    Abstract Diethylpyrocarbonate (DEPC) covalent labeling-mass spectrometry (CL-MS) has been extensively utilized to study protein structure and interactions owing to its ease of use, commercial availability, and broad labeling of nucleophilic residues. During typical CL-MS experiments with DEPC, the extent of labeling is kept low to avoid any structural perturbations resulting from covalent modification of the protein. In this study, we demonstrate that proteins can be labeled more extensively via DEPC and still provide accurate structural information. To show this, we modeled labeling kinetics over a range of DEPC concentrations and used molecular dynamics simulations to investigate the molecular-level effects of extensive labeling on the protein structure. Our results indicate that higher extents of DEPC labeling do not significantly perturb the protein structure and can lead to improved precision, detectability of labeled peptides, and protein structural resolution. Furthermore, higher extents of labeling enable better identification of protein-ligand binding sites where lower extents of modification provide ambiguous results.

    Abstract The politics of health care policy create opportunities driven by public demands for lowering costs and providing health security, constrained by fear of loss of what people have now, fears that will be inflamed by opponents. The best course toward reform will provide people a choice of improved public coverage and the ability to keep improved employer coverage. There are a variety of ways to design these proposals, some of which can be characterized as Medicare for All. Inclusion of a Medicare/public plan in the Affordable Care Act marketplaces is a policy option but one of relatively low significance.

    Abstract Mast cells are residents of the tubular gastrointestinal (GI) tract, where they play an important role in host defence and other vital functions. Dysregulation of mast cells has been implicated in the pathogenesis of several neoplastic, inflammatory, and functional disorders, some of which may manifest with GI symptoms. Surgical pathologists must therefore confront when and how to evaluate GI biopsies for mast cells, and whether such decisions should be based on morphologic criteria, clinical context, or direct request from clinical colleagues. The pathologist's role in evaluation of mast cell infiltrates is best defined in the diagnosis of systemic mastocytosis, where the utility of morphologic assessment coupled with ancillary studies is well established. In contrast, in nonneoplastic mast cell disorders such as mast cell activation syndrome, irritable bowel syndrome, or so-called 'mastocytic enterocolitis', a role for histopathology, if any, is controversial. Despite this, pathologists have seen a sharp increase in requests for mast cell quantification in the latter setting, despite these requests not being supported by published evidence. Moreover, what constitutes a 'normal' number of mast cells in a luminal GI biopsy is not well established. As a result, there is considerable variation in how these requests are handled in practice. This review evaluates and summarizes the published evidence relating to mast cell evaluation in endoscopic GI biopsies in various clinical scenarios, with a goal of providing practical, evidence-based guidance for the surgical pathologist when approached with requests for mast cell quantification in GI biopsies.

    Abstract Abstract In natural sciences, mineral exploration has a high network centrality. For industries with high technological- and knowledge proximity, transfer effects are an important function for innovation. Despite the high level of proximity between mineral exploration and other natural sciences, scholars hardly examine transfers from and to mineral exploration. This paper analyzes obstacles and mechanisms of transfer effects in and from mineral exploration and finds answers on how to institutionalize knowledge and technology transfer (KTT). The study employs a qualitative research design. The underlying database consists of 16 expert interviews, from the fields of natural science. The results show that KTT between areas as diverse as mineral exploration, healthcare, and arts are possible. A lack of interdisciplinary exchange and rigid scientific structures is the main inhibitor of KTT. Before this study, evidence for KTT from and to smaller industries is mostly anecdotal. The study is among the few, which investigates KTT concerning functional transfer opportunities.

    Abstract Inflammatory bowel diseases (IBD) are chronic inflammatory diseases affecting the gastrointestinal (GI) tract. Patients with IBD, besides other non-neoplastic complications, are also at increased risk of GI malignancies such as colorectal cancer, small bowel adenocarcinoma and lymphoma. The principal purpose of imaging in patients with IBD to assess complications and to stage a clinically known cancer. In addition, the goal of imaging has expanded to include the diagnosis of GI malignancies in clinical situations where colonoscopy cannot be performed or is incomplete. In addition, imaging allows the detection of cancers in patients where the development of either disease-related or treatment-related neoplasia is clinically suspected. The purpose of this review is to present the different imaging techniques used to detect GI malignancies in IBD patients and describe the radiological appearances of GI malignancies in IBD patients.
