    Abstract Currently, it is acknowledged that gout is caused by uric acid (UA). However, some studies have revealed no correlation between gout and UA levels, and growing evidence suggests that 2,8-dihydroxyadenine (2,8-DHA), whose structural formula is similar to UA but is less soluble, may induce gout. Hence, we hypothesized that uroliths from hyperuricemia (HUA) patients, which is closely associated with gout, may contain 2,8-DHA. In this study, 2,8-DHA in uroliths and serum of HUA patients were determined using HPLC. Moreover, bioinformatics was used to investigate the pathogenic mechanisms of 2,8-DHA nephropathy. Subsequently, a mouse model of 2,8-DHA nephropathy established by the gavage administration of adenine, as well as a model of injured HK-2 cells induced by 2,8-DHA were used to explore the pathogenesis of 2,8-DHA nephropathy. Interestingly, 2,8-DHA could readily deposit in the cortex of the renal tubules, and was found in the majority of these HUA patients. Additionally, the differentially expressed genes between 2,8-DHA nephropathy mice and control mice were found to be involved in inflammatory reactions. Importantly, CCL2 and IL-1β genes had the maximum degree, closeness, and betweenness centrality scores. The expressions of CCL2 and IL-1β genes were significantly increased in the serum of 24 HUA patients with uroliths, indicating that they may be significant factors for 2,8-DHA nephropathy. Further analysis illustrated that oxidative damage and inflammation were the crucial processes of 2,8-DHA renal injury, and CCL2 and IL-1β genes were verified to be essential biomarkers for 2,8-DHA nephropathy. These findings revealed further insights into 2,8-DHA nephropathy, and provided new ideas for its diagnosis and treatment.

    Abstract Polygonatum cyrtonema Hua is a traditional Chinese medicinal plant acclaimed for its therapeutic potential in diabetes and various chronic diseases. Its rhizomes are the main functional parts rich in secondary metabolites, such as flavonoids and saponins. But their quality varies by region, posing challenges for industrial and medicinal application of P. cyrtonema. In this study, 482 metabolites were identified in P. cyrtonema rhizome from Qingyuan and Xiushui counties. Cluster analysis showed that samples between these two regions had distinct secondary metabolite profiles. Machine learning methods, specifically support vector machine-recursive feature elimination and random forest, were utilized to further identify metabolite markers including flavonoids, phenolic acids, and lignans. Comparative transcriptomics and weighted gene co-expression analysis were performed to uncover potential candidate genes including CHI, UGT1, and PcOMT10/11/12/13 associated with these compounds. Functional assays using tobacco transient expression system revealed that PcOMT10/11/12/13 indeed impacted metabolic fluxes of the phenylpropanoid pathway and phenylpropanoid-related metabolites such as chrysoeriol-6,8-di-C-glucoside, syringaresinol-4'-O-glucopyranosid, and 1-O-Sinapoyl-D-glucose. These findings identified metabolite markers between these two regions and provided valuable genetic insights for engineering the biosynthesis of these compounds.

    Abstract In the present study, we investigated the intervention effects of a purified Polygonatum cyrtonema polysaccharide (PCP) on high-fat diet (HFD)-induced atherosclerosis in male and female LDLr⁻/⁻ mice. Results showed that HFD caused severe dyslipidemia, atherosclerotic lesions, oxidative damages and inflammation in male and female mice, and these effects seemed to be more pronounced in males than in females. However, the above variations could be dose-dependently reversed by PCP treatment, and the intervention effects on males were greater than those on females. Nuclear factor kappa-B (NF-κB), mitogen-activated protein kinase (MAPKs) and protein kinase B (Akt) are 3 pivotal signaling pathways mediating the development of atherosclerosis. Consistently, PCP was also found to significantly decrease the phosphorylation of p65, p38, extracellular-regulated kinase 1/2 (ERK1/2) and Akt, and increase the protein expression of inhibitor of NF-κB (IκB) in the aortas of male and female mice induced by HFD. Taken together, these findings indicated that PCP could be effective for the prevention of atherosclerosis, and the intervention effect of PCP on male mice was more obvious than that of female mice.

    Abstract In our aging society, dysphagia and malnutrition are growing concerns, necessitating intervention. Liquid nutrition support offers a practical solution for traditional dietary issues, but it raises a key issue: the potential for post-meal glucose spikes impacting efficacy. This study examined the effects of supplementation of Polygonatum cyrtonema Hua polysaccharide (PCP), konjac glucomannan (KGM) and their combination on acute phase postprandial glycemic response and long-term glucose metabolism in T2DM mice on a complete nutritional liquid diet. KGM was more effective in reducing postprandial glucose response, while PCP was more prominent in ameliorating long-term glucose metabolism. The KGM-PCP combination demonstrated superior outcomes in fasting blood glucose, insulin, and glucose homeostasis. PCP and KGM also influenced the composition and abundance of the gut microbiome, with the H-PCP group showing optimal performance. Moreover, the KGM-PCP combination improved body weight, lipid homeostasis, and liver health the most. PCP potentially regulates glycemia through metabolic pathways, while KGM improves glycemic metabolism by reducing postprandial glucose levels in response to viscous intestinal contents. This research identifies the structure, viscosity properties, and hypoglycemic effects of KGM and PCP in complete nutritional liquid diet fed T2DM mice, enabling their strategic utilization as hypoglycemic components in nutritional administration and glycemic regulation.

    Abstract Ethnopharmacological relevance: Controversy persists regarding the treatment of immunoglobulin A nephropathy (IgAN), thereby highlighting the demand for safer more effective therapeutic drugs. Although supplementary treatment using Yi-Shen-Hua-Shi (YSHS) granules has distinct advantages with respect to improving renal function in IgAN, a lack of clarity regarding the underlying mechanisms limits their clinical application.
    Aim of the study: In this study, we aimed to elucidate the therapeutic mechanisms underlying the efficacy of YSHS granules in the treatment of IgAN.
    Materials and methods: A rat model of IgAN was established based on lipopolysaccharide, carbon tetrachloride, and bovine serum albumin induction. In order to evaluate the effects of YSHS granules, we performed a range of techniques, including immunofluorescence assays, hematoxylin and eosin staining, and flow cytometry, to assess inflammation, immunity, and other relevant factors. Direct data-independent acquisition-mass spectrometry (DIA-MS) analysis and parallel reaction monitoring (PRM) were used for functional characterization and quantitative validation of differentially expressed proteins (DEPs), and Western blot analysis is used to identify downstream proteins associated with DEPs.
    Results: Compared with the model group, the levels of proteinuria, urine red blood cells, serum creatinine, blood urea nitrogen, low-density lipoprotein-cholesterol, triglycerides, and pathological kidney damage were reduced in the YSHS group. A high dose of YSHS granules was found to raise the levels of CD8 T cells and reduce the CD4/CD8 ratio in the peripheral serum. To examine the mechanisms underlying the therapeutic effects YSHS granules, we performed direct DIA-MS analysis to identify proteins that were differentially expressed among the model, YSHS, and control groups. A total of 29 proteins were identified as being commonly expressed in all three groups. Further KEGG and protein-protein interaction (PPI) network analysis revealed that YSHS granules can contribute to the regulation of N-glycosylation-associated proteins, such as ALG3 and STT3A, in rats with IgAN. Detected changes in the expression of ALG3 and STT3A were consistent with the PRM results. We also established that the administration of YSHS granules can contribute to regulation of the ALG3-associated PPAR-γ/NF-κB signaling pathway.
    Conclusions: Our findings in this study provide evidence to indicate the efficacy of YSHS granules in the treatment of IgAN, the putative underlying mechanisms of which involve the modulation of N-glycosylation, mediated via the PPAR-γ/NF-κB pathway.

    Abstract Qing-Jin-Hua-Tan-Decoction (QJHTD), a classic famous Chinese ancient prescription, has been used for treatment of pulmonary diseases since Ming Dynasty. A total of 22 prototype compounds of QJHTD absorbed into rat blood were chosen as candidates for the pharmacological network analysis and molecular docking. The targets from the intersection of compound target and ALI disease targets were used for GO and KEGG enrichment analyses. Molecular docking was adopted to further verify the interactions between 22 components and the top 20 targets with higher degree values in the component-target-pathway network.

    Abstract Polygonatum cyrtonema Hua, also known as Huangjing, is an important traditional Chinese herb for treating diabetes and asthma. This plant is commercially planted (approximately 700 ha) in the highland of Anhui Province in China (Liu et al. 2016). In June 2019, brown spots on the leaves of P. cyrtonema Hua were frequently observed in the field in Jinzhai County, Anhui Province. Disease incidence varied from 20 to 30% in a survey site (3 ha) of approximately 25,000 plants. Initially circular or elliptical, reddish colored spots with well-defined brown margin were observed on infected leaves. With time, the spots expanded to large elliptical or shuttle-shaped, pale to dark brown lesions within 10-15 days. To identify the pathogen, symptomatic tissues from the lesion margin were excised from fifteen infected leaves, surface sterilized with 2% NaClO for 2 min, 70% ethanol for 30 s, rinsed three times with sterile water, air dried, plated on potato dextrose agar (PDA) medium supplemented with cefotaxime sodium salt (150 μg/ml), and incubated at 26°C for 3 to 4 days in the dark. Eight morphologically similar fungal isolates were obtained by single-sporing. On PDA, colonies were initially white, and turned pale gray with age. On the reverse side, colonies were initially pale brown and later turned dark black. Conidia were hyaline, curved or slightly curved, aseptate, with a truncate base and acute apex, and measured 14.38 to 20.57 × 2.03 to 5.28 μm (n = 50). The appressoria were solitary, dark brown, ovoid or irregularly shaped, with diameters from 5.61 to 12.43 μm (n = 50). These morphological characteristics are consistent with the description of a Colletotrichum species (Damm et al. 2009). For molecular identification, the primers ITS4/ITS5, T1/T2 and GDF/GDR were used to amplify the partial region of rDNA-ITS, β-tubulin (TUB2) and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) from fresh mycelia of all eight isolates, respectively. The resulting sequences of ITS, TUB2 and GAPDH were identical with each other, and sequences from strain PCCS-3 were submitted to Genbank. BlastN revealed that ITS (MN305792), TUB2 (MN307740) and GAPDH (MN307739) shared 99.8%, 100%, 98.8% similarity with the published ITS, TUB2 and GADPH sequences from the ex-epitype culture of C. spaethianum CBS 167.49 (Damm et al. 2009), respectively. To prove Koch's postulates, a pathogenicity test was conducted by spraying 3 ml of conidial suspension of each the eight isolates (105 conidia/ml) on three-month-old healthy P. cyrtonema seedlings (3 seedlings per isolate). As a control, three seedlings were sprayed with 3 ml of distilled water. All inoculated plants were maintained in a moist chamber (RH>90%) at 26°C, with an 8-h photoperiod under fluorescent light. The disease assays were repeated thrice. Brown spots identical to those observed in field were noted 7 to 10 days after inoculation, whereas control plants remained symptomless. The same pathogen was reisolated and confirmed as C. spaethianum by sequencing of ITS, GAPDH and Tub2, fulfilling Koch's postulates. Presently, C. spaethianum causing leaf spot on Polygonatum odoratum was reported from Jilin Province in China (Liu et al. 2020). To our knowledge, this is the first report of C. spaethianum infecting P. cyrtonema in Anhui Province, China. Our result expanded the geographic location of C. spaethianum as the pathogen on plants belonging to Asparagaceae family in China, and effective control strategies need to be considered to minimize the losses.

    Abstract Glyoxal and methylglyoxal are important volatile organic compounds in the atmosphere. The gas–particle partitioning of these carbonyl compounds makes significant contributions to O 3 formation. In this study, both the gas- and particle-phase glyoxal and methylglyoxal concentrations at the foot and top of Mount Hua were determined simultaneously. The results showed that the gaseous-phase glyoxal and methylglyoxal concentrations at the top were higher than those at the foot of the mountain. However, the concentrations for the particle phase showed the opposite trend. The average theoretical values of the gas–particle partitioning coefficients of the glyoxal and methylglyoxal concentrations (4.57 × 10 −10 and 9.63 × 10 −10 m 3 μg −1 , respectively) were lower than the observed values (3.79 × 10 −3 and 6.79 × 10 −3 m 3 μg −1 , respectively). The effective Henry’s law constants (eff.K H ) of the glyoxal and methylglyoxal were in the order of 10 8 to 10 9 mol/kgH2O/atm, and they were lower at the foot than they were at the top. The particle/gas ratios (P/G ratios) of the glyoxal and methylglyoxal were 0.039 and 0.055, respectively, indicating more glyoxal and methylglyoxal existed in the gas phase. The factors influencing the partitioning coefficients of the glyoxal and methylglyoxal were positively correlated with the relative humidity (RH) and negatively correlated with the PM 2.5 value. Moreover, the partitioning coefficient of the glyoxal and methylglyoxal was more significant at the top than at the foot of Mount Hua.
