    Abstract Methods: The potential active ingredients and corresponding potential targets of BSYS Capsule were obtained from the TCMSP, BATMAN-TCM, Swiss Target Prediction platform, and literature research. Disease targets of CNSD were explored through the GeneCards and the DisGeNET databases. The matching targets of BSYS in CNSD were identified from a Venn diagram. The protein-protein interaction (PPI) network was constructed using bioinformatics methods. Gene Ontology (GO) function and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were performed to predict the mechanisms of BSYS. Furthermore, the neuroprotective effects of BSYS were evaluated using a cell model of hydrogen peroxide- (H
    Results: A total of 59 potential bioactive components of BSYS Capsule and 227 intersection targets were obtained. Topological analysis showed that AKT had the highest connectivity degrees in the PPI network. Enrichment analysis revealed that the targets of BSYS in the treatment of CNSD were the PI3K-Akt and MAPK signaling pathway, among other pathways. GO analysis results showed that the targets were associated with various biological processes, including apoptosis, reactive oxygen species metabolic process, and response to oxidative stress, among others.     Abstract Remyelination is a refractory feature of demyelinating diseases such as multiple sclerosis (MS). Studies have shown that promoting oligodendrocyte precursor cell (OPC) differentiation, which cannot be achieved by currently available therapeutic agents, is the key to enhancing remyelination. Bu Shen Yi Sui capsule (BSYSC) is a traditional Chinese herbal medicine over many years of clinical practice. We have found that BSYSC can effectively treat MS. In this study, the effects of BSYSC in promoting OPCs differentiation and remyelination were assessed using an experimental autoimmune encephalomyelitis (EAE) model in vivo and cultured OPCs in vitro.     Abstract The impact of human interference on the ecological environment has attracted a significant amount of attention. In this study, hemeroby index (HI) was constructed to quantify the degree of human disturbance, and the relationship between HI and landscape pattern index was explored in a newly metropolitan area. The main objectives of this study were to analyze the temporal and spatial characteristics of landscape pattern and human disturbance in the process of urbanization of county-level cities in China, and to explore the relationship between the landscape pattern index and human disturbance under different disturbance intensity. The conclusions showed that: (1) the degree of human interference in the new area is on the rise, with a slow increase from 1980 to 2010, but since 2010, human interference has increased significantly. The diffusion of human disturbance intensity has obvious spatial directivity, spreading from east to west. (2) The impact of human activities on landscape pattern is shown as increasing fragmentation and increasing number of landscape types. With the enhancement of human activities, the heterogeneity and fragmentation of landscape types in the region reach their highest points. With the continuous increase of human activities, on a certain scale, the landscape types will gradually tend to be the same, and the same type of landscape patches will become one piece and tend to be integrated. This phenomenon is particularly obvious at the patch type level. (3) There is an inflection point value between human disturbance and landscape pattern index in landscape or patch type. On both sides of the inflection point value, the landscape pattern parameters and human disturbance have obvious opposite trends. (4) In the low interference range (1 ≤ HI < 4), with the increase of human interference, more heterogeneous structures (shown in the increase of SHDI) are brought to the landscape, there is more landscape fragmentation (shown in the decrease of LPI, CONTRACT, and AI), and the patch shape and landscape structure tend to be complex (shown in the increase of LSI, SQP, ED, and other indexes); in the high interference range (4 ≤ HI ≤ 7), due to human intervention, the increase of LPI, contract, and AI indicates that intense human activities turn the landscape into a broken and scattered structure and tend to be consistent and homogeneous. LSI, SQP, and ED were negatively correlated with HI at this stage, indicating that with the enhancement of human activities, the complexity of landscape shape decreased.

    Abstract Remyelination is a refractory feature of demyelinating diseases such as multiple sclerosis (MS). Studies have shown that promoting oligodendrocyte precursor cell (OPC) differentiation, which cannot be achieved by currently available therapeutic agents, is the key to enhancing remyelination. Bu Shen Yi Sui capsule (BSYSC) is a traditional Chinese herbal medicine over many years of clinical practice. We have found that BSYSC can effectively treat MS. In this study, the effects of BSYSC in promoting OPCs differentiation and remyelination were assessed using an experimental autoimmune encephalomyelitis (EAE) model in vivo and cultured OPCs in vitro. The results showed that BSYSC reduced clinical function scores and increased neuroprotection. The expression of platelet-derived growth factor receptor α (PDGFR-α) was decreased and the level of 2′,3′-cyclic nucleotide 3′-phosphodiesterase (CNPase) was increased in the brains and spinal cords of mice as well as in OPCs after treatment with BSYSC. We further found that BSYSC elevated the expression of miR-219 or miR-338 in the serum exosomes of mice with EAE, thereby suppressing the expression of Sox6, Lingo1, and Hes5, which negatively regulate OPCs differentiation. Therefore, serum exosomes of BSYSC-treated mice (exos-BSYSC) were extracted and administered to OPCs in which miR-219 or miR-338 expression was knocked down by adenovirus, and the results showed that Sox6, Lingo1, and Hes5 expression was downregulated, MBP expression was upregulated, OPCs differentiation was increased, and the ability of OPCs to wrap around neuronal axons was improved. In conclusion, BSYSC may exert clinically relevant effects by regulating microRNA (miR) levels in exosomes and thus promoting the differentiation and maturation of OPCs.

    Abstract Background: Bu Shen Yi Sui capsule (BSYS) is a traditional Chinese medicine prescription that has shown antineuroinflammatory and neuroprotective effects in treating multiple sclerosis (MS) and its animal model of experimental autoimmune encephalomyelitis (EAE). Microglia play an important role in neuroinflammation. The M1 phenotype of microglia is involved in the proinflammatory process of the disease, while the M2 phenotype plays an anti-inflammatory role. Promoting the polarization of microglia to M2 in MS/EAE is a promising therapeutic strategy. This study is aimed at exploring the effects of BSYS on microglial polarization in mice with EAE.

    Abstract Electrochemically active bacteria (EAB) exhibit promising prospects for space exploration and life support systems. However, the effects of the space environment on EAB are unclear. In this study, the effects of simulated microgravity on the current generation of mixed-culture EAB were illustrated, and the underlying mechanism was elucidated. The results demonstrated that the electrochemical activity of mixed-culture EAB was enhanced, which was mainly due to the enrichment of Geobacter and the increase in EAB biomass. Additionally, the genes and proteins of the biofilm changed obviously under simulated microgravity conditions, including: I) genes related to signal transfer, II) genes related to cell wall synthesis, and III) genes related to riboflavin synthesis. This study first revealed the enrichment in EAB abundance, the increase in EAB biomass, and the promotion of current generation under simulated microgravity.

    Abstract The burden of incidence rate and mortality of cancer is increasing rapidly, and the development of precise intervention measures for cancer detection and treatment will help reduce the burden and pain of cancer. At present, the sensitivity and specificity of tumor markers such as CEA and CA-125 used clinically are low, while PET, SPECT, and other imaging diagnoses with high sensitivity possess shortcomings, including long durations to obtain formal reports and the inability to identify the molecular pathological type of cancer. Cancer surgery is limited by stage and easy to recur. Radiotherapy and chemotherapy often cause damage to normal tissues, leading to evident side effects. Aptamers can selectively and exclusively bind to biomarkers and have, therefore, gained attention as ligands to be targeted for cancer detection and treatment. Gold nanoparticles (AuNPs) are considered as promising nano carriers for cancer diagnosis and treatment due to their strong light scattering characteristics, effective biocompatibility, and easy surface modification with targeted agents. The aptamer-gold nanoparticles targeting delivery system developed herein can combine the advantages of aptamers and gold nanoparticles, and shows excellent targeting, high specificity, low immunogenicity, minor side effects, etc., which builds a bridge for cancer markers to be used in early and efficient diagnosis and precise treatment. In this review, we summarize the latest progress in the application of aptamer-modified gold nanoparticles in cancer targeted diagnosis and delivery of therapeutic agents to cancer cells and emphasize the prospects and challenges of transforming these studies into clinical applications.

    Abstract This study examines the relationship and risk spillover between Bitcoin, crude oil, and six traditional markets (the US stock, Chinese stock, gold, bond, currency, and real estate markets) from 2019 to 2020, during which the coronavirus disease 2019 (COVID-19) outbreak occurred as well. We first discuss the static relationship between Bitcoin and these markets using a quantile-on-quantile model and examine the dynamic relationship using a time-varying copula model. A conditional value-at-risk model is subsequently used to estimate the risk spillover between the markets studied. The empirical results reveal that the relationship between these markets is always time-varying, and the COVID-19 outbreak has revealed such changes in the relationship between Bitcoin and other traditional financial markets. The risk of all single markets has enhanced because of the pandemic. Further, the risk spillover of these markets has also changed dramatically since the COVID-19 outbreak during which the Bitcoin market has played an important role and exerted a significant impact on the crude oil market, and the four other markets (US stock, gold, Chinese stock, and real estate markets). Overall, our findings indicate that investors and policymakers need to be made aware of the risk spillover between Bitcoin, crude oil, and other traditional markets and that flexible hedge strategies and policies should be implemented in response to the challenges and economic recession observed following the COVID-19 outbreak.
