    Abstract Purpose: To evaluate workflow efficiency and diagnostic quality of a free-breathing 3D stack-of-stars gradient echo (Radial GRE) sequence compared to a breath-hold 3D Cartesian gradient echo (Cartesian GRE) sequence for needle position control in MR-guided liver interventions.
    Materials and methods: 12 MR-guided liver interventions were performed on a 1.5 T Siemens Aera and analyzed retrospectively. 15 series of the Radial GRE sequence were compared to 14 series of the Cartesian GRE sequence regarding the time interval between two consecutive live-scans for needle tracking (Tracking-2-Tracking-Time). The quality of both sequences was compared by the SNR within comparable slices in liver and tumor ROIs. The CNR was calculated by subtraction of the SNR values. Subjective image quality scores of three radiologists were assessed and inter-rater reliability was tested by Fleiss' kappa. Values are given as mean ± SD. P-values < 0.05 were considered as significant.
    Results: The median Tracking-2-Tracking-Time was significantly shorter for the Radial GRE sequence, 185 ± 42 s vs. 212 ± 142 s (p = 0.04) and the median SNR of the liver and tumor ROIs were significantly higher in the Radial GRE sequence, 249 ± 92 vs. 109 ± 67 (p = 0.03) and 165 ± 74 vs. 77 ± 43 (p = 0.02). CNR between tumor and liver ROIs showed a tendency to be higher for the Radial GRE sequence without significance, 68 ± 48 vs. 49 ± 32 (p = 0.28). Mean subjective image quality was 3.33 ± 1.08 vs. 2.62 ± 0.95 comparing Radial and Cartesian GRE with a Fleiss' kappa of 0.39 representing fair inter-rater reliability.
    Conclusion: A free-breathing 3D stack-of-stars gradient echo sequence can simplify the workflow and reduce intervention time, while providing superior image quality. Under local anesthesia, it increases patient comfort and reduces potential risks for needle dislocations in MR-guided liver interventions by avoiding respiratory arrests for needle position control.

    Abstract Two-dimensional culture remains widely employed to determine the bioavailability of orally delivered drugs. To gain more knowledge about drug uptake mechanisms and risk assessment for the patient after oral drug admission, intestinal in vitro models demonstrating a closer similarity to the in vivo situation are needed. In particular, Caco-2 cell-based Transwell® models show advantages as they are reproducible, cost-efficient, and standardized. However, cellular complexity is impaired and cell function is strongly modified as important transporters in the apical membrane are missing. To overcome these limitations, primary organoid-based human small intestinal tissue models were developed recently but the application of these cultures in pre-clinical research still represents an enormous challenge, as culture setup is complex as well as time- and cost-intensive. To overcome these hurdles, we demonstrate the establishment of primary organoid-derived intestinal cell lines by immortalization. Besides exhibiting cellular diversity of the organoid, these immortalized cell lines enable a standardized and more cost-efficient culture. Further, our cell line-based Transwell®-like models display an organ-specific epithelial barrier integrity, ultrastructural features and representative transport functions. Altogether, our novel model systems are cost-efficient with close similarity to the in vivo situation, therefore favoring their use in bioavailability studies in the context of pre-clinical screenings.

    Abstract The objective of this study was to evaluate stearic acid supplementation (C18:0) on the production, composition, fatty acid profile and the expression of lipogenic genes in the mammary gland of late lactating ewes. We used 30 primiparous and multiparous Lacaune ewes with a mean body weight (BW) of 66.5 ± 9.4 kg and body condition score (BCS) of 3.0 ± 0.5 at late lactation (122 ± 12 DEL), producing 1.0 ± 0.3 kg of milk/day. The treatments were: Control and C18 (28 g/animal/d of C18:0). When compared with Control, C18:0 reduced silage dry matter intake (DMI) by 13.1 % (P = 0.0003), milk production by 8.1 % (P = 0.05), lactose production by 3.3 % (P = 0.0004), and lactose content by 3.4 % (P = 0.0002). There was no treatment effect on the production and content of fat, protein, and total solids. The gene expression of acetyl-CoA carboxylase alpha promoter 2 (ACACA PII) and fatty acid synthase (FASN) were reduced by 30 % (P = 0.02) and 26.1 % (P = 0.04), respectively. The C18:0 supplementation is not effective in increasing the fat content in late lactating dairy ewes and reduces DMI, milk production, lactose levels, and gene expression involved in fatty acid synthesis.

    Abstract This paper provides an introduction to and discussion of SIGNAL, an industry-scale process data querying language and engine for large-scale cloud-based systems that is developed by SAP Signavio. SIGNAL is optimized for fast read access to process data in event log format and utilizes an in-memory columnar store to this end. To facilitate usability, SIGNAL uses an SQL-like syntax with additional domain-specific querying features and in particular row-pattern matching-based temporal operators. Also, the paper highlights research challenges related to process querying that are informed by the implementation and application of SIGNAL.
