    Abstract The impact of host adaptive immune response on COVID-19 has now become a critical issue in absence of specific therapy and immunotherapies. In SARS CoV-2 infection, the immune response is thought to contribute both to the pathogenesis of the disease and to protection during its resolution. While mild cases develop an immune response that contributes to host protection, immunity of severely infected patients is a balance between harmful and protective immune responses. The severity of the disease has raised many questions about the kinetic, amplitude and the quality of adaptive immunity to the virus and its generation during the early phases of infection in severe, mild and asymptomatic patients.     Le rôle protecteur de la réponse immunitaire adaptative de l’hôte au cours de l’infection par le SARS-CoV-2 est devenu une question critique en l’absence d’un traitement spécifique, d‘un vaccin préventif ou d’une immunothérapie. Au cours de l’infection par le SARS-CoV-2, la réponse immunitaire contribuerait à la défense de l’hôte dans la majorité des cas, mais serait responsable de sa pathogénèse chez certains malades. Notamment, au cours des formes sévères, un déséquilibre entre les réponses immunitaires innée et adaptative pourrait être fatal. Au cours de la COVID-19, de nombreuses questions se posent sur la génération de l’immunité spécifique contre les diverses protéines du virus, la cinétique, la fonction des anticorps, ainsi que la qualité des réponses des lymphocytes effecteurs CD4+ et CD8+ pour la protection de l’hôte. L’étude bio-informatique des épitopes T et B des coronavirus a soulevé la question de l’immunité croisée entre le SARS-COV-2 et d’autres coronavirus sources d’infection bénigne ou responsables de pneumopathies graves telles que le MERS-CoV et le SARS-CoV. Dans cette revue, nous faisons le point sur les réponses immunitaires adaptatives au cours de la COVID-19 et leurs rôles potentiels dans la protection des personnes infectées.

    Abstract Vaccination is one of the major public health advances in modern medicine. But in order to improve the effectiveness of existing vaccines and develop new ones, we need to know more about the mechanisms of action that lead to protective immunity and the strategies that lead to sustainable defence. Cutaneous vaccination is a promising procedure because of the richness of innate cells in the skin and their role in the quality, intensity and persistence of adaptive responses. A Systems Biology approach is used to probe immunological dynamic post-vaccination and to simultaneously analyse a large number of variables: antibody and cytokines levels, gene expression or even cellular outcomes. Immunological data have been collected from an influenza vaccination clinical trial and, thanks to this approach, provided insight into the impact of the immunization route on the quality of immune responses. An innate gene signature is also identified in order to predict the intensity of immune responses.

    Abstract Immunomonitoring is the study of an individual's immune responses over the course of vaccination or infection. In the infectious context, exploring the innate and adaptive immune responses will help to investigate their contribution to viral control or toxicity. After vaccination, immunomonitoring of the correlate(s) and surrogate(s) of protection is a major asset for measuring vaccine immune efficacy. Conventional immunomonitoring methods include antibody-based technologies that are easy to use. However, promising sensitive high-throughput technologies allowed the emergence of holistic approaches. This raises the question of data integration methods and tools. These approaches allow us to increase our knowledge on immune mechanisms as well as the identification of key effectors of the immune response. However, the depiction of relevant findings requires a well-rounded consideration beforehand about the hypotheses, conception, organization and objectives of the immunomonitoring. Therefore, well-standardized and comprehensive studies fuel insight to design more efficient, rationale-based vaccines and therapeutics to fight against infectious diseases. Hence, we will illustrate this review with examples of the immunomonitoring approaches used during vaccination and the COVID-19 pandemic.

    Abstract HIV-1 is responsible for a global pandemic of 35 million people and continues to spread at a rate of >2 million new infections/year. It is widely acknowledged that a protective vaccine would be the most effective means to reduce HIV-1 spread and ultimately eliminate the pandemic, whereas a therapeutic vaccine might help to mitigate the clinical course of the disease and to contribute to virus eradication strategies. However, despite more than 30 years of research, we do not have a vaccine capable of protecting against HIV-1 infection or impacting on disease progression. This, in part, denotes the challenge of identifying immunogens and vaccine modalities with a reduced risk of failure in late stage development. However, progress has been made in epitope identification for the induction of broadly neutralizing antibodies. Thus, peptide-based vaccination has become one of the challenges of this decade. While some researchers reconstitute envelope protein conformation and stabilization to conserve the epitope targeted by neutralizing antibodies, others have developed strategies based on peptide-carrier vaccines with a similar goal. Here, we will review the major peptide-carrier based approaches in the vaccine field and their application and recent development in the HIV-1 field.

    Abstract Vaccination has been a major advance for health care, allowing eradication or reduction of incidence and mortality of various infectious diseases. However, there are major pathogens, such as Human Immunodeficiency Virus (HIV) or the causative agent of malaria, for which classical vaccination approaches have failed, therefore requiring new vaccination strategies. The development of new vaccine strategies relies on the ability to identify the challenges posed by these pathogens. Understanding the pathogenesis and correlates of protection for these diseases, our ability to accurately direct immune responses and to vaccinate specific populations are such examples of these roadblocks. In this respect, the use of a robust, cost-effective and predictive animal model that recapitulates features of both human infection and vaccination is currently a much-needed tool. We discuss here the major limitations faced by modern vaccinology and notably, the development of humanized mice for assessing the immune system, along with their potential as vaccine models.
