    Abstract Duhuo, a member of the Angelica family, is widely used to treat ailments such as rheumatic pain. It possesses a diverse array of bioactivities, including anti-tumor, anti-inflammatory, and analgesic properties, as recent pharmacological research has revealed. Nevertheless, the mtDNA of Angelica species remains relatively unexplored. To address this gap, we sequenced and assembled the mtDNA of A. biserrata to shed light on its genetic mechanisms and evolutionary pathways. Our investigation indicated a distinctive multi-branched conformation in the A. biserrata mtDNA. A comprehensive analysis of protein-coding sequences (PCGs) across six closely related species revealed the presence of 11 shared genes in their mitochondrial genomes. Intriguingly, positive selection emerged as a significant factor in the evolution of the atp4, matR, nad3, and nad7 genes. In addition, our data highlighted a recurring trend of homologous fragment migration between chloroplast and mitochondrial organelles. We identified 13 homologous fragments spanning both chloroplast and mitochondrial genomes. The phylogenetic tree established a close relationship between A. biserrata and Saposhnikovia divaricata. To sum up, our research would contribute to the application of population genetics and evolutionary studies in the genus Acanthopanax and other genera in the Araliaceae family.

    Abstract Photoperiod is an important environmental factor that influences seasonal reproduction behavior in birds. Birds translate photoperiodic information into neuroendocrine signals through deep brain photoreceptors (DBPs). OPN5 has been considered candidate DBPs involved in regulating seasonal reproduction in birds. We found that OPN5 could mediate light to regulate the follicle development in ducks. In this study, we further verified the effect of OPN5 on follicular development in Shan Partridge ducks by immunizing against the extracellular domain (ECD) of OPN5. We investigated the specific regulatory mechanism of photoperiod mediated by OPN5 on the reproductive activity of ducks. The trial randomly divided 120 Shan Partridge ducks into 3 groups with different treatments: the immunization of OPN5 group was done at d0, d15, d30, and d40 with 1 mL of vaccine containing OPN5 protein (thus containing 1, 1, 0.5, and 0.5 mg of OPN5-KLH protein), and the control group (CS and CL groups) was injected at the same time with the same dose of OPN5-uncontained blank vaccine. The group of CS (900 lux), OPN5 (600 lux), and CL (600 lux) lasted for 40 d in 12 L:12 D photoperiods, respectively. Then, the groups of CS, OPN5, and CL subsequently received 12 L:12 D, 12 L:12 D, and 17 L:7 D light treatments for 33 d, respectively. The ducks were caged in 3 constant rooms with the same feeding conditions for each group, free water, and limited feeding (150 g per duck each day). Duck serum and tissue samples were collected at d 40, d 62, and d 73 (n = 12). It was found that before prolonged light, the group of immunization (group OPN5) and the group of strong light intensity (group CS) were higher than the group of CL in egg production. Subsequent to prolonged light, the group CL in egg production rose about the same as the group immunization, while the strong light group (group CS) was lower. Group OPN5 increased the ovarian index of ducks, and both the immunization of group OPN5 and group CL (extended light) increased the thickness of the granular layer and promoted the secretion of E2, P4, LH, and PRL hormones. Compared with group CS, group CL and OPN5 increased the mRNA level and protein expression of OPN5 in the hypothalamus on d 62 and d 73 (P < 0.05). The gene or protein expression patterns of GnRH, TRH, TSHβ, DIO2, THRβ, VIP, and PRL were positively correlated with OPN5, whereas the gene expression patterns of GnIH and DIO3 were negatively correlated with OPN5. The results showed that immunization against OPN5 could activate the corresponding transmembrane receptors to promote the expression of OPN5, up-regulate the expression of TSHβ and DIO2, and then regulate the HPG axis-related genes to facilitate the follicular development of Shan Partridge ducks. In addition, in this experiment, prolonging the photoperiod or enhancing the light intensity could also enhance follicle development, but the effect was not as significant as immunizing against OPN5. Our results will offer beneficial data and more supportive shreds of evidence in favor of elucidating the role of OPN5 in relation to photoperiods and reproduction.

    Abstract Background: Although the traditional Chinese medicine Shan-Zhu-Yu may be efficacious against depression, its mechanism of action is unknown. In this study, we aimed to explore the possible mechanisms of action of Shan-Zhu-Yu in the treatment of depression using network pharmacology.
    Methods: The active ingredients and targets of Shan-Zhu-Yu were obtained from the Traditional Chinese Medicine System Pharmacology Database (TCMSP) database and converted into gene names using UniProt. Then, the target genes of depression were collected using GeneCards and OMIM. Drug disease intersection genes were obtained using a Venn tool, and a protein-protein interaction network was constructed using STRING. Cytoscape was used to construct an active ingredients-targets-drug-disease network. GO and KEGG pathway enrichment analyses were performed using DAVID. Furthermore, Autodock was used to evaluate drug and target binding and explore possible molecular mechanisms.
    Results: We identified 9721 disease genes, 13 active ingredients, 50 target genes, and 48 drug disease intersecting genes. The results of the GO enrichment analysis suggested that Shan-Zhu-Yu affects the activity of G protein-coupled amine, neurotransmitter, steroid hormone, nuclear, and G protein-coupled neurotransmitter receptors in the treatment of depression by acting on hormone and nuclear receptor binding. The main signaling pathways were associated with neuroactive ligand-receptor interaction, calcium, cGMP-PKG, apoptosis, estrogen, p53, and AGE-RAGE.     Abstract The Helan Shan is located on the front edge of the expanding Tibetan Plateau. However, how the topography here responds to this propagation and its precise time constraints remain unknown. Based on the response process of fluvial landforms and tectonic evolution, we conducted a quantitative landform analysis of the Helan Shan region. Here, the spatial distribution features of various geomorphic indices were coupled, demonstrating that the mountain is tilting toward the west and north. The steeper downstream and gentle upper reaches indicate that the fluvial landforms have experienced an accelerated incision event, which can be attributed to the tectonic activity along the East Helan Shan Fault. Furthermore, the response time of the tectonic knickpoints ranges from 0.1 to 1.4 Ma based on the paleochannel reconstruction method. Combined with previous studies on low temperature thermochronology and active tectonics, we proposed a tectonic transformation model where the Helan Shan shifted the tilting model from the southwest to the northwest, as induced by the northeast expansion of the Tibetan Plateau from 0.1 to 1.4 Ma.

    Abstract The Tian Shan is one of the most active intracontinental orogenic belts in the world. It has undergone complex deformation that has resulted in the formation of several fold-and-thrust belts (FTBs) in the piedmonts and intermontane basins. Investigating the deformation histories of these FTBs is important for understanding the tectonic propagation processes of the Tian Shan. Here, we gain insight into these crustal shortening processes by deciphering the Houyanshan structure, a typical fold-thrust belt in the eastern Chinese Tian Shan. We first describe a curved thrust ramp and related fold pairs of the structure using high-resolution remote sensing photography, deformation of fluvial terraces, and field-based geological cross-section. Combined with deformed terrace records and optically stimulated luminescence (OSL) dating results, the kinematic style allows us to yield a geologic shortening rate of 1.6 ± 0.2 mm/a since ~52 ka. Second, to reduce uncertainty in the seismic interpretation and quantify the amount and time of crustal shortening, we interpret three seismic reflection profiles by using the theory of quantitative fault-related fold, area-depth-strain (ADS), and reverse modeling analyses. These profiles provide direct evidence that this structure connects by means of a listric thrust ramp to a shallow detachment level. ADS analysis reveals that the maximum shortening of the Huoyanshan structure is ~4.5 km, which is consistent with the result of quantitative inverse modeling. Each of the structural analysis methods gives similar parameters, and the high consistency of results greatly improves the soundness of a given geologic interpretation. Finally, the shortening rate and total shortening amount suggest that the structure may have formed at 1.8–3.7 Ma, which is nearly synchronous around the Tibetan Plateau.     Abstract The phenomenon in which the warming rate of air temperature is amplified with elevation is termed elevation-dependent warming (EDW). It has been clarified that EDW can accelerate the retreat of glaciers and melting of snow, which can have significant impacts on the regional ecological environment. Owing to the lack of high-density ground observations in high mountains, there is widespread controversy regarding the existence of EDW. Current evidence is mainly derived from typical high-mountain regions such as the Swiss Alps, the Colorado Rocky Mountains, the tropical Andes and the Tibetan Plateau–Himalayas. Rare evidence in other mountain ranges has been reported, especially in arid regions. In this study, EDW features (regional warming amplification and altitude warming amplification) in the Chinese Tian Shan (CTM) were detected using a unique high-resolution (1 km , 6-hourly) air temperature dataset (CTMD) from 1979 to 2016. The results showed that there were significant EDW signals at different altitudes on different timescales. The CTM showed significant regional warming amplification in spring, especially in March, and the warming trends were greater than those of continental China with respect to three temperatures (minimum temperature, mean temperature and maximum temperature). The significance values of EDW above different altitude thresholds are distinct for three temperatures in 12 months. The warming rate of the minimum temperature in winter showed a significant elevation dependence ( p <0.01 ), especially above 3000 m . The greatest altitudinal gradient in the warming rate of the maximum temperature was found above 4000 m in April. For the mean temperature, the warming rates in June and August showed prominent altitude warming amplification but with different significance above 4500 m .     Abstract Climate changes have substantial impacts on the geographic distribution of montane lakes and evolutionary dynamics of cold-adapted species. Past climate cooling is hypothesized to have promoted the dispersal of cold-adapted species via montane lakes, while future climate warming is thought to constrain their distributions. We test this hypothesis by using phylogeographic analysis and niche modeling of the Holarctic crustacean Gammarus lacustris with global sampling comprised of 567 sequenced individuals and 3180 occurrence records. We found that the species arose in Tian Shan in Central Asia and dispersed into montane lakes along the Alps, Himalayas, Tibet, East Asia, and the North American Rocky Mountain ranges, with accelerated diversification rates outside Tian Shan. Climatically suitable regions for geographic lineages of G. lacustris were larger during cooling periods (LGM), but smaller during warming periods (Mid-Holocene). In the future (2070) scenario, potential distributions in the Himalayas, North Tibet, South Tibet and North America are predicted to expand, whereas ranges in East Asia, Europe and Tian Shan will decline. Our results suggest that Mid-Miocene-to-Pleistocene continuous cooling promoted multiple independent dispersal events out of Tian Shan due to increased availability of montane lakes via "budding" of lineages. Montane lakes are conduits through which cold-adapted amphipods globally dispersed, dominating circumboreal lakes. However, future climate warming is likely to force organisms to shift upward in altitude and northward in latitude, leading to a future change in local populations. These findings highlight the importance of conservation of montane lakes, especially in the context of climate change.
