    Abstract Objectives: At the onset of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic in the United States, testing was limited to the Centers for Disease Control and Prevention-developed reverse transcription polymerase chain reaction assay. The urgent and massive demand for testing prompted swift development of assays to detect SARS-CoV-2. The objective of this study was to assess the accuracy of these newly developed tests.
    Methods: The American Proficiency Institute sent 2 test samples to 346 clinical laboratories in order to assess the accuracy of SARS-CoV-2 assays. The positive sample, containing 5,175 viral copies/mL, was fully extractable with SARS-CoV-2 viral capsid protein and RNA. The negative sample, with 3,951 viral copies/mL, contained recombinant virus particles with sequences for targeting human RNAase P gene sequences.
    Results: Of the laboratories submitting results, 97.4% (302/310) correctly detected the virus when present and 98.3% (296/301) correctly indicated when the virus was not present. Among incorrect results reported in this proficiency challenge, 76.9% (10/13) were likely related to clerical error. This accounts for 1.6% (10/611) of all reported results.
    Conclusions: Overall performance in this SARS-CoV-2 RNA detection challenge was excellent, providing confidence in the results of these new molecular tests and assurance for the clinical and public health decisions based on these test results.

    Abstract Leptospira is a genus of bacteria that includes free-living saprophytic species found in water or soil, and pathogenic species, which are the etiologic agents of leptospirosis. Besides all the efforts, there are only a few proteins described as virulence factors in the pathogenic strain L. interrogans. This work aims to perform L. biflexa serovar Patoc1 strain Paris global proteome and to compare with the proteome database of pathogenic L. interrogans serovar Copenhageni strain Fiocruz L1-130. We identified a total of 2327 expressed proteins of L. biflexa by mass spectrometry. Using the Get Homologues software with the global proteome of L. biflexa and L. interrogans, we found orthologous proteins classified into conserved, low conserved, and specific proteins. Comparative bioinformatic analyses were performed to understand the biological functions of the proteins, subcellular localization, the presence of signal peptide, structural domains, and motifs using public softwares. These results lead to the selection of 182 low conserved within the saprophyte, and 176 specific proteins of L. interrogans. It is anticipated that these findings will indicate further studies to uncover virulence factors in the pathogenic strain. This work presents for the first time the global proteome of saprophytic strain L. biflexa serovar Patoc, strain Patoc1. SIGNIFICANCE: The comparative analysis established an array of specific proteins in pathogenic strain that will narrow down the identification of immune protective proteins that will help fight leptospirosis.

    Abstract Introduction: The coronavirus 2019 pandemic (COVID-19) has resulted in major changes in lifestyle practices and healthcare delivery. The goal of this study was to examine changes in practice and service outcomes in a telehealth program before and after the federal and private telehealth policy expansion during the COVID-19 pandemic. These findings are particularly useful to understand what may be needed to overcome telehealth challenges in future disasters.
    Methods: We conducted a cross-sectional analysis of virtual visits through a statewide telehealth center embedded in a large academic healthcare system. Primary outcomes of this study were changes in telehealth visits pre- and post-policy expansions among at-risk populations.
    Results: A total of 2,132 telehealth visits were conducted: 1,530 (71.8 percent) patients were female, 1,561 (73.2 percent) were between the ages 18-50, 1,576 (74 percent) were uninsured, and 1,225 (57.5 percent) were from rural regions. The average number of telehealth visits per day increased from 14 to 33 visits post-expansion. A significant change in patient characteristics was found among senior, uninsured, and rural patients after the telehealth expansion.There was an 11 percent decrease in telehealth visits from very high vulnerability regions post-expansion compared to pre-expansion. There was a 15 percent decrease in visits resulting in prescription post-expansion (p-value<0.01).
    Conclusions: COVID-19 policy expansions expanded telehealth utilization among at-risk populations such as senior, uninsured, and rural patients while decompressing hospitals and emergency rooms and maintaining positive patient experiences. Further regulations are needed around virtual visits unintended consequences, software certification, and guidelines for workforce training.

    Abstract Magnetic resonance imaging (MRI) is a well-known and widespread imaging modality for neuroscience studies and the clinical diagnoses of neurological disorders, mainly due to its capability to visualize brain microstructures and quantify various metabolites. Additionally, its noninvasive nature makes possible the correlation of high-resolution MRI from ex vivo brain samples with histology, supporting the study of neurodegenerative disorders such as Alzheimer's or Parkinson's disease. However, the quality and resolution of ex vivo MRI highly depend on the availability of specialized radiofrequency coils with maximized filling factors for the different sizes and shapes of the samples to be studied. For instance, small, dedicated radiofrequency (RF) coils are not always commercially available in ultrahigh field whole-body MRI scanners. Even for ultrahigh field preclinical scanners, specific RF coils for ex vivo MRI are expensive and not always available. Here, we describe the design and construction of two RF coils based on the solenoid geometry for ex vivo MRI of human brain tissues in a 7T whole-body scanner and for ex vivo MRI of marmoset brain samples in a 9.4T preclinical scanner. We designed the 7T solenoid RF coil to maximize the filling factor of human brain samples conditioned on cassettes for histology, while the 9.4T solenoid was constructed to accommodate marmoset brain samples conditioned in 50 ml centrifuge tubes. Both solenoid designs operate in transceiver mode. The measured B1+ maps show a high level of homogeneity in the imaging volume of interest, with a high signal-to-noise ratio over the imaging volume. High-resolution (80 µm in plane, 500 µm slice thickness) images of human brain samples were acquired with the 7T solenoid, while marmoset brain samples were acquired with an isotropic resolution of 60 µm using the 9.4T solenoid coil.

    Abstract Abstract This report describes the photoacoustic and electron paramagnetic resonance investigations of Brazilian and Cuban zeolites. Photoacoustic optical absorption measurements indicate the presence of iron (Fe 3 +) ions with their respective transition bands for both zeolites. Two species of manganese (Mn 2 + and Mn 3 +) were identified in the Cuban sample and the electronic transitions assigned. Iron and manganese ions were confirmed through nonradiative relaxation ( τ) and characteristic diffusion ( τ β) times evaluation, whose values were found to be τ BRA = 5.40 ms, τ CUB = 4.60 ms, τ β BRA = 387 μs and τ β CUB = 305 μs. Crystal field (Dq- BRA/Dq- CUB = 1048 cm − 1/945 cm − 1) plus Racah (B- BRA/B- CUB = 457 cm − 1/813 cm − 1 and C- BRA/C- CUB = 3655 cm − 1/2496 cm − 1) parameters were assessed as well. Paramagnetic resonance corroborated Fe 3 + ions present in the Brazilian zeolite occupying sites showing axial and/or rhombic symmetry distortions. For the Cuban sample, results reveal the characteristic hyperfine sextet lines of Mn 2 + overlapping the Fe 3 + line. Values of Landé factor and isotropic hyperfine splitting constant were found to be 2.0 and 9.7 mT, respectively. This tells us that the Mn 2 + lies in octahedral symmetry probably replacing calcium ions and point towards an ionic bonding character of the Mn 2 + and its surroundings.

    Abstract Objective This study was conducted to evaluate the effects of the provision of a protein-rich supplement on productive performance, and metabolic profile on grazing suckling female beef calves in tropical conditions during 150 d of experimentation. Methods Fifty-six Nellore suckling female calves, and their respective dams were distributed in a completely randomised design and made to undergo two treatments as follows: UNS (without supplementation), and SUP (supplementation with 5 g/kg body weight [BW] of a protein supplement). Throughout the experiment, animal performance and metabolic profile were evaluated. Also, ureagenesis and gluconeogenesis were assessed for gene expression. Results SUP female calves showed a higher voluntary intake (p≤0.03) of the diet components evaluated, digestibility of organic matter (p≤0.02) and microbial nitrogen production (MICN; p≤0.02) compared to UNS female calves. In its turn, serum urea nitrogen (p≤0.01) and insulin-like growth factor-1 (p≤0.03) levels and ureagenesis (p≤0.04) increased in SUP female calves compared to UNS female calves. Blood glucose and triglyceride levels were not affected by supplementation. The average daily gain (ADG) from SUP female calves was higher (p≤0.02) compared with UNS female calves. However, supplementation did not affect the body measures of the animals. Conclusion In summary, provision of a protein-rich supplement improves the intake and nutrients digestibility, ADG and final BW and increases metabolic indicators of the protein status in grazing suckling female beef calves in tropical conditions.
