Abstract To assess the effect of population genetic polymorphism on forensic research, we investigated the genetic polymorphisms of Chinese Kyrgyz group (n = 98) and evaluated forensic application values in Chinese Kyrgyz group and other 26 reference populations at 90 autosomal SNPs, and then combined with 34 SNPs and 37 STRs on Y chromosome to reveal the genetic background of Kyrgyz group in autosomal and Y-chromosomal inheritances, respectively. The 90 autosomal SNPs and 34 Y-chromosomal SNPs were sequenced base on next generation sequencing technology, and 37 Y-chromosomal STRs were analyzed by capillary electrophoresis platform. The results showed that cumulative power of discrimination and cumulative power of exclusion of 90 autosomal SNPs in the panel met the application need of forensic genetics in Kyrgyz group. The forensic effectivenesses of the panel were high in all 27 populations, although there were genetic differences among these populations. The forensic effectiveness of the panel was relatively higher in the European populations, but relatively lower in the African populations. The population genetic results indicated that the Kyrgyz group had the relatively closer genetic relationships with the reference East Asian populations at autosomal SNPs, and there were gene exchanges between the Kyrgyz group and East Asian, European populations based on the analytical results of autosomal SNPs, Y-chromosomal SNPs and STRs.

Abstract Drought and salt stresses impose major constraints on soybean production worldwide. However, improving agronomically valuable soybean traits under drought conditions can be challenging due to trait complexity and multiple factors that influence yield. Here, we identified a nuclear factor Y C subunit (NF-YC) family transcription factor member, GmNF-YC14, which formed a heterotrimer with GmNF-YA16 and GmNF-YB2 to activate the GmPYR1-mediated abscisic acid (ABA) signalling pathway to regulate stress tolerance in soybean. Notably, we found that CRISPR/Cas9-generated GmNF-YC14 knockout mutants were more sensitive to drought than wild-type soybean plants. Furthermore, field trials showed that overexpression of GmNF-YC14 or GmPYR1 could increase yield per plant, grain plumpness, and stem base circumference, thus indicating improved adaptation of soybean plants to drought conditions. Taken together, our findings expand the known functional scope of the NF-Y transcription factor functions and raise important questions about the integration of ABA signalling pathways in plants. Moreover, GmNF-YC14 and GmPYR1 have potential for application in the improvement of drought tolerance in soybean plants.

Abstract Olfactory dysfunction significantly impedes the life quality of patients. Neuropeptide Y (NPY) is not only a neurotrophic factor in the rodent olfactory system but also an orexigenic peptide that regulates feeding behavior. NPY increases the olfactory receptor neurons (ORNs) responsivity during starvation; however, whether NPY can promote differentiation of human ORNs remains unexplored. This study investigates the effect of NPY on the differentiation of human olfactory neuroepithelial cells in vitro. Human olfactory neuroepithelium explants were cultured on tissue culture polystyrene dishes for 21 days. Then, cells were cultured with or without NPY at the concentration of 0.5 ng/mℓ for 7 days. The effects of treatment were assessed by phase contrast microscopy, immunocytochemistry and western blot analysis. The further mechanism was evaluated with NPY Y1 receptor-selected antagonist BIBP3226. NPY-treated olfactory neuroepithelial cells exhibited thin bipolar shape, low circularity, low spread area, and long processes. The expression levels of Ascl1, βIII tubulin, GAP43 and OMP were significantly higher in NPY-treated cells than in controls (p < 0.05). NPY-treated olfactory neuroepithelial cells expressed more components of signal transduction apparatuses, G

Abstract Hydrotreating is an effective method for bio-crude upgrading. However, the high cost of catalysts and gaseous hydrogen consumption limits its application. In this research, oak wood hydrothermal liquefaction bio-crude upgrading was conducted by using Zn as heterogeneous hydrogen producer and low-cost Ni supported Y zeolite synthesized from rice husk as catalyst. The Y zeolite was obtained by two-step synthetic route, using different Tetramethylammonium hydroxide (TMAOH)/silica mole ratios of 0, 0.03 and 0.08. Synthesized Y zeolite showed better performance than commercial one due to the higher total acid sites (24–28% higher). Compared with template-free Y zeolite, the introduction of TMAOH template brings more mesopore structure to the synthesized zeolite, improving its catalytic activity. The yield of upgraded oil increased from 65.5% of blank upgrading test to 80.0% of the synthesized 10%Ni/HY (0.08), and the coke amount reduced from 14.3% to 3.9%. The H/C ratio increased, and the O/C ratio decreased, consequently the higher heating value (HHV) raised from 30.04 to 33.13 MJ/kg. The area percentages of hydrogenation products: cyclopentanones and hydrogenated lignin-derived compounds increased in the catalytic upgraded oil samples consuming furan derivatives and lignin-derived compounds. Deactivation of the synthesized catalyst was observed due to the zeolite structure collapse, Ni active site oxidation, coke deposition and presence of ZnO impurity. The one-pot catalytic hydrothermal treatment of oak wood was also conducted with interesting result, bio-crude yield of 39.0% and HHV of 32.61 MJ/kg, which greatly simplified the process by combining the bio-crude production and upgrading steps together.
