    Abstract Intense and unaccustomed eccentric exercise has been extensively studied for its ability to induce muscle damage. However, the underlying mechanism of this phenomenon still requires further clarification. This knowledge gap arises from the need for explanation of the eccentric contraction through the sliding filament theory. The two-filament sarcomere model, which is consisted of thin and thick filaments, forms the basis of the sliding filament theory. The mechanisms of concentric and isometric contractions at the cellular and molecular levels are effectively described by this model. However, when relying solely on the cross-bridge swing, the sliding filament theory fails to account for specific observations, such as the stability of the descending limb of the force-length relationship curve. Recent evidence indicated that titin and the extracellular matrix (ECM) may play a protective role by interacting with the thick and thin filaments. During an eccentric contraction, titin serves as a third filament in the sarcomere, which helps regulate changes in passive force. The two-filament sarcomere model has limitations in explaining eccentric contraction, thus this compensates for those shortcomings. The present review explored the potential of replacing the two-filament sarcomere model with a three-filament sarcomere model, incorporating thin filaments, thick filaments and titin. This revised model offers a more comprehensive explanation of eccentric contraction phenomena. Furthermore, the sliding filament theory was investigated in the context of the three-filament sarcomere model. The double-layer protection mechanism, which involves increased titin stiffness and the ECM during eccentric contraction was explored. This mechanism may enhance lateral force transmission between muscle fibers and the ECM, resulting in sarcolemma and ECM shear deformation. These findings provided insight into the mechanism of eccentric exercise-induced skeletal muscle damage. Considering the three-filament sarcomere model and the double-layer protection mechanism, the present review offered a more logical and comprehensive understanding of the mechanism behind eccentric exercise-induced muscle damage.

    Abstract Electronic conduction in solid-polymer electrolytes is generally not desired, which causes leakage of electrons or energy loss, and the electronically conductive domains at electrode-electrolyte interfaces can lead to continuous decomposition of electrolytes and shorting issues. However, it is noticed in this work that in an insulating matrix, the conductive domains at certain aspects could also have positive effects on the electrolyte performance with proper control. This work evaluates the limitation and benefits of electronically conductive domains in a solid-polymer electrolyte system and discusses the approach to improve the electrolyte physicochemical properties with densified local electric field distribution, enhanced bulk dielectric property, and charge transfer. By deliberately introducing the conductive domains in a regular solid-polymer electrolyte, stable cycle life, low overpotential, and promising full cell performance could be achieved.

    Abstract Cisplatin resistance is a major cause of therapeutic failure in patients with high-grade serous ovarian cancer (HGSOC). Long noncoding RNAs (lncRNAs) have emerged as key regulators of human cancers; however, their modes of action in HGSOC remain largely unknown. Here, we provide evidence to demonstrate that lncRNA Platinum sensitivity-related LncRNA from Ascites-Derived Exosomes (PLADE) transmitted by ascites exosomes enhance platinum sensitivity in HGSOC. PLADE exhibited significantly decreased expression in ascites exosomes and tumor tissues, as well as in the corresponding metastatic tumors from patients with HGSOC cisplatin-resistance. Moreover, HGSOC patients with higher PLADE expression levels exhibited longer progression-free survival. Gain- and loss-of-function studies have revealed that PLADE promotes cisplatin sensitivity by suppressing cell proliferation, migration and invasion, and enhancing apoptosis in vitro and in vivo. Furthermore, the functions of PLADE in increasing cisplatin sensitivity were proven to be transferred by exosomes to the cultured recipient cells and to the adjacent tumor tissues in mouse models. Mechanistically, PLADE binds to and downregulates heterogeneous nuclear ribonucleoprotein D (HNRNPD) by VHL-mediated ubiquitination, thus inducing an increased amount of RNA: DNA hybrids (R-loop) and DNA damage, consequently promoting cisplatin sensitivity in HGSOC. Collectively, these results shed light on the understanding of the vital roles of long noncoding RNAs in cancers.

    Abstract Plankton-bacteria interactions may play essential roles in maintaining the persistence of pathogenic Vibrio spp. However, the actual plankton host of the toxigenic Vibrio parahaemolyticus that harbors thermostable direct hemolysin (TDH) and TDH-related hemolysin (TRH) remains unclear. To answer this question, we measured the abundance of toxigenic and nontoxigenic Vibrio parahaemolyticus in different types of plankton by most probable number (MPN)-PCR. We next investigated the dynamics of the microbiomes of rearing water, copepods, Chlorella, four predominant diatom genera (Nitzschia, Melosira, Skeletonema and Chaetoceros) and the gut of the shrimp in two recirculated shrimp farming systems from April to September using high-throughput 16 S rRNA amplicon sequencing. The survival of trh-positive and trh-negative V. parahaemolyticus in seawater and different plankton hosts was examined under low temperature and starvation conditions. The results showed that copepods harbored the highest proportion of trh-positive V. parahaemolyticus, followed by diatoms. Chitinous diatoms (CD) harbored a high proportion of Vibrionaceae, of which a high abundance of V. parahaemolyticus was found in summer. In contrast, Vibrio spp. is rarely found in Chlorella and nonchitinous diatoms. Taxon-specific associations were also observed, including a relatively high abundance of Vibrio and Halodesulfovibrio on copepods and covariation of Aeromonas and Bacillus inside the Chlorella. The survival assays showed that, in comparison to trh-negative V. parahaemolyticus, trh-positive V. parahaemolyticus showed better survival in copepods and CD under starvation conditions and maintained high persistence in the above hosts at low temperature. In conclusion, the results herein suggested that chitinous plankton might provide protection for V. parahaemolyticus, especially trh-positive V. parahaemolyticus, and improve their persistence under harsh conditions. Our study provided in-depth insights into the persistence of V. parahaemolyticus in the environment, which would promote targeted disease prevention measures.

    Abstract The exploration of brain activity and its decoding from fMRI data has been a longstanding pursuit, driven by its potential applications in brain-computer interfaces, medical diagnostics, and virtual reality. Previous approaches have primarily focused on individual subject analysis, highlighting the need for a more universal and adaptable framework, which is the core motivation behind our work. In this work, we propose fMRI-PTE, an innovative auto-encoder approach for fMRI pre-training, with a focus on addressing the challenges of varying fMRI data dimensions due to individual brain differences. Our approach involves transforming fMRI signals into unified 2D representations, ensuring consistency in dimensions and preserving distinct brain activity patterns. We introduce a novel learning strategy tailored for pre-training 2D fMRI images, enhancing the quality of reconstruction. fMRI-PTE's adaptability with image generators enables the generation of well-represented fMRI features, facilitating various downstream tasks, including within-subject and cross-subject brain activity decoding. Our contributions encompass introducing fMRI-PTE, innovative data transformation, efficient training, a novel learning strategy, and the universal applicability of our approach. Extensive experiments validate and support our claims, offering a promising foundation for further research in this domain.

    Abstract The sustainable future of modern society relies on the development of advanced energy systems. Alkali metals, such as Li, Na, and K, are promising to construct high-energy-density batteries to complement the fast-growing implementation of renewable sources. The stripping/deposition of alkali metals is compromised by serious dendrite growth, which can be intrinsically eliminated by using molten alkali metal anodes. Up to now, most of the conventional molten alkali metal-based batteries need to be operated at high temperatures. To decrease the operating temperature, we extended the battery chemistry to multielement alloys, which provide more flexibility for wide selection and rational screening of cost-effective and fusible metallic electrodes. On the basis of an integrated experimental and theoretical study, the depressed melting point and enhanced interfacial compatibility are elucidated. The proof-of-concept molten sodium battery enabled by the Bi-Pb-Sn fusible alloy not only circumvents the use of costly Ga and In elements but also delivers attractive performance at 100 °C, holding great promise for grid-scale energy storage.

    Abstract With the wide applications of deep neural network models in various computer vision tasks, more and more works study the model vulnerability to adversarial examples. For data-free black box attack scenario, existing methods are inspired by the knowledge distillation, and thus usually train a substitute model to learn knowledge from the target model using generated data as input. However, the substitute model always has a static network structure, which limits the attack ability for various target models and tasks. In this paper, we propose a novel dynamic substitute training attack method to encourage substitute model to learn better and faster from the target model. Specifically, a dynamic substitute structure learning strategy is proposed to adaptively generate optimal substitute model structure via a dynamic gate according to different target models and tasks. Moreover, we introduce a task-driven graph-based structure information learning constrain to improve the quality of generated training data, and facilitate the substitute model learning structural relationships from the target model multiple outputs. Extensive experiments have been conducted to verify the efficacy of the proposed attack method, which can achieve better performance compared with the state-of-the-art competitors on several datasets.
