    Abstract The allocation of carbon emission reduction responsibility is severe issue in China for these years. In case to find a fairer and more effective way to divided the responsibility to each region of China, this paper examines embodied carbon emissions (ECEs) transfers in China's inter-regional trade by applying value-added extended decomposition model. This study allows policymakers to trace CO2 emissions at regional levels and provides three key findings. Firstly, using novel data on the physical consumption of energy by region, we observe a strong and robust negative association between the regional direct CO2 emission coefficient and the regional economic development level. Secondly, employing the latest inter-regional input-output table of China to calculate ECEs and uncover transfer characteristics via inter-regional trade, results show that central region, eastern region and northern region are the three highest ECEs regions. Thirdly, ECEs in value-added trade are generally transferred from inland China to coastal areas of China. Northeast region, north coastal region, central region and northwest region are net ECEs outflow regions, the rest regions are net ECEs inflow regions.

    Abstract Precipitation is one of the many important natural factors impacting agriculture and natural resource management. Although statistics have been applied to investigate the non-stationary trend and the unpredictable variances of precipitation under climate change, existing methods usually lack a sound physical basis that can be generally applied in any location and at any time for future extrapolation, especially in tropical areas. Physically, the formation of precipitation is a result of ascending air which reduces air pressure and condenses moisture into drops, either by irregular terrain or atmospheric phenomena (e.g., via frontal lifting). Thus, in this paper, pressure change events (PCEs) will be used as a physical indicator of the stability of atmospheric systems to reveal the impact of temperature on precipitation in the tropical areas of Florida. By using data from both national and regional weather observation networks, this study segments the continuous observation series into PCE sequences for further analysis divided by dry and wet seasons. The results reveal that the frequency and intensity of PCE are highly associated with the occurrences of weather events. Decreasing pressure favors precipitation, and may turn extreme when the temperature and air moisture are sufficient to fuel the process. With similar intensity, decreasing pressure change events (DePCEs) generally bear a higher probability of precipitation (POP) and precipitation depth (PD) than increasing pressure change events (InPCEs). The frequency of alternating between InPCEs and DePCEs is subject to the temperature of the season and climate. Due to the seasonal fluctuations of weather characteristics, such as temperature and relative humidity, the dependence of extreme precipitation on these characteristics can be interpreted via PCE. A 7% increase rate of precipitation vs. temperature rise, determined by the Clausius—Clapeyron (C—C) relationship, can be observed from extreme precipitation with variances in the season and PCE types. ...

    Abstract Although surgery followed by platinum-based therapy is effective as a standard treatment in the early stages of ovarian cancer, the majority of cases are diagnosed at advanced stages, leading to poor prognosis. Thus, the identification of novel therapeutic drugs is needed. In this study, we assessed the effectiveness of bepridil-a calcium channel blocker-in ovarian cancer cells using two cell lines: SKOV-3, and SKOV-3-13 (a highly metastatic clone of SKOV-3). Treatment of these cell lines with bepridil significantly reduced cell viability, migration, and invasion. Notably, SKOV-3-13 was more sensitive to bepridil than SKOV-3. The TGF-β1-induced epithelial-mesenchymal transition (EMT)-like phenotype was reversed by treatment with bepridil in both cell lines. Consistently, expression levels of EMT-related markers, including vimentin, β-catenin, and Snail, were also substantially decreased by the treatment with bepridil. An in vivo mouse xenograft model was used to confirm these findings. Tumor growth was significantly reduced by bepridil treatment in SKOV-3-13-inoculated mice, and immunohistochemistry showed consistently decreased expression of EMT-related markers. Our findings are the first to report anticancer effects of bepridil in ovarian cancer, and they suggest that bepridil holds significant promise as an effective therapeutic agent for targeting metastatic ovarian cancer.

    Abstract Background: Although cytoreductive surgery followed by adjuvant chemotherapy is effective as a standard treatment for early-stage ovarian cancer, the majority of ovarian cancer cases are diagnosed at the advanced stages with dissemination to the peritoneal cavity, leading to a poor prognosis. Therefore, it is crucial to understand the cellular and molecular mechanisms underlying metastasis and identify novel therapeutic targets.
    Objective: In this study, we aimed to elucidate the mechanisms underlying gene expression alterations during the acquisition of metastatic potential and characterize the metastatic subpopulations within ovarian cancer cells.
    Methods: We conducted single-cell RNA sequencing of two human ovarian cancer cell lines: SKOV-3 and SKOV-3-13, a highly metastatic subclone of SKOV-3. Suppression of NFE2L1 expression was performed through siRNA-mediated knockdown and CRISPR-Cas9-mediated knockout.
    Results: Clustering and pseudotime trajectory analysis revealed pro-metastatic subpopulation within these cells. Furthermore, gene set enrichment analysis and prognosis analysis indicated that NFE2L1 could be a key transcription factor in the acquisition of metastasis potential. Inhibition of NFE2L1 significantly reduced migration and viability of both cells. In addition, NFE2L1 knockout cells exhibited significantly reduced tumor growth in a mouse xenograft model, recapitulating in silico and in vitro results.
    Conclusion: The results presented in this study deepen our understanding of the molecular pathogenesis of ovarian cancer metastasis with the ultimate goal of developing treatments targeting pro-metastatic subclones prior to metastasis.

    Abstract Idiopathic pulmonary fibrosis (IPF) is considered as a chronic, fibrosing interstitial pneumonia with unknown mechanism. The present work aimed to explore the function, biogenesis and regulatory mechanism of circELP2 in pulmonary fibrosis and evaluate the value of blocking circELP2-medicated signal pathway for IPF treatment. The results showed that heterogeneous nuclear ribonucleoprotein L initiated reverse splicing of circELP2 resulting in the increase of circELP2 generation. The biogenetic circELP2 activated the abnormal proliferation and migration of fibroblast and extracellular matrix deposition to promote pulmonary fibrogenesis. Mechanistic studies demonstrated that cytoplasmic circELP2 sponged miR-630 to increase transcriptional co-activators Yes-associated protein 1 (YAP1) and transcriptional co-activator with PDZ-binding motif (TAZ). Then, YAP1/TAZ bound to the promoter regions of their target genes, such as mTOR, Raptor and mLST8, which in turn activated or inhibited the genes expression in mitochondrial quality control pathway. Finally, the overexpressed circELP2 and miR-630 mimic were packaged into adenovirus vector for spraying into the mice lung to evaluate therapeutic effect of blocking circELP2-miR-630-YAP1/TAZ-mitochondrial quality control pathway in vivo. In conclusion, blocking circELP2-medicated pathway can alleviate pulmonary fibrosis, and circELP2 may be a potential target to treat lung fibrosis.

    Abstract BACKGROUND: Polygonatum cyrtonema Hua is cultivated for its edible and medical value. The steam‐processed rhizome of P. cyrtonema is the main form for daily consumption and it has been used traditionally in tonics for treating various age‐related disorders. The aim of our study was to compare the physicochemical properties and antioxidant activity of polysaccharides respectively extracted from crude P. Cyrtonema (PCPC), and steam‐processed P. cyrtonema (PCPS), and to explore a possible underlying antioxidant mechanism. RESULTS: The PCPC with a molecular weight of 4.35 × 10³ Da mainly consisted of fructose and trace amounts of glucose, whereas PCPS with 4.24 × 10⁴ Da was composed of fructose, arabinose, glucose, xylose, mannose, galacturonic acid and glucuronic acid. The PCPC had a triple‐helical conformation whereas PCPS was a random coil. Both exhibited free radicals‐ scavenging activity in vitro. In a mouse model of oxidative damage, PCPC or PCPS treatment significantly reversed histopathological alterations, reactive oxygen species (ROS) accumulation and the reduction of antioxidant enzyme activity. They both also promoted Nrf2 nuclear transport by decreasing Keap‐1 expression and increasing HO‐1 expression. Both in vitro and in vivo, PCPS exhibited more potent antioxidant activity than PCPC. CONCLUSION: Overall, the results suggest that PCPS has a stronger effect on the prevention of oxidative damage by activating Nrf2/HO‐1 antioxidant signaling. This study demonstrates the role of steam‐processed P. cyrtonema rhizome and provides valuable perspective for PCPS as a functional agent. © 2022 Society of Chemical Industry.

    Abstract In the pharmaceutical industry, the use of artificial intelligence (AI) has seen consistent growth over the past decade. This rise is attributed to major advancements in statistical machine learning methodologies, computational capabilities and the increased availability of large datasets. AI techniques are applied throughout different stages of drug development, ranging from drug discovery to post-marketing benefit-risk assessment. Kolluri et al. provided a review of several case studies that span these stages, featuring key applications such as protein structure prediction, success probability estimation, subgroup identification, and AI-assisted clinical trial monitoring. From a regulatory standpoint, there was a notable uptick in submissions incorporating AI components in 2021. The most prevalent therapeutic areas leveraging AI were oncology (27%), psychiatry (15%), gastroenterology (12%), and neurology (11%). The paradigm of personalized or precision medicine has gained significant traction in recent research, partly due to advancements in AI techniques \cite{hamburg2010path}. This shift has had a transformative impact on the pharmaceutical industry. Departing from the traditional "one-size-fits-all" model, personalized medicine incorporates various individual factors, such as environmental conditions, lifestyle choices, and health histories, to formulate customized treatment plans. By utilizing sophisticated machine learning algorithms, clinicians and researchers are better equipped to make informed decisions in areas such as disease prevention, diagnosis, and treatment selection, thereby optimizing health outcomes for each individual.
