    Abstract The COVID-19 pandemic caused by the previously unknown SARS-CoV-2 Betacoronavirus made it extremely important to develop simple and safe cellular systems which allow manipulation of the viral genome and high-throughput screening of its potential inhibitors. In this review, we made an attempt at summarizing the currently existing data on genetic engineering systems used to study not only SARS-CoV-2, but also other viruses from the Coronaviridae family. In addition, the review covers the basic knowledge about the structure and the life cycle of coronaviruses.

Traditional classification is based on histological properties of tumors and distinguishes different grades of meningioma malignancy. However, knowledge about different molecular mechanisms of tumor provided new data on genetic features of meningiomas. The authors analyze current available data on the main driver mutations, new classifications based on molecular genetic characteristics and potential targets for therapy.

    Title translation New classification and approaches to the treatment of schwannomatosis.
    Abstract Schwannomatoses is a new classification unit for all the hereditary diseases caused by chromosome 22 damage followed by multiple benign neoplasms of the peripheral and central nervous system. Schwannomatosis occurs as a result of damage to different genes: NF2, SMARCB1, LZRT1, loss of heterozygosity of the long arm of chromosome 22. Nevertheless, clinical manifestations are similar. Molecular diagnostics not only confirms the diagnosis, but also predicts the course of disease. Thus, the most severe clinical manifestations are observed in patients with violation of semantic sequences and reading frame shift in exons 2-13 of the NF2 gene. A more favorable course with less number of tumors is observed in patients with somatic mosaicism. Stereotactic irradiation and surgery are the main treatment options for schwannomatosis. However, there is evidence of effective targeted therapy with bevacizumab (inhibitor of vascular endothelial growth factor). Bevacizumab is used in patients with bilateral vestibular schwannomas and high risk of hearing loss, as well as for intramedullary tumor growth control.

    Abstract Adipose tissue is the most convenient source of cellular material for regenerative medicine as it can be obtained in significant quantities via cosmetic liposuction, lipoaspiration of subcutaneous fat or by excision of fat deposits. Adipose tissue consists of adipocytes and cells, which are the part of the stromal-vascular fraction (SVF). Different cell populations can be isolated from SVF, among which the population of adipose tissue stem cells (adipose-derived stem cells, ADSC) is especially important for regenerative medicine. SVF can be obtained relatively easily from adipose tissue (adipose tissue is an alternative to bone marrow in terms of being a source of stem cells) and used to treat various pathologies. Recent studies show that SVF not only has a therapeutic effect similar to that of ADSC, but in some cases is even more effective. The article provides the analysis of the main methods of SVF obtainment, characteristics of SVF cellular composition, its potential for use in clinical medicine and its main advantages over other sources of cellular material, including­ ADSC cultured in vitro, for regenerative medicine.

    Abstract We present a novel process for generating synthetic datasets tailored to assess asset allocation methods and construct portfolios within the fixed income universe. Our approach begins by enhancing the CorrGAN model to generate synthetic correlation matrices. Subsequently, we propose an Encoder-Decoder model that samples additional data conditioned on a given correlation matrix. The resulting synthetic dataset facilitates in-depth analyses of asset allocation methods across diverse asset universes. Additionally, we provide a case study that exemplifies the use of the synthetic dataset to improve portfolios constructed within a simulation-based asset allocation process.

    Title translation Clinical and electrophysiological parameters of alcohol dependence and its comorbidity with affective disorders.
    Abstract Objective: To identify characteristics of and possible differences in clinical and electroencephalographic parameters in patients with alcohol dependence with- and without comorbid affective disorders.
    Materials and methods: Ninety-five patients, including 74 with an established diagnosis of alcohol dependence and 21 with alcohol dependence and affective disorders, were examined. Duration of alcohol dependence and affective disorder (years), number of hospitalizations and suicidal attempts were analyzed as anamnestic data. Hamilton's anxiety and depression scale (HDRS), the Alcohol Use Disorders Identification Test (AUDIT), the General clinical impression scale (CGI-s) were used as psychometric tools. The study of bioelectric activity of the brain was carried out using a 16-channel encephalograph. The background electroencephalogram was recorded, the values of absolute spectral power and coherence of theta, alpha and beta rhythms were analyzed.
    Results and conclusion: Patients with comorbidity of alcohol dependence and affective disorders have a significantly higher risk of suicidal behavior, a greater frequency of hospitalizations, higher levels of anxiety compared with patients without affective disorders. Electrophysiological data have shown that patients with comorbidity of alcohol dependence and affective disorders have higher values of spectral power of alpha-rhythm in all parts of the cerebral cortex (except for temporal lobes) and beta-rhythm in the occipital cortex, as well as an increase in interhemispheric coherence in all frequency ranges.

    Abstract Objective. To study neurophysiological correlates of inhibitory control to determine the features of inhibitionprocesses in alcohol dependence.Materials and methods. 77 patients with alcohol dependence were examined (42 men and 35 women) (F10.2 according to ICD-10). Patients were examined using a test to assess inhibitory control – Go/No – go. According to the task performance, patients were divided into two groups: group 1 – without inhibitory control impairments, group 2 – with impaired inhibitory control. During execution of test, electroencephalogram recordings were made according to the 10–20 system. The values of spectral power and coherence of θ-, α- and β-rhythms were analyzed. Statistical processing was carried out using nonparametric Mann–Whitney U-test and Wilcoxon W-test.Results. In patients with impaired inhibitory control, there was a decrease in the spectral power of the α-rhythm in the frontal cortex (p = 0.003), whereas in patients without inhibitory control disorders – in the Central cortex (p = 0.036). Patients with impaired inhibitory control responded by increasing β-power to cognitive stimulus in the occipital (p = 0.014), left temporal (p = 0.009) and right temporal (p = 0.008) cortex, while patients without inhibitory control disorders showed an increase in β-power only in the occipital (p = 0.007) and left temporal (p = 0.002) cortex. According to coherence data, patients with impaired inhibitory control have greater involvement of brain structures during the “Go/No – go” test in all frequency ranges.Conclusions. Patients with and without impaired inhibitory control have regional differences in changes in brain bioelectric activity during the “Go/No – go” test.
