    Abstract The volume deals with the juridical and institutional framework of athletic and artistic games from archaic Greece to late antique Rome. The agones' rules and regulations, the funding by public contributions and wealthy private sponsors were approached as well as the athletes' training and prizes and privileges, which determined the athletes'-and especially the winner's- social status. Moreover attention was drawn onto the end of the classical Greek agones from a Roman perspective.The Second Vienna Colloquium on Ancient Legal History addressed the juridical and institutional framework of athletic and artistic games in classical antiquity. The chronological frame ranged from Archaic times to Late Antiquity (8th cent. BCE - 6th cent. CE). The symposium covered three main areas of interest. Firstly, the agones' rules and regulations, the funding by public contributions and wealthy private sponsors as well as the athletes' training were approached. Prizes and privileges, which determined the athletes'-and especially the winner's- social status and were thus crucial to the agones' appeal, were then attended to by several papers. In contrast to the conditions under the Imperium Romanum, where the emperor enforced nation-wide regulations, the "politics of sports" followed different rules in the small-scaled world of the classical Greek poleis. Thus, differences in the conception of sports between Greeks and Romans were traced and exemplarily demonstrated on the basis of the gladiatorial games. Lastly, attention was drawn onto the end of the classical Greek agones from a Roman perspective. The now Christian empire couldn't tolerate the pagan agones anymore and instead developed new forms of athletic competitions and public entertainment: late antique circus games and chariot races

    Abstract In eukaryotic organisms, proteins are typically translated from monocistronic messenger RNAs containing a single coding sequence (CDS). However, recent long transcript sequencing identified 87 nuclear polycistronic mRNAs in Chlamydomonas reinhardtii natively carrying multiple co-expressed CDSs. In this study, we investigated the dynamics of 22 short intergenic sequences derived from these native polycistronic loci by their application in genetic constructs for synthetic transgene expression. A promising candidate sequence was identified based on the quantification of transformation efficiency and expression strength of a fluorescence reporter protein. Subsequently, the expression of independent proteins from one mRNA was verified by cDNA amplification and protein molecular mass characterization. We demonstrated engineered bicistronic expression in vivo to drive successful co-expression of several terpene synthases with the selection marker aphVIII. Bicistronic transgene design resulted in significantly increased (E)-α-bisabolene production of 7.95 mg L

    Abstract Downstream processing is the bottleneck in the continuous manufacturing of monoclonal antibodies (mAbs). To overcome throughput limitations, two different continuous processes with a novel convective diffusive protein A membrane adsorber (MA) were investigated: the rapid cycling parallel multi-column chromatography (RC-PMCC) process and the rapid cycling simulated moving bed (RC-BioSMB) process. First, breakthrough curve experiments were performed to investigate the influence of the flow rate on the mAb dynamic binding capacity and to calculate the duration of the loading steps. In addition, customized control software was developed for an automated MA exchange in case of pressure increase due to membrane fouling to enable robust, uninterrupted, and continuous processing.
