEGs), 36 of which were changed in DSS-induced mice. And the genes of Per2, Per3, Npy and Serpina3m were closely related to colitis and also restored by S-Nb with different extent. Remarkably, these DEGs modulated the biological functions of colitis mice, including extracellular region, response to external stimulus, MAPK signaling pathway and arginine and proline metabolism.
    Conclusions: These data indicated that SLBZS and S-Nb blunted DSS-induced colitis by modulating differentially expression gene profile and biological functions based on their ginsenosides and rich compounds.

    Abstract Chemoselective deprotonative functionalization of benzylic C–H bonds is challenging, because the arene ring contains multiple aromatic C(sp²)–H bonds, which can be competitively deprotonated and lead to selectivity issues. Recently it was found that bimetallic [MN(SiMe₃)₂ M = Li, Na]/Cs⁺ combinations exhibit excellent benzylic selectivity. Herein, is reported the first deprotonative addition of toluenes to Weinreb amides mediated by LiN(SiMe₃)₂/CsF for the synthesis of a diverse array of 2-arylacetophenones. Surprisingly, simple methyl benzoates also react with toluenes under similar conditions to form 2-arylacetophenones without double addition to give tertiary alcohol products. This finding greatly increases the practicality and impact of this chemistry. Some challenging substrates with respect to benzylic deprotonations, such as fluoro and methoxy substituted toluenes, are selectively transformed to 2-aryl acetophenones. The value of benzylic deprotonation of 3-fluorotoluene is demonstrated by the synthesis of a key intermediate in the preparation of Polmacoxib.

    Abstract The characteristics, sources and risk assessment of heavy metal pollution in community garden soil of Lin’an District were evaluated. The 28 soil samples from community garden were collected for determination of 7 heavy metal elements. The Geostatistical analysis, Spearman correlation coefficient, Principal component analysis and PMF model have explored sources of heavy metal pollution. The health risk assessment model has assessed ecological risk of heavy metals. The results revealed that average concentration of As, Cd, Cr, Cu, Ni, Pb and Zn were 16.0, 0.158, 76.1, 34.6, 45.8, 20.9 and 166 mg kg⁻¹, respectively. Whereas As, Cd, Cr, Cu, Ni and Zn were higher than background values. The spatial distribution of heavy metal pollution in the southwest of the study area was higher than northeast. The pollution sources of Cd, Cu, Ni and Zn in the study area were due to agricultural activities (42.9%), Cr and Pb were from traffic sources (36.2%), and As was domestic pollution (20.9%) according to Spearman correlation coefficient, Principal component analysis and PMF model. The non-carcinogenic risks of As (5.39), Cr (3.53) and Ni (2.07) have a value of 1, which indicated significant risk. The potentially toxic elements have not exceeded maximum threshold of USEPA, with regard to carcinogenic risk, while As (3.37E−05) and Cr (5.74E−05) have exceeded the safety range. It is concluded that soils of community gardens are facing pollution problem due to potentially toxic elements which require environmental monitoring of the soil to reduce risk of human health.
