    Abstract Purpose: Traditional treatment regimens for advanced prostate cancer, especially castration-resistant prostate cancer, result in low survival times with severe side effects.     Methods: The anti-cancer effect of VC and/or SAS on prostate cancer cells was assessed using WST-8, colony formation and annexin V-FITC/PI FACS assays. Changes in cellular ROS and GSH levels were determined to verify our hypothesis. Finally, BALB/c nude mice bearing prostate cancer xenografts were used to assess the anti-cancer effects of single or combined VC and SAS therapies.
    Results: We found that SAS could potentiate the short- and long-term cytotoxicity of VC in prostate cancer cells. We also found that the synergistic effect of SAS and VC led to significant cellular GSH depletion, resulting in increased ROS accumulation. This synergistic effect could be reversed by the antioxidant N-acetyl-L-cysteine (NAC). The synergistic effect of SAS and VC was also noted in prostate cancer xenografts and correlated with immunohistochemistry results.
    Conclusions: Our results strongly indicate that SAS, a relatively non-toxic drug that targets cystine transporters, in combination with VC may be superior to their single applications in the treatment of prostate cancer.

    Abstract Sequential latent variable models (SLVMs) are essential tools in statistics and machine learning, with applications ranging from healthcare to neuroscience. As their flexibility increases, analytic inference and model learning can become challenging, necessitating approximate methods. Here we introduce neural adaptive smoothing via twisting (NAS-X), a method that extends reweighted wake-sleep (RWS) to the sequential setting by using smoothing sequential Monte Carlo (SMC) to estimate intractable posterior expectations. Combining RWS and smoothing SMC allows NAS-X to provide low-bias and low-variance gradient estimates, and fit both discrete and continuous latent variable models. We illustrate the theoretical advantages of NAS-X over previous methods and explore these advantages empirically in a variety of tasks, including a challenging application to mechanistic models of neuronal dynamics. These experiments show that NAS-X substantially outperforms previous VI- and RWS-based methods in inference and model learning, achieving lower parameter error and tighter likelihood bounds.

    Abstract As more and more pre-trained language models adopt on-cloud deployment, the privacy issues grow quickly, mainly for the exposure of plain-text user data (e.g., search history, medical record, bank account). Privacy-preserving inference of transformer models is on the demand of cloud service users. To protect privacy, it is an attractive choice to compute only with ciphertext in homomorphic encryption (HE). However, enabling pre-trained models inference on ciphertext data is difficult due to the complex computations in transformer blocks, which are not supported by current HE tools yet. In this work, we introduce $\textit{THE-X}$, an approximation approach for transformers, which enables privacy-preserving inference of pre-trained models developed by popular frameworks. $\textit{THE-X}$ proposes a workflow to deal with complex computation in transformer networks, including all the non-polynomial functions like GELU, softmax, and LayerNorm. Experiments reveal our proposed $\textit{THE-X}$ can enable transformer inference on encrypted data for different downstream tasks, all with negligible performance drop but enjoying the theory-guaranteed privacy-preserving advantage.

    Abstract We present the training recipe and results of scaling up PaLI-X, a multilingual vision and language model, both in terms of size of the components and the breadth of its training task mixture. Our model achieves new levels of performance on a wide-range of varied and complex tasks, including multiple image-based captioning and question-answering tasks, image-based document understanding and few-shot (in-context) learning, as well as object detection, video question answering, and video captioning. PaLI-X advances the state-of-the-art on most vision-and-language benchmarks considered (25+ of them). Finally, we observe emerging capabilities, such as complex counting and multilingual object detection, tasks that are not explicitly in the training mix.

    Abstract In the progression of X-ray-based radiotherapy for the treatment of cancer, the incorporation of nanoparticles (NPs) has a transformative impact. This study investigates the potential of NPs, particularly those comprised of high atomic number elements, as radiosensitizers. This aims to optimize localized radiation doses within tumors, thereby maximizing therapeutic efficacy while preserving surrounding tissues. The multifaceted applications of NPs in radiotherapy encompass collaborative interactions with chemotherapeutic, immunotherapeutic, and targeted pharmaceuticals, along with contributions to photodynamic/photothermal therapy, imaging enhancement, and the integration of artificial intelligence technology. Despite promising preclinical outcomes, the paper acknowledges challenges in the clinical translation of these findings. The conclusion maintains an optimistic stance, emphasizing ongoing trials and technological advancements that bolster personalized treatment approaches.     Abstract Liver X receptors α and β are members of the nuclear receptor family, which comprise a flexible N-terminal domain, a DNA binding domain, a hinge linker, and a ligand binding domain. Liver X receptors are important regulators of cholesterol and lipid homeostasis by controlling the transcription of numerous genes. Key to their transcriptional role is synergetic interaction among the domains. DNA binding domain binds on DNA; ligand binding domain is a crucial switch to control the transcription activity through conformational change caused by ligand binding. The Liver X receptors form heterodimers with retinoid X receptor and then the liganded heterodimer may recruit other necessary transcription components to form an active transcription complex.
