    Abstract The Liang guang Small-spotted pig is a well-known Chinese indigenous pig that is valued for its exceptional meat quality. However, the Liang guang Small-spotted pig has a lower semen storage capacity, shorter storage time and worse semen quality compared to Duroc. Pig sperm used for artificial insemination (AI) loses part of vitality and quality when being stored in commercial solutions. Serious vitality losses and short shelf life of the semen are particularly prominent in Liang guang Small-spotted pig. In this study, the metabolites in seminal plasma and spermatozoa of Duroc and Liang guang Small-spotted pigs were identified using UHPLC-Q-TOF/MS technology. The findings indicated forty distinct metabolites concentrating on energy metabolic substrates and antioxidant capacity in Liang guang Small-spotted pig and Duroc seminal plasma, including D-Fructose, succinate, 2-dehydro-3-deoxy-d-gluconate, alanine betaine, citrate, carnitine, acetylcarnitine and so on. Seventeen different metabolites were explored, with a focus on glycerophospholipid metabolism in Liang guang Small-spotted pig and Duroc spermatozoa, primarily including glycerol 3-phosphate, acetylcarnitine, phosphatidylcholine (PC) 16:0/16:0, palmitoyl sphingomyelin, acetylcholine, choline, glycerophosphocholine, betaine, L-carnitine, creatinine and others. This study reveals the metabolite profile of spermatozoa and seminal plasma among different pig breeds and might be valuable for understanding the mechanisms that lead to sperm storage capacity. Metabolites involved in energy metabolism, antioxidant capacity and glycerophospholipid metabolism might be key to the poor sperm storage capacity in Liang guang Small-spotted pig.

    Abstract Background: Dengue virus (DENV) is a major public health threat. However, there are no specific therapeutic drugs for DENV. Many Chinese heat-cleaning formulas, such as Liang-Ge-San (LGS), have been frequently used in the virus-induced diseases. The antiviral effect of LGS has not been reported yet.
    Purpose: In this study, the effect of LGS on the inhibition of dengue virus serotype 2 (DENV-2) was investigated and the relevant mechanism was explored.
    Methods: High-performance liquid chromatography was applied to analyze the chemical characterization of LGS. The in vitro antiviral activities of LGS against DENV-2 were evaluated by time-of-drug-addition assay. The binding of heat shock protein 70 (Hsp70) and envelope (E) protein or caveolin1 (Cav1) were analyzed by immunofluorescence and immunoprecipitation assays. Then the role of Cav1 in the anti-DENV-2 effects of LGS was further examined. DENV-2 infected Institute of Cancer Research suckling mice (n = 10) and AG129 mice (n = 8) were used to examine the protective effects of LGS.
    Results: It was found that geniposide, liquiritin, forsythenside A, forsythin, baicalin, baicalein, rhein, and emodin maybe the characteristic components of LGS. LGS inhibited the early stage of DENV-2 infection, decreased the expression levels of viral E and non-structural protein 1 (NS1) proteins. LGS also reduced E protein and Hsp70 binding and attenuated the translocation of Hsp70 from cytoplasm to the cell membrane. Moreover, LGS decreased the binding of Hsp70 to Cav1. Further study showed that the overexpression of Cav1 reversed LGS-mediated E protein and Hsp70 inhibition in the plasma membrane. In the in vivo study, LGS was highly effective in prolonging the survival time, reducing viral loads.
    Conclusion: This work demonstrates for the first time that LGS exerts anti-DENV-2 activity in vitro and in vivo. LGS decreases DENV-2-stimulated cytoplasmic Hsp70 translocation into the plasma membrane by Cav1 inhibition, thereby inhibiting the early stage of virus infection. These findings indicate that LGS may be a candidate for the treatment of DENV.

    Abstract Caridina stellata sp. nov. is described from streams in Guangxi, south-western China. The new species clearly belongs to “Caridina serrata group” of the genus and shows a morphological similarity with C. cantonensis Yu, 1938, C. serrata Stimpson, 1860 and C. pacbo Do et al. 2020. Caridina stellata is distinguished from congeners, based on differences in its male first pleopod and appendix masculina morphology, along with COI and 16S rRNA molecular evidence. The first pleopod endopod in male is rectangle, about 0.70 × length of exopod, about 3.7–3.9 × as long as proximally wide, inner margin concave, bearing nearly equal spine setae, outer margin bearing nearly equal long and dense spine setae; appendix interna well developed, arising from distal 1/5 of endopod, reaching to end of endopod, with cincinuli distally. The new species displays a unique and brightly coloured pattern and, therefore, can be easily recognised in the field. Liang & Zhou, 1993 described C. cavernicola from the Lenggu Cave, Du’an County, Guangxi. However, the description was based exclusively on two females. We have collected specimens of both sexes near the type locality and describe herein the previously unknown male and present morphological data on females. Data on the habitat, ecology and levels of threat of the two species are provided and suggest that they should be categorised as vulnerable (VU) under the current IUCN Criteria.

    Abstract We have previously shown that Liang-Yan-Yi-Zhen-San (LYYZS), an ancient Chinese herbal formula, can promote the browning of white adipose tissue. In this study, we sought to determine which active ingredients of LYYZS mediated its effects on the browning of white adipose tissue. Employing ultra-high performance liquid chromatography-Q-Exactive HF mass spectrometry, a total of 52 LYYZS ingredients were identified. On this basis, 1,560 ingredient-related targets of LYYZS were screened using the HERB databases. Meanwhile, RNA sequencing analysis of the inguinal white adipose tissue of mice produced a total of 3148 genes that were significantly differentially expressed following LYYZS treatment and differentially expressed genes regarded as browning-related targets. Through the network pharmacological analysis, a total of 136 intersection targets were obtained and an ingredient-target-pathway network was established. According to network pharmacology analysis, 10 ingredients containing trans-cinnamaldehyde, genistein, daidzein, calycosin, arginine, coumarin, oleic acid, isoleucine, palmitic acid and tyrosine were regarded as active ingredients of browning of white adipose tissue. Integrated evaluation using chemical analysis, transcriptomics and network pharmacology provides an efficient strategy for discovering the active ingredients involved in how LYYZS promotes the browning of white adipose tissue.

    Abstract The commercial value of green raisins produced by the shade-air drying in Liang-fang was 2–3 times as high as that of yellow-brown raisins. This study systematically evaluated the effect of the temperature, humidity, and light of sun drying and shade-air drying on raisins. The sugar, acids, texture and color parament of raisins were compared with different drying conditions. The results indicated that shade-air drying can offer a dark and conducive light environment with the maximum temperature below 40 °C, and a lower humidity by segregating rainwater during the day as well. Then, the contents of acids of raisins in sun drying were lower compared with the shade-air drying and had a lower hardness and a* values of raisins. In summary, low light and ultraviolet intensity, suitable temperature and appropriate humidity environments were the basal and key factors for the formation of the green raisins. The present study not only revealed the crucial factors during shade-air drying in Liang-fang in Turpan, but also provided a scientific basis for improving the Liang-fang conditions and producing green raisins in other regions.
