    Abstract The increasing knowledge on skin physiology, formulation science and nanotechnology has led to continuous improvements in cosmetics, and introduction of dermocosmetics has been increasing particularly for the management of skin disorders such as acne, eczema, psoriasis, etc. Nowadays, research has been focused on the development of products which can efficiently administer active compounds to the target skin layers while minimizing side effects. The use of multifunctional lipid nanoparticles for cosmetic and dermocosmetic purposes is promising not only because biocompatible ingredients are used in their composition, but also because of their ability to show enhanced skin penetration. Although the introduction of liposomes has been a hallmark of lipid nanoparticles, development of novel systems capable of encapsulating active compounds with tunable release profiles, that show good stability, are easy to manufacture and handle remains a necessity. Solid lipid nanoparticles (SLN) were introduced as alternative formulations for emulsions, liposomes and polymeric nanoparticles, whereas nanostructured lipid carriers (NLC) were developed later as second-generation nanoparticles. However, both SLN and NLC show many inherited advantageous properties to be used for dermal applications including ability to provide occlusion and photoprotective effect and skin hydration, and various SLN and NLC based products are already in the market. This review provides an overview on the current state-of-art of SLN and NLC particularly for cosmetic and dermocosmetic purposes, discuss their formulation composition, structures and preparation techniques. Their use for the topical delivery of active compounds in different skin disorders is highlighted along with examples of commercialized products.

    Abstract Purple basil is widely used medicinally and aromatically due to its essential oil properties, and it is a plant that stands out with its antibacterial, antimutagenic and antioxidant properties. It has been determined that purple basil has positive effects on health such as appetizing, insomnia, sputum, diuretical and antispasmodic. Purple basil contains high concentrations of anthocyanins and with these properties it stands out as a potential new source of stable red pigments for the food industry. In recent years, it has been seen that purple basil has been processed into different products such as tea and sherbet in order to benefit from its functional properties. With the processing of purple basil into different products, it is seen that the consumption of basil and basil products has increased by consumers. In this study, purple basil sherbet samples were concentrated at different voltage gradient values (14, 17 and 20 V/cm) under atmospheric conditions with ohmic heating up to 25% soluble solid content (SSC) and the changes in their rheological properties during the concentration process was investigated. The changes in the rheological properties of the purple basil samples were determined in the range of 1-148 s-1 shear rate values and measurement temperature was 25°C. The compatibility of different rheological models (Newton, Power-Law and Herschel-Bulkley) to the experimental data was statistically evaluated to determine the flow behavior index of the purple basil sherbet samples. It has been determined that the shear stress values increased as the shear rate values increased in all the process conditions. Similarly, at the same shear rate values, it was determined that the shear stress values increased as the SSC values increased. It has been determined that the best suitable rheological model was Herschel-Bulkley Model for all purple basil sherbet concentrates.     Abstract Melanomas harboring an activating BRAFV600 mutation account for 50% of all advanced melanomas. The approval of BRAF-targeted therapy revolutionized treatment of these patients with achievement of impressive responses. However, development of resistance to these drugs is a significant problem, and as such, duration of response remains low, with median progression free survival of around 11-15 months. Immune checkpoint blockers exploit the immune system to eradicate cancer and can produce durable disease control that results in long-term, treatment-free survival in some patients. These drugs have shown very impressive survival in patients with BRAF-mutated melanoma. Thus, there is a need to continue to utilize emerging data to achieve long-term disease control for patients with advanced melanoma. Combining targeted therapy with immune therapy may be one possible way to achieve this goal. In this review, the mechanisms of action of these two pathways, including the mechanistic basis of this combination, are summarized, along with results of completed and ongoing trials in triple therapy.

    Abstract In this study, black cumin oil (BCO) oleogel was produced using natural waxes (beeswax: BW, sunflower wax: SFW, and carnauba wax: CW) to offer a healthy alternative to shortening. Oiling‐off was observed only in the BW oleogel (3.88 ± 0.02%). The CW oleogel had the lowest Eₐ (354.82 kJ/mol), indicating greater resistance to thermal degradation. The highest G′ and G″ were observed in the CW oleogel, while the lowest was observed in the BW oleogel. All oleogel samples displayed weak gel properties (tan δ > 0.1), but the lowest tan δ value belonged to the CW oleogel (0.12). After 4 weeks of storage, the peroxide values of the oleogels were lower than those of bulk oil (p < .05). Finally, the oxidation results showed that the BCO structuring by the waxes effectively protected it from oxidation over eight weeks of storage. NOVELTY IMPACT STATEMENT: The most important innovation of the present study was to investigate the effect of BCO oleogelation on the stability of thymoquinone, the main phenolic compound of BCO. The BCO oleogels had higher thymoquinone content during the first half of the storage period compared with bulk BCO, showing that the oleogel network provides a protective effect against oxidation and thermal degradation.

    Abstract Purpose of review: While anti-PD-1 antibodies have been a breakthrough in the treatment of patients with advanced melanoma, a substantial proportion of patients are still refractory to or progress after treatment with anti-PD-1 immunotherapy. Here, we review the post anti-PD-1 therapy alternatives that may be possible for patients with unresectable or metastatic stage 3 or 4 melanoma.
    Recent findings: Currently available treatment options include BRAF-targeted and MEK inhibitor-targeted therapies for those with BRAFV600 mutant melanoma, while for patients with BRAF-WT melanoma or those who have already received prior BRAF-targeted therapy, options include anti-CTLA-4 therapy, alone or in combination with anti-PD-1 therapy, or for selected patients, clinical trials that may incorporate other immune checkpoint inhibitors or co-stimulatory agonists, oncolytic virotherapies, adoptive cellular therapies, or other novel agents. Participation in clinical trials is critical in order to delineate what more effective treatment options are and which group of patients after receiving prior anti-PD-1 therapy.

    Abstract Objective: The prevalence of radiological involvement is considered a poor prognostic factor for coronavirus disease 2019 (COVID- 19). In our study, we aimed to investigate the threshold value of the chest X-ray (CXR) score, which would require the patient to be transferred to the intensive care unit (ICU) through scoring the CXR of COVID-19 patients receiving in-patient treatment.
    Material and methods: The patients that tested positive for COVID-19 on the basis of the polymerase chain reaction (PCR) test, who was hospitalized in our hospital between the dates of March 15 and May 30, 2020, were included in the study. The CXRs of these patients taken during hospitalization were scored. The threshold value of the CXR score of the patients transferred to the ICU was calculated. Patients were grouped according to the threshold value of the CXR score, and demographic data and other recorded parameters were compared.
    Results: A total of 301 patients were included in this retrospective cross-sectional study. It was determined that finding of a CXR score threshold value of 5 or above during hospitalization predicted a transfer to the ICU with 90% sensitivity and 80% specificity. The use of broad-spectrum antibiotics, the laboratory parameters (leukocyte, neutrophil, C-reactive protein (CRP), procalcitonin, troponin, D-dimer, ferritin), and rate of transfer to ICU were found to be significantly higher in the group with a CXR score of 5 and above, compared to the group with CXR score below 5 (P < .001 in each).
    Conclusion: Finding high CXR scores during hospitalization due to COVID-19 may act as a warning in terms of the severity of the disease. It may be beneficial to examine the chest X-ray images during hospitalization with utmost care and take these images into account.

    Abstract Objectives The aim of this study is to evaluate the accuracy and completeness of the answers given by Chat Generative Pre-trained Transformer (ChatGPT) (OpenAI OpCo, LLC, San Francisco, CA), to the most frequently asked questions on different topics in the field of periodontology. Methods The 10 most frequently asked questions by patients about seven different topics (periodontal diseases, peri-implant diseases, tooth sensitivity, gingival recessions, halitosis, dental implants, and periodontal surgery) in periodontology were created by ChatGPT. To obtain responses, a set of 70 questions was submitted to ChatGPT, with an allocation of 10 questions per subject. The responses that were documented were assessed using two distinct Likert scales by professionals specializing in the subject of periodontology. The accuracy of the responses was rated on a Likert scale ranging from one to six, while the completeness of the responses was rated on a scale ranging from one to three. Results The median accuracy score for all responses was six, while the completeness score was two. The mean scores for accuracy and completeness were 5.50 ± 0.23 and 2.34 ± 0.24, respectively. It was observed that ChatGPT's responses to the most frequently asked questions by patients for information purposes in periodontology were at least "nearly completely correct" in terms of accuracy and "adequate" in terms of completeness. There was a statistically significant difference between subjects in terms of accuracy and completeness (P<0.05). The highest and lowest accuracy scores were peri-implant diseases and gingival recession, respectively, while the highest and lowest completeness scores were gingival recession and dental implants, respectively. Conclusions The utilization of large language models has become increasingly prevalent, extending its applicability to patients within the healthcare domain. While ChatGPT may not offer absolute precision and comprehensive results without expert supervision, it is apparent that those within the field of periodontology can utilize it as an informational resource, albeit acknowledging the potential for inaccuracies.

    Abstract Introduction: Breakthroughs in targeted therapy have significantly improved outcomes for many patients with advanced melanoma, including those with BRAFV600 mutant disease. Targeted therapy for BRAFV600-mutant metastatic melanoma includes combinations of BRAF inhibitors and MEK inhibitors, which improve response rates and prolong progression-free survival (PFS) and overall survival (OS) in these patients. However, while durable responses have been observed, many patients develop acquired resistance to these drugs.
    Areas covered: Recent clinical trial updates and ongoing studies with targeted therapy for BRAF-V600 mutant melanoma are reviewed.
    Expert opinion: Although BRAF targeted therapy remains an effective treatment for BRAF-mutant for melanoma, ongoing trials are exploring combinations with other targeted therapeutics and immunotherapeutics to determine whether tumor responses can be prolonged, and these drugs are increasingly utilized in the neoadjuvant and adjuvant settings.
