    Abstract Here we report a three-component auto-tandem reaction of 1,3-enyne-tethered carbonyls, organoboronic reagents, and suitable nucleophiles catalyzed by palladium, proceeding through consecutive intramolecular vinylogous addition, Suzuki coupling, and allylic alkylation. This process exhibited high chemo- and regioselectivity with 1,3,4-trifunctionalization of the 1,3-enyne motif, and a wide range of 2H-chromenes, 1,2-dihydroquinolines, benzo[b]oxepines, 1,7-annulated indoles, and other frameworks were efficiently constructed in fair to good yields and E/Z selectivity.

    Abstract TRIzol is a monophasic solution of phenol and guanidine isothiocyanate used for the extraction of RNA, DNA and proteins from tissues or cells. However, few studies have described its application to DNA extraction due to its time-consuming procedure. We present a TRIzol-modified method of extracting DNA from tissues using the TRIzol reagent and a silica column, which requires only one-third of the time required for the classic extraction procedure. Spectrophotometric analysis showed that the 260/280 and 260/230 nm optical density ratio of the DNA extracted using the TRIzol-modified method is ideal and equal to that obtained by the classic method and commercial DNAiso methods. The DNA extracted by the TRIzol-modified method had the same performance in a restriction enzyme digestion and quantitative PCR as that extracted using the classic method. Using the TRIzol-modified method saves time, simplifies the DNA extraction procedure, and facilitates various molecular biology assays.

    Abstract A cascade assembly between isatin-derived Morita–Baylis–Hillman carbonates and o-hydroxybenzylideneacetones has been developed under the relay catalysis of Pd(PPh₃)₄ and DBU, affording a spectrum of 1,2,3,4-tetrahydrodibenzo[b,d]furan architectures incorporating a spirooxindole motif in moderate to good yields with excellent diastereoselectivity. The fused indole analogues were similarly furnished by employing the benzylideneacetones having an o-TsNH group.

    Abstract Background: Due to the complex histological type and anatomical structures, there has been considerable debate on the classification of adenocarcinoma of the esophagogastric junction (AEG), especially Siewert II AEG. Furthermore, neither the American Joint Committee on Cancer (AJCC) 7th tumor-node-metastasis (TNM) [esophageal adenocarcinoma (E) or gastric cancer (G)] nor the AJCC 8th TNM (E or G) accurately predicted the prognosis of patients with Siewert II AEG.
    Objective: This study aimed to investigate the factors influencing the survival and prognosis of patients with Siewert II AEG and establish a new and better prognostic predictive model.
    Design: A retrospective study.
    Methods: Patients with Siewert II AEG, retrieved from the Surveillance, Epidemiology, and End Results (SEER) databases, were assigned to the training set. Patients retrieved from a single tertiary medical center were assigned to the external validation set. Significant variables were selected using univariate and multivariate Cox regression analyses to construct the nomogram. Nomogram models were assessed using the concordance index (C-index), a calibration plot, decision curve analysis (DCA), and external validation.
    Results: Age, tumor grade, and size, as well as the T, N, and M stages, were included in the nomograms. For the SEER training set, the C-index of the nomogram was 0.683 (0.665-0.701). The C-index of the nomogram for the external validation set was 0.690 (0.653-0.727). The calibration curve showed good agreement between the nomogram estimations and actual observations in both the training and external validation sets. The DCA showed that the nomogram was clinically useful.
    Conclusion: The new predictive model showed significant accuracy in predicting the prognosis of Siewert II AEG.
