    Abstract Multisource unsupervised domain adaptation (MUDA) is an important and challenging topic for target classification with the assistance of labeled data in source domains. When we have several labeled source domains, it is difficult to map all source domains and target domain into a common feature space for classifying the targets well. In this article, a new progressive multisource domain adaptation network (PMSDAN) is proposed to further improve the classification performance. PMSDAN mainly consists of two steps for distribution alignment. First, the multiple source domains are integrated as one auxiliary domain to match the distribution with the target domain. By doing this, we can generally reduce the distribution discrepancy between each source and target domains, as well as the discrepancy between different source domains. It can efficiently explore useful knowledge from the integrated source domain. Second, to mine assistance knowledge from each source domain as much as possible, the distribution of the target domain is separately aligned with that of each source domain. A weighted fusion method is employed to combine the multiple classification results for making the final decision. In the optimization of domain adaption, weighted hybrid maximum mean discrepancy (WHMMD) is proposed, and it considers both the interclass and intraclass discrepancies. The effectiveness of the proposed PMSDAN is demonstrated in the experiments comparing with some state-of-the-art methods.

    Abstract Environmentally persistent free radicals (EPFRs) are emerging pollutants stabilized on or inside particles. Although the toxicity of EPFR-containing particles has been confirmed, the conclusions are always ambiguous because of the presence of various compositions. A clear dose-response relationship was always challenged by the fact that the concentrations of these coexisted components simultaneously changed with EPFR concentrations. Without these solid dose-response pieces of evidence, we could not confidently conclude the toxicity of EPFRs and the description of potential EPFR risks. In this study, we established a particle system with a fixed catechol concentration but different reaction times to obtain particles with different EPFR concentrations. Caenorhabditis elegans (C. elegans) in response to different EPFR concentrations was systematically investigated at multiple biological levels, including behavior observations and biochemical and transcriptome analyses. Our results showed that exposure to EPFRs disrupted the development and locomotion of C. elegans. EPFRs cause concentration-dependent neurotoxicity and oxidative damage to C. elegans, which could be attributed to reactive oxygen species (ROS) promoted by EPFRs. Furthermore, the expression of key genes related to neurons was downregulated, whereas antioxidative genes were upregulated. Overall, our results confirmed the toxicity from EPFRs and EPFR concentration as a rational parameter to describe the extent of toxicity.

    Abstract Background: Because of the wide use of information and communication technologies (ICT) in healthcare, medical students' knowledge and skills of modern ICT have been considered essential for their successful learning and future careers. According to Bandura's self-efficacy, enhancing ICT self-efficacy, which might be affected by technology experience, could be a pathway to improving ICT literacy and competence, which should be one focus of medical educationalists. However, there is a lack of suitable measurements of medical students' self-efficacy and a clear understanding of its relationship with technology experience.
    Materials and methods: We conducted a literature review and direct consultation with an expert panel to identify potential items for the ICT self-efficacy scale. Based on the data collected in a survey of 486 first-year medical students in China, the exploratory factor analysis (EFA) was employed to confirm the structure of the final version. Furthermore, we used linear regressions to quantify the association between ICT self-efficacy and technology experience measured by the age of first access to the Internet, the age at first ownership of a personal computer (PC) or a laptop, and that of a smartphone.
    Results: The EFA results derived 15 items of four factors, with 67.02% of the total variance explained:
    Conclusion: The ICT scale developed is a reliable and valid task-specific measure to assess ICT Self-Efficacy for medical students. In addition, enhancing students' technology experience might improve their ICT self-efficacy.

    Abstract Depression is a prevalent and debilitating psychiatric illness. However, the antidepressant drugs currently prescribed are only effective in a limited group of patients. Histone modifications mediated by histone acetylation are considered to play an important role in the pathogenesis and treatment of depression. Recent studies have revealed that histone deacetylase inhibitors may be involved in the pathogenesis of depression and the underlying mechanism of the antidepressant therapeutic action. Here, we first conducted virtual screening of histone deacetylase-5 (HDAC5) inhibitors against HDAC5, a target closely related to depression, and identified compound T2943, further verifying its inhibitory effect on enzyme activities in vitro. After stereotaxic injection of T2943 into the hippocampus of mice, the antidepressant effect of T2943 was evaluated using behavioral experiments. We also used different proteomic and molecular biology analyses to determine and confirm that T2943 promoted histone 3 lysine 14 acetylation (H3K14ac) by inhibiting HDAC5 activity. Following the overexpression of adenoviral HDAC5 in the hippocampus of mice and subsequent behavioral analyses, we confirmed that T2943 exerts antidepressant effects by inhibiting HDAC5 activity. Our findings highlight the efficacy of targeting HDAC5 to treat depression and demonstrate the potential of using T2943 as an antidepressant.

The Cronbach’s alphas for the four subscales and the overall scale ranged from 0.78 to 0.89. Regression results demonstrated a significant association of ICT self-efficacy with age at first ownership of a personal computer (PC) and the mediation role of the general self-efficacy in the ICT self-efficacy’s association with the age at first ownership of a personal smartphone.Conclusion The ICT scale developed is a reliable and valid task-specific measure to assess ICT Self-Efficacy for medical students. In addition, enhancing students’ technology experience might improve their ICT self-efficacy.

    Abstract Objective: The obesity epidemic and its metabolic complications continue to be a major global public health threat with limited effective treatments, especially drugs that can be taken orally. Peptides are a promising class of molecules that have gained increased interest for their applications in medicine and biotechnology. In this study, we focused on looking for peptides that can be administrated orally to treat obesity and exploring its mechanisms.
    Design: Here, a 9-amino-acid peptide named D3 was designed and administered orally to germ-free (GF) mice and wild-type (WT) mice, rats and macaques. The effects of D3 on body weight and other basal metabolic parameters were evaluated. The effects of D3 on gut microbiota were evaluated using 16S rRNA amplicon sequencing. To identify and confirm the mechanisms of D3, transcriptome analysis of ileum and molecular approaches on three animal models were performed.
    Results: A significant body weight reduction was observed both in WT (12%) and GF (9%) mice treated with D3. D3 ameliorated leptin resistance and upregulated the expression of uroguanylin (UGN), which suppresses appetite via the UGN-GUCY2C endocrine axis. Similar effects were also found in diet-induced obese rat and macaque models. Furthermore, the abundance of intestinal
    Conclusion: Our results indicated that D3 is a novel drug candidate for counteracting diet-induced obesity as a non-toxic and bioactive peptide. Targeting the UGN-GUCY2C endocrine axis may represent a therapeutic strategy for the treatment of obesity.
