Abstract Matrix metalloproteinase 9 (MMP-9) plays an important role in wound healing. However, overexpression of MMP-9 leads to the degradation of the newly formed extracellular matrix, which delays wound healing, ultimately leading to chronic wounds. Therefore, timely monitoring of the MMP-9 activity using simple, cost-effective methods is important to prevent the formation of chronic wounds. In this work, ferrocene-modified MMP-9 cleavage peptide (Fc-MG) modified carboxymethyl chitosan hydrogels were prepared as electrochemical biosensors. In the presence of MMP-9, the peptide chain is sheared, and the electrochemically active ferrocene segment is released. Therefore, analyzing the electrochemical activity of hydrogels using differential pulse voltammetry (DPV) can be used to determine MMP-9 activity. The results showed that the DPV peaks were correlated with the MMP-9 concentration in phosphate-buffered saline (PBS, pH 7.4) and Dulbecco's modified Eagle's medium (DMEM). Specifically, the corresponding coefficient of determination (R

Abstract Since combined sewer systems are still considered as a threat to urban water environment, understand their pollution transport process and load distribution characteristics during rainfall-runoff events would assist to mitigate urban stormwater pollution. In this study, built-up and wash-off processes of particulate pollutants on road surface and in sewer were synchronously observed during eight natural rainfall events in the combined sewer system in Zhuhai, Southern China. Field observation results showed the significant influence of particles size distribution characteristics on TSS concentration. High TSS concentrations (Road runoff, >200 mg/L; combined sewer runoff, >150 mg/L) usually contained relatively coarse particles (>100 μm) during the eight rainfall events, but the counter argument. Combined sewer runoff had a particle size coarser than road runoff. The relationship between the event means concentration (EMC) of TSS from road and combined sewer runoff varied with rainfall. EMC of TSS in road runoff were higher than that in sewer runoff during small rainfall, while that in combined sewer runoff were higher during large rainfall. The results also showed that 65.2 % of total nitrogen (TN) loadings and 62.3 % of total phosphorous (TP) loadings in the catchment came from combined sewer sediments, while 58.6 % of chemical oxygen demand (COD) loadings came from road runoff, which were mainly determined by the content of each pollutant in the sewer sediments and road-deposited sediments (RDS). Additionally, high pollutant loads phases (drainage of pollutant loads faster than runoff at a given incremental time during an event) of road runoff occurred earlier than that of combined sewer runoff. These findings can provide a basis for the type selection and scale determination of stormwater control measures in combined sewer systems.

Abstract Real-time sensing of reactive oxygen species (ROS) and timely scavenging of excessive ROS in physiological environments are critically important in the diagnosis and prevention of ROS-related diseases. To solve the mismatch problem between conventional rigid ROS biosensors and biological tissues in terms of both modulus and composition, here, we present a flexible ferrocene-based hydrogel biosensor designed for oxidative stress detection and antioxidation treatment. The hydrogel was fabricated through a supramolecular assembly of ferrocene-grafted polyethylenimine (PEI-Fc), sodium alginate (SA), and polyvinyl alcohol (PVA). Multiple non-covalent interactions, including electrostatic interactions between PEI-Fc and SA, hydrophobic interactions and π-π stacking among ferrocene groups, and the PVA crystalline domain, synergistically improve the mechanical properties of the PVA/SA/PEI-Fc hydrogel. The flexible PVA/SA/PEI-Fc hydrogel biosensor exhibited a broad detection range for hydrogen peroxide (H

Abstract The objective of this study was to evaluate the safety and efficacy of percutaneous pedicle screw fixation combined with bone cement augmentation in the management of stage III Kümmell disease without nerve deformity. A retrospective analysis was conducted on 17 patients diagnosed with stage III Kümmell disease without nerve deformity, who underwent treatment with percutaneous pedicle screw fixation combined with bone cement augmentation between April 2019 and 2022. Preoperative, postoperative, and final follow-up clinical outcome measures were collected, including Visual Analog Scale scores and Oswestry Disability Index scores. Additionally, lateral radiography was used to measure the Cobb angle and height of the anterior border of the affected vertebral body. Operative time, volume of injected bone cement, intraoperative cement leakage, and other complications were recorded. All patients underwent successful surgery, resulting in significant reductions in Visual Analog Scale scores, Oswestry Disability Index scores, and Cobb angle postoperatively; meanwhile there was a significant increase in height of the anterior border of the affected vertebral body. No major complications occurred during the follow-up period. In conclusion, percutaneous pedicle screw short-segment fixation combined with bone cement augmentation appears to be an effective surgical option for treating stage III Kümmell disease without nerve deformities.

Abstract Background. Systemic lupus erythematosus (SLE) is an autoimmune disease involving multiple organs, with atypical clinical manifestations and indefinite diagnosis and treatment. So far, the etiology of the disease is not completely clear. Current studies have known the interaction of genetic system, endocrine system, infection, environment, and other factors. Due to abnormal immune function, the human body, with the participation of various immune cells such as T cells and B cells, abnormally recognizes autoantigens, so as to produce a variety of autoantibodies and combine them to form immune complexes. These complexes will stay in the skin, kidney, serosa cavity, large joints, and even the central nervous system, resulting in multisystem damage of the body. The disease is heterogeneous, and it can show different symptoms in different populations and different disease stages; patients with systemic lupus erythematosus need individualized diagnosis and treatment. Therefore, we aimed to search for SLE immune-related hub genes and determine appropriate diagnostic genes to provide help for the detection and treatment of the disease. Methods. Gene expression data of whole blood samples of SLE patients and healthy controls were downloaded from the GEO database. Firstly, we analyzed and identified the differentially expressed genes between SLE and the normal population. Meanwhile, the single-sample gene set enrichment analysis (ssGSEA) was used to identify the activation degree of immune-related pathways based on gene expression profile of different patients, and weighted gene coexpression network analysis (WGCNA) was used to search for coexpressed gene modules associated with immune cells. Then, key networks and corresponding genes were found in the protein-protein interaction (PPI) network. The above corresponding genes were hub genes. After that, this study used receiver operating characteristic (ROC) curve to evaluate hub gene in order to verify its ability to distinguish SLE from the healthy control group, and ...

Abstract Road-deposited sediments (RDS) on urban impervious surfaces are important carriers of heavy metals (HMs), and can contribute to urban runoff pollution. With the outbreak of COVID-19, chlorinated disinfectants (CDs) have been extensively sprayed on these surfaces. This practice may have a superposed or priming effect on HMs contaminants in RDS, yet this remains unknown. This study examined the effects of seven CDs concentration gradients (0, 250, 500, 1000, 2000, 5000, 60,000 mg/L) on the leaching and chemical forms of HMs (Cd, Cr, Ni, Pb, and Zn) in seven particle size fractions (<44, 44-63, 63-105, 105-149, 149-250, 250-450, 450-1000 μm). The results showed that CDs can promote the leaching of HMs in RDS, at the recommended CDs dose (2000 mg/L), except for Pb, the leaching amounts increased by 21.8%-237.2% compared with the untreated RDS. The alteration in the leaching were primarily attributed to the redistribution of chemical forms of HMs in RDS, specifically, the acid-extractable fractions percentage increased by 0.23%-24.39%, and the reducible fractions percentages decreased by 3.21%-38.35%. The lower oxidation-reduction potential (ORP) and alkalinity of CDs as strong oxidants were responsible for the redistribution of forms. The leaching and chemical forms of HMs vary among different particle sizes, but in any case, finer particle sizes (< 105 μm) still dominate their contribution. The current control measure of street sweeping is ineffective in removing these particles. These findings will facilitate the development of strategies for controlling urban diffuse pollution from RDS during the pandemic. Finally, this study suggests potential directions for future research.
