    Abstract The serotonin content of growing fruits and of germinating seeds of Juglans regia has been studied. In the embryo 0.4-0.6 mg serotonin/g FW were found; in contrast no serotonin was detectable in the fleshy pericarp and in the seed coat. Serotonin was also not detectable in leaves, stems and roots of the adult plant. Most of the serotonin found in the embryo is formed after abscission of the seeds. During the synthesis of serotonin there are no dramatic changes in the chemical composition of the seeds (Tables 3-5).The formation of serotonin could be followed in isolated cotyledons and under sterile conditions. This serotonin formation is stimulated by exogenous tryptophan (Fig. 2). That tryptophan acts as a precursor of serotonin could be demonstrated with labelled DL-tryptophan (benzene ring (14)C) (U). The possibility of stimulating serotonin formation in isolated cotyledons by the addition of tryptophan is limited to a certain stage of development and cannot be observed with material from fully matured seeds (Fig. 3).No serotonin was found in callus tissue and adventitious roots formed by isolated cotyledons; all the serotonin remained in the cotyledons. This was also the case in young seedlings, in which only the cotyledons showed the characteristic high serotonin content, whereas leaves, stems and roots contained no serotonin (Table 6).From these data we conclude that serotonin formation in the embryo of Juglans regia is not a special type of nitrogen storage but a way of ammonia detoxification in which ammonia from protein amino acid degradation is incorporated into serotonin via tryptophan.

    Abstract Unnatural mirror image l-configured oligonucleotides (L-ONs) are a convenient substance class for the application as complementary in vivo recognition system between a tumor specific antibody and a smaller radiolabeled effector molecule in pretargeting approaches. The high hybridization velocity and defined melting conditions are excellent preconditions of the L-ON based methodology. Their high metabolic stability and negligible unspecific binding to endogenous targets are superior characteristics in comparison to their d-configured analogs. In this study, a radiopharmacological evaluation of a new l-ONs based pretargeting system using the epidermal growth factor receptor (EGFR) specific antibody cetuximab (C225) as target-seeking component is presented.     Abstract The antileishmanial activity of the essential oil (EO) from Chenopodium ambrosioides L. has been demonstrated in vitro and in animal models, attributed to the major components of the EO. This study focused on the effects of the three major EO compounds carvacrol, caryophyllene oxide (Caryo), and the antileishmanial endoperoxide ascaridole (Asc) on mitochondrial functions in Leishmania tarentolae promastigotes (LtP). EO and Caryo were able to partially inhibit the leishmanial electron transport chain, whereas other components failed to demonstrate a direct immediate effect. Caryo demonstrated inhibition of complex III activity in LtP and in isolated complex III from other species. The formation of superoxide radicals was studied in Leishmania by electron spin resonance spectroscopy in the presence of iron chelators wherein selected compounds failed to trigger a significant immediate additional superoxide production in LtP. However, upon prolonged incubation of Leishmania with Asc and especially in the absence of iron chelators (allowing the activation of Asc), an increased superoxide radical production and significant impairment of mitochondrial coupling in Leishmania was observed. Prolonged incubation with all EO components resulted in thiol depletion. Taken together, the major components of EO mediate their leishmanicidal activity via different mitochondrial targets and time profiles. Further studies are required to elucidate possible synergistic effects of carvacrol and Asc and the influence of minor compounds.
